Serum isoferritin levels were detected by ELISA in 96 normal, 11 cases of hepatocellular carcinoma (HCC), 28 breast cancer (BC), 31 lung cancer (LC), 26 breast fibroma, 11 pneumonia and 11 tuberculosis. The results reveal significant differences of serum isoferritin levels between the normals and the patients, and between the malignant cases and benign cases (P < 0.01). Serum isoferritin demonstrates higher sensitivity in detecting HCC, LC and BC and thus is of great value in the differential diagnosis of these cancers.

The effects of fibronectin (FN) on cytodifferentiation and cell cycle in a human hepatoma cell line QGY-7703 were studied by flow cytometry, electron microscopy, etc. The results indicated that FN inhibited cell proliferation and mitosis; significant differences were noted between the control group and the experimental group (P < 0.01). FN blocked the cell cycle in phase S; cell spreading improved with less cell overlapping, and the growth of monolayer cell was partially restored. The number of microvilli and marginal ruffles decreased and that of mitochondria, Golgi complex, endoplasmic reticulum increased. The findings suggested that FN could partially restore QGY-7703 cell normal phenotype and probably be useful for the treatment of tumor.

Nine patients with inoperable hepatoma were treated by using hepatic arterial embolization 131I and chemotherapeutic agent gelatin microsphere (131I-CA-GM). The emission CT after operation detected that the microspheres were concentrated on tumor area. The ratio between the radioactivity in tumor and that in liver was 4.1:1. A case died of ictopic embolization; the others survived 3, 4, 5, 19, 24, 7, 8, and 12 months respectively. Three of them were still alive. 131I-CA-GM has triple anticarcinogenic actions, including the arterial occlusion, targeting chemotherapy and internal radiation. The microspheres can selectively accumulate in the tumor artery and can be easily traced by gamma-camera or emission CT. 131I-CA-GM is a hopeful embolic agent for the treatment of liver cancer, but some problems about ectopic arterial embolization should be further studied.

A new operative procedure for hepatic segmentectomy was performed by dissecting the hepatic hila to occlude blood vessels to and from hemihepatis, one lobe or one segment which bore hepatoma. Forty-three liver cancers with 33 of them in stage IV, 4 metastatic liver cancers and 16 hemangiomas were resected by this method. The types of hepatic segmentectomy consisted of monosegmentectomy, multisegmentectomy, peripheral and central segmentectomy, segmentectomy after hepatic arteria embolization and resegmentectomy for recurrent hepatic cellular carcinoma etc. The vascular occlusion lasted as long as 120 minutes for the right portal vein and 70 minutes for the left. Intravenous infusion of extract from Salive mliltiorrhiza before vascular occlusion could prevent hepatic cells from reperfusion injury. The mentioned maneuvers resulted in mild postoperative liver dysfunction, rapid recovery and hepatocytes regeneration. Two patients died within 30 days postoperatively, constituting an operative mortality of 3.1%.

Type-A LDH (LD5), isolated from normal C57 mouse liver, H22 hepatoma and S180 sarcoma after a sequential ion-exchange and affinity chromatography, was examined to be quite pure. Studies on the enzyme kinetics and immunological properties showed that there were no differences in optimal pH, optimal temperature, subunit MW., isoelectric point between normal and malignant (H22 hepatoma and S180 sarcoma) LD5. Km to pyruvate decreased in malignant LD5 while LD5 content and percentage increased. Three LD5 antibodies raised against rabbits showed cross-reaction. These antibodies as probes are useful in the isolation of LD5-mRNAs from G57 mouse liver H22 hepatoma and S180 sarcoma.

