Although there are many complementary and supplementary types of diagnostic imaging of the liver, a logical sequence for most liver pathology begins with high-quality ultrasound. It is noninvasive and inexpensive but very operator dependent. In other settings, CT may be the preferable screening modality, as it gives an excellent picture of the global anatomy and is easily reproducible. Frequently, the two modalities are complementary and indicated; on other occasions, one method will suffice. Radionuclide evaluation of the liver is usually reserved for hepatobiliary imaging for biliary obstruction or the question of acute cholecystitis and tagged-red cell scanning for hemangioma. It is also frequently used for gallium scanning in
hepatoma
, but
lymphoma
and inflammatory diseases are also gallium avid. The invasive imaging tests of the liver--angiography and transhepatic and endoscopic retrograde cholangiography--are performed when insufficient information is obtained by the other methods (as in diagnostic transhepatic cholangiography) or when the procedure offers therapy (as for biliary drainage, percutaneous transhepatic removal of common bile duct stones, percutaneous cholecystosis with gallstone dissolution and liver embolization, or the angiographic evaluation for portal shunting or liver resection). The impact of magnetic resonance imaging and fourth-generation raid angiotomography CT scanning has yet to be felt. The use of intraoperative ultrasound is to be encouraged prior to liver resection because it can demonstrate lesions as small as 3 mm in diameter.

In the United States, New York City has had the greatest number of subjects at risk of AIDS for the longest period of time. This population therefore serves as an indicator of changes in cancer risk which may emerge among persons at risk from AIDS. Using a proportional incidence method, we surveyed cancers occurring among single (a surrogate for homosexual) young men and married young men in Manhattan, the rest of New York City, and the remainder of New York State. The baseline period was established earlier to be 1973-76, during which time no cases of Kaposi's sarcoma were observed among single men in Manhattan. By 1985, the frequency of Kaposi's sarcoma in this group was increased 1,850-fold (compared with expected cases derived from other registries). In the same group, the increase of non-Hodgkin's lymphoma was 6.2-fold (p for trend less than 0.0001), with excesses of Burkitt's lymphoma and immunoblastic
lymphoma
being most noticeable. Diagnoses of Hodgkin's disease increased markedly in 1985 but not earlier. Since this pattern did not follow that of the AIDS epidemic in this area, we suggest that Hodgkin's disease is not an AIDS-associated tumor.
Hepatoma
was diagnosed more frequently in single young men during the 1980s but similar increases also were observed in married men and thus may be unrelated to AIDS. Thus, only Kaposi's sarcoma and non-Hodgkin's lymphoma appear to be AIDS-associated tumors, at least so far. With better treatment and longer survival, it remains possible that other tumors will emerge as part of the AIDS epidemic.

Thymus and lymph node lymphocytes in L-1210 ascite
lymphoma
, Lewis tumor, 22a
hepatoma
have been studied by NMR. Lymphocyte 1H-NMR spectra can to a certain extent reflect the state of immune system of an organism, in particular, they can sense both transformation of lymphocytes, and development of several tumors which are not accompanied by transformation of lymphocytes. In the latter case, changes in lymphocyte spectra takes place at terminal stages of carcinogenesis and are less pronounced than at transformation of lymphocytes. NMR spectra parameters can also distinguish between reaction of males and females to tumor formation.

All of the sterols were found to possess cytotoxicity when incubated with
hepatoma
(HTC) and
lymphoma
(RDM-4) cells in culture at 10-30 microM concentrations.

Sixty-eight patients with various malignancy was examined for their natural killer (NK) cell activity against 14 target cell lines. The group consisted of 10 patients with gastric cancer, 10 patients with lung cancer, 8 patients with
hepatoma
, 11 patients with cancer of female genital organs, 14 patients with malignant
lymphoma
and 15 patients with acute myelogenous leukemia (AML). The target cells from a variety of lineage were selected to examine the disease-related specificity in NK cell activity. The peripheral mononuclear cells from patients with gastric cancer did not show a decrease in NK activity against 14 targets including gastric cancer cell lines. Other patients except for AML demonstrated low NK activity against one or two target cells out of 14 targets. Whereas, NK activity in patients with AML was remarkably depressed against 10 target cells out of 14. At single cell assay, killing ability rather than binding activity to target was markedly impaired in AML. Comprehensively, the data demonstrated the marked difference in the NK level between the patients with solid tumor and the patients with hematopoietic malignancy. There existed neither disease-related specificity in NK cytolysis, nor correlation in NK levels and clinical severity in the patients with malignancy. These results suggested that it was very difficult to evaluate the anti-cancer capacity in patients with malignancy by NK activity alone.

