Effects of single doses of kumari asav, kumari kalp, arogyavardhini and tamra bhasma on lysosomal enzymes (

and beta-glucuronidase) of rat liver and kidney were studied during
hepatitis
induced by single 0.3 ml/kg body wt dose of CCl4. Histologically all the drugs showed significant hepatoprotection. While
acid phosphatase
activities of liver and kidney were suppressed, activities of beta-glucuronidase were enhanced by these drugs. The results indicate that
acid phosphatase
and beta-glucuronidase behave differently, although they are lysosomal in nature.

Activity of acid hydrolases,
acid phosphatase
, cathepsin D, DNA- and RNAases in lysosomal fractions of liver tissue and peripheric blood lymphocytes as well as concentration of circulating immune complexes in blood serum were studied in thymectomized rats and in thymectomized rats with chronic heliotrinic
hepatitis
; thymosin and alimentary factors were used for treatment and correction of the impairments observed. The rate of thymus hormones deficiency was found to be responsible for impairments of functional activity of lysosomes in lymphocytes and liver tissue. Activation of the lysosomal enzymes studied was detected within early periods (40 days) after thymectomy, while a decrease in the enzymatic activity was observed within later periods and to the end of the experiment (190 and 370 days). Besides, concentration of circulating immune complexes was increased in liver tissue and the most distinct increase occurred within 370 days after thymectomy. Activity of lysosomal enzymes in liver tissue and lymphocytes and content of circulating immune complexes in blood were normalized in thymectomized animals after treatment with thymosin and alimentary factors.

Activities of the enzymes beta-glucuronidase,
acid phosphatase
, acid DNAase, acid RNAase, and acid protease have been measured in the lysosomal and supernatant fractions of mouse liver cells and monkey kidney cells before and after infection with mouse
hepatitis
virus and vaccinia virus, respectively. In the infected cells there was easily measurable release of lysosomal enzymes into the supernatant fraction. Evidence was presented that this is not an artefact of homogenization and precedes cell degeneration demonstrable histologically. It is suggested that release of lysosomal enzymes may explain some of the biochemical changes found in infected cells and may contribute to the cytopathic effects of some viruses.

We have studied the protective effect of chitosan on isoniazid- and rifampicin-induced hepatotoxicity with respect to the changes in the levels of diagnostic marker enzymes (in serum), lipid components and lipid peroxidation (in serum and liver). The oral administration of antitubercular drugs caused a significant elevation in the levels of diagnostic marker enzymes and cholesterol, triglycerides, free fatty acids and lipid peroxidation in serum and liver of experimental rats. There was a slight decline in the level of phospholipids in liver tissue also observed. Co-administration of chitosan significantly prevented the antitubercular drugs-induced elevation in the levels of serum diagnostic marker enzymes (alanine amino transferase, aspartate amino transferase, lactate dehydrogenase,
acid phosphatase
and alkaline phosphatase) in experimental groups of rats. It exerted a significant antilipidemic effect against isoniazid- and rifampicin-induced
hepatitis
by maintaining the levels cholesterol, triglycerides, free fatty acids and phospholipids in serum and liver at near normalcy. A tendency to prevent the isoniazid- and rifampicin-induced lipid peroxidation was also observed. The results of the present study indicated that the hepatoprotective effect of chitosan might be ascribable to its antilipidemic effect and/or antioxidant property.