The characters of liver isoferritin from Wistar rat, normal human and patient with hepatocellular carcinoma (HCC) were studied. The ratio of iron to protein of liver isoferritin from patient with HCC were 0.021, which accounts for about one-twelfth of that of liver isoferritin from normal human. The molecular weights of liver isoferritin from normal human and the patient with HCC were 20.5 kd and 21.5 kd, respectively. The electrophoresis patterns of liver isoferritin from both Wistar rat and patient with HCC showed one band, but two bands from normal human. There was cross reaction in immunity among isoferritins from normal human liver, Wistar rat liver and horse spleen by double immunodiffusion. The PAGE patterns of liver isoferritin from different sources could be clearly differentiated. The data presented may serve as a good reference for further study of the relationship between the structure and function of ferritin and may be used practically in clinical medicine.

The hepatocellular carcinoma (HCC) cell line, QGY-7703, derived from a Chinese patient, was used to immunize the BALB/c mice. Fifteen hybridomas producing McAb that reacted with QGY-7703 cells were isolated from 858 hybridomas created in three cell fusions. In further studies two McAb, namely, AQGY1 and A-QGY2, were selected which specifically stained HCC cells grown in vitro. The reactivity of these McAb was not removed by the absorption by homogenates of the normal liver, but was by homogenates of HCC cells. A-QGY1 and A-QGY2 also reacted definitely with HCC cells in liver tissues of HCC patients, but neither with other cells in the tissues nor with nontransformed liver tissues of the same patients. Furthermore these two McAb stained the adult or fetal liver tissues, nor all of the other normal or tumor tissues that had been tested. Blocking and absorbing experiments revealed that A-QGY1 and AQGY2 antigens had no immunohomogenicity with antigens such as HBsAg, HBcAg, HBeAg, AFP and CEA. The specificity of these two McAb may be used potentially for the sero diagnosis, histologic identification, radioimmunoimaging and destruction of human hepatocellular carcinoma.

Thin-layer IEF, an analytical technique of high resolution for protein, was used to identify the liver isoferritin in Wistar rat, normal human and patients with hepatocellular carcinoma (HCC) respectively. The focusing conditions were 2000 V, 25W, 10 degrees C, 2h. The results showed that the IEF bands of the liver isoferritin in Wistar rat, normal human and patients with HCC were 7, 6, 3 and the pI of the liver isoferritin were 5.76-4.75, 5.80-4.85, 4.50-4.43 respectively. The method and data in this paper might be good reference for in-depth study of the structure and function of ferritin and its applications in clinical medicine.

Molecular weight of human hepatocellular carcinoma (HCC)-associated antigen-HL2 antigen was detected to be about 50,000 dalton by SDS-PAGE and Western blot. One hundred and twenty-seven paraffin sections of cancers and pathological liver tissue were tests by ABC immunohistochemical staining. MAb HL2 reacted with 62.7% HCC and also with 8 of the 10 stomach carcinomas but only with a few other digestive system carcinomas (pancreas carcinoma, rectum carcinoma and duodenum carcinoma) and hepatitis. MAb HL2 was negative in hepatocirrhosis and other tumors (vesica carcinoma, skin carcinoma, breast carcinoma, neurogliocytoma and carcinoma of the lung). The results suggest that MAb HL2 has values in diagnosis of HCC and differential diagnosis between HCC and other tumours. HL2 antigen might be a new tumor-associated antigen (TAA). Further study of HL2 antigen will significantly show the actions of TAA in the occurrence and development of tumor.

This paper reports the result of an experiment on the anticarcinogenic effect of liver targeted drug delivery system-mitoxantrone-polybutylcyanoacrylate-nanospheres (DHAQ-PBCA-NS) by using a model of heterotopic and orthotopic transplantation of human hepatocellular carcinoma (HCC) in nude mice. No significant difference was noted between the anti-HCC effects of mitoxantrone and dexerubicin, but the anti-orthotopic transplanting HCC effect of mitoxantrone-nanospheres was obviously higher than that of mitoxantrone and dexerubicin by comparison of tumor inhibition rates and proliferating cell nuclear antigen (PCNA). The result showed the applicability of mitoxantrone-nanospheres.