We have found that genomes of human T cell leukemia-
lymphoma
virus type I (HTLV-I), BK virus (BKV), and a hepatitis B virus (HBV) DNA sequence integrated into DNA of a
hepatoma
-derived cell line contain binding sites for nuclear factor 1 (NF-1), a cellular protein which binds to adenoviral and putative cellular origins of DNA replication. We suggest that cellular origins of DNA replication acquired by oncoviruses may play a role in malignant transformation after reintegration into the cellular genome by providing new targets for cellular factors initiating DNA replication and by perturbing the temporal order of replication.

Hepatocellular carcinoma
is strongly associated with hepatitis B virus carrier patients who usually have HBV sequences integrated in the chromosomal DNA of liver cells. To assess the possible effects of HBV regulatory sequences (e.g., the enhancer) on expression of neighboring host genes we have screened for cellular genes that are both overexpressed and adjacent to integrated HBV sequences in
hepatocellular carcinoma
cells. The cloned cDNA for one such gene encodes a protein similar to the E. coli L-3 ribosomal protein which is thought to play a role in mRNA binding to the ribosome. The protein encoded by the cDNA localizes to the nucleolus and is also found in ribosomes; possibly it is the mammalian homologue of L-3 (MRL3). The expression of MRL3 is higher in colon carcinoma and
lymphoma
cell lines than in normal liver, placenta and diploid fibroblasts, and is also higher in fetal than in adult liver. Therefore, MRL3 overexpression seems to be a property of rapidly dividing cells and is not directly linked to oncogenesis.

Chalone-like proteoglycans (PG) have been isolated from 22a
hepatoma
, spontaneous adenocarcinoma of the mammary gland of C3H/He mice and from transplanted X-ray-induced
lymphoma
in C57Bl mice. It has been found that the biological activity of these PG was considerably weaker than that of PG taken from the liver, mammary gland and spleen of cattle. PG from 22a
hepatoma
produced a weaker tissue-specific antimitotic effect as against PG from the liver when acting on the liver cells of 6-day C57Bl mice, while the
lymphoma
PG under these conditions was ineffective. PG from the liver and mammary gland inhibited the growth of certain transplanted tumours in vitro. The
lymphoma
PG has stimulated the tumour growth.

The expression of 7 cellular oncogenes in a human
hepatoma
cell line PLC/PRF/5 was studied using Northern blot analyses. Among the oncogenes tested, c-abl, c-fes, c-fms, c-myc, c-Ha-ras and c-sis were expressed. The oncogene c-Ki-ras was not expressed. The length of the mRNAs expressed was almost consistent with published data. Compared to the oncogene expression in Daudi
lymphoma
cells, the same kind of oncogenes were expressed in PLC/PRF/5 cells, but the intensity of the signal in each oncogene expression was stronger in Daudi cells than in PLC/PRF/5 cells. Considering the cellular localization and the function of each oncogene, the oncogene survey in
hepatoma
cells broadens the knowledge of hepatocarcinogenesis and the character of human
hepatoma
cells.

The antitumor effects of 5 different subtypes of human type I interferons (IFN-alpha(Le), rIFN-alpha A, rIFN-alpha D, IFN-beta, and rIFN-beta) were studied on 3 human cancer cell lines (Daudi
lymphoma
, PLC/PRF/'
hepatoma
, and G361 melanoma). IFN-alpha(Le) was the most potent against Daudi cells. The effects of IFNs were cytostatic. On the other hand, 500 IU/ml of IFN-alpha(Le), IFN-beta and 5000 IU/ml of rIFN-beta showed cytocidal effect against PLC/PRF/5 cells. The G361 cells were the least sensitive to the IFNs tested. This is the first report on the antitumor effect of rIFN-beta isolated from insect cells. The effect of this rIFN-beta was similar to that of IFN-beta.
