The fate of lectin labeled internalized plasma membrane in the ascites tumor form of the Chang rat
hepatoma
growing under in vivo and in vitro conditions was investigated cytochemically. Ascites cells were incubated in Convanavalin A (Con A) and horseradish peroxidase (PO), either with or without prior glutaraldehyde fixation and subsequently treated with 3',3-diaminobenzidine. In cells fixed before Con-A-PO labeling the reaction product was localized as a continuous and even layer upon the external surface of the plasma membrane. If unfixed cells were treated with Con A, coupled with PO at 4 degrees C and reincubated in
phosphate
buffered saline at 37 degrees C for varying periods of time, the Con-A-PO layer was of irregular thickness. In as little as 15 min of reincubation endocytotic vesicles containing PO positive material were closely associated with GERL components of the Golgi Apparatus. Localization of acid phosphatase (ACPase) within GERL vesicles, similar in size and location to those containing Con-A-PO reaction product, indicates that the Con-A-PO labeled vesicles may be a component of the Golgi apparatus in

Agents that increase (metabolic inhibitors) or decrease (hormones) the sensitivity of the cells to killing by antibody plus complement were examined for their effects on the chemical, physical, and enzymic composition of the cells. The effects of these agents on the chemical and enzymic characteristics of isolated plasma membrane and intracellular fractions of these cells were also measured. Adriamycin treatment resulted in a decrease in the amount of ribonucleoprotein and smooth endoplasmic reticulum isolated from the cells as compared to untreated cells. Hormone (insulin or hydrocortisone)-treated cells were enhanced in their yield of this fraction but were decreased in their yield of mitochondria as compared to controls. Carbohydrate metabolism by four rat

cell lines in culture, namely, Reuber H35, MH1C1, RLC, and HTC, has been investigated. Glucose utilization by H35 and MH1C1 cells is lower than that by RLC and HTC cells. The four cell lines also differ with respect to the accumulation of lactic acid in the growth medium; in particular, H35 cells show uptake of lactic acid, rather than accumulation in the medium. Specific activities of a number of enzymes involved in glycolysis, gluconeogenesis, pentose

pathway, and glycogen formation were determined in the four cell lines. A direct relationship between the differences was found for the activities of some enzymes belonging to carbohydrate metabolism, namely, hexokinase, pyruvate kinase, aldolases A and B, glucose-6-phosphate dehydrogenase, and phosphogluconate dehydrogenase and the differences found for glucose utilization by the different cell lines.

(Tris-BP) and the hair-dye component 2,4-diaminoanisole (2,4-DAA) were studied by possible cytotoxic effects in rat
hepatoma
cells grown in culture and in suspensions of isolated rat hepatocytes. Cell growth of Reuber cells was inhibited by 50% at 50 microgram/ml Tris-BP and 20 microgram/ml 2,4-DAA, respectively. At 200 microgram/ml Tris-BP protein synthesis in Reuber cells was reduced by 40%, whereas 50% inhibition of protein synthesis in isolated hepatocytes was seen at 100 microgram/ml. IC50 of 2,4-DAA with respect to protein synthesis was found at 400 microgram/ml in Reuber cells and at 3600 microgram/ml in MH1C1 cells, whereas in the isolated hepatocytes IC50 was 650 microgram/ml. DNA synthesis was inhibited by 50% at 225 microgram/ml Tris-BP in Reuber cells. At 500 microgram/ml 2,4-DAA DNA synthesis in Reuber and MH1C1 cells was inhibited by more than 80%.

cell line (N1-S1/FdUrd) to 5-fluorouracil (FUra) was reversed by the inclusion of inosine in the culture medium. As the concentration of inosine in the medium was increased, there was a marked increase in the uptake of [14C]FUra and conversion to nucleotides with a corresponding increase in the incorporation into RNA. While FUra alone had no effect on this resistant cell line, the combination of FUra plus inosine altered the levels of ribose 1-
phosphate
but not 5-phosphoribosyl 1-pyrophosphate, altered the maturation of precursor ribosomal RNA by blocking the formation of 18S RNA, altered the methylation of the ribosomal RNA, and caused inhibition of the growth of these cells. No evidence was obtained that fluorodeoxyuridine 5'-monophosphate was formed in the N1-S1/FdUrd cells as a result of treatment with FUra plus inosine. In addition, the metabolism of [3H]deoxycytidine in the presence of FUra plus inosine in the intact N1-S1/FdUrd cells did not indicate significant inhibition of thymidylate synthetase as evidenced by the levels of deoxyuridine 5'-monophosphate or conversion to thymidine 5'-monophosphate.

remained constant, there was a significant decrease in

, magnesium, fluoride and parathyroid hormone. Corresponding to these results, negative balances could be seen during the course of a hemofiltration treatment: for
phosphate
a mean value of -593 mg, for magnesium -8.4 mEq and for fluoride -458 microgram. When a calcium content of 3.75 mEq/l was used in the substitution solution, an only slightly positive calcium balance of +1.51 mEq/l (mean value) was found. A significant correlation between calcium and fluid balance was demonstrated by means of 197 filtration treatments of one patient: the calcium balance became negative whenever the fluid loss was greater than 3.86 liters. After a 3-month period no significant changes in the above parameters were found, which indicates, that disturbances in the calcium-
phosphate
-parathyroid hormone metabolism do not only lie in a reduced renal elimination. Even though our results do not indicate that hemofiltration treatment induces or increases the chances of renal osteodystrophy, the calcium concentration of the substitution solution should be increased to 4.0 mEq/l, in order to guarentee a positive calcium balance even by forced filtration.
