The selenium concentration in foods grown and consumed and in plasma, red blood cells, and toenails of people living in the district of Chita in the transbaikalian part of Russia were studied in August 1991. Preliminary results from the area have suggested low selenium intakes and the possible occurrence of cardiomyopathy (
Keshan disease
) in the population. A low selenium concentration in foods grown locally was found: mean selenium concentration in wheat grains was 1, 5, and 28 micrograms/kg, respectively, in three villages studied, that of oats was between 3-6 micrograms/kg, and of cow's milk 10-27 micrograms/kg dry matter. The selenium concentration of bread was considerably higher, between 87-337 micrograms/kg dry wt, presumably because wheat imported from the US had been used for baking. Occasional samples of pork, beef, and mutton contained between 32-218 micrograms selenium/kg dry wt. Low selenium concentrations were observed in samples of soil and river water. The mean plasma selenium concentration of 52 persons was 1.02 mumol/L, including 33 children and 19 adult subjects. The selenium concentrations in red blood cells and toenails were 1.95 mumol/L and 0.61 mg/kg, respectively. No symptoms of
heart disease
caused by selenium deficiency were observed. It is concluded that the selenium status of people was fairly good thanks to the contribution to dietary intake of imported wheat with a high selenium content. As the selenium concentration was very low in foods grown in the area, the selenium intake of the population will be reduced to a very low level if only locally produced foods are consumed.

Keshan
Disease (KSD) is an endemic
heart disease
and moniliformin (MF) has been suggested as one of the etiological factors. In this paper, thin layer chromatographic (TLC) and high pressure liquid chromatographic (HPLC) methods were used for the determination of MF in the rice and corn samples collected from KSD areas and non-KSD areas. One hundred and twenty-three rice samples were analyzed and showed MF contamination in only 8 samples (KSD areas: 8.4% positive; non-KSD areas: 2.5% positive) ranging from 73.6 to 265.3 ng/g (mean concentration: KSD areas 156.3 ng/g; non-KSD areas 179.5 ng/g). One hundred and four corn samples in KSD areas and non-KSD areas were determinated by HPLC method, 45.2% samples were contaminated with MF (KSD areas: 81.4%; non-KSD areas: 19.7%) ranging from 52.3 to 1116.0 ng/g (mean concentration: KSD areas 488.9 ng/g; non-KSD areas 457.4 ng/g). The results showed that the contamination of MF in grains were significantly different between rice and corn, but not between the grains from the KSD areas and non-KSD areas, then casting doubt on the role of MF as an etiological factor of KS.

. Several large population based intervention trials using oral administration of sodium selenite tablets showed significant reduction of
Keshan disease
incidence. Based on the above evidence, it was concluded that selenium deficiency is the major cause of
Keshan disease
, although other etiological factors could not be ruled out. in pigs and damage of tissues including cardiac injury, apoptosis in the liver, reduction in the immune responses in spleen and cerebral lesions in chickens. However, it is well know that plasma biomarkers are not specific and also show alterations in various diseases including those caused by Se deficiency. Therefore, new definition biomarkers are needed to improve disease surveillance and reduce unnecessary chicken losses due to Se deficiency. To identify new biomarkers for Se deficiency, we performed exploratory heart, liver, spleen, muscle, vein, and artery proteomic screens to further validate the biomarkers using Venn analysis, GO enrichment, heatmap analysis, and IPA analysis. Based on the bioinformatics methods mentioned above, we found that differentially expressed genes and proteins are enriched to the PI3K/AKT/mTOR signal pathway and insulin pathway. We further used western blot to detect the expression of proteins related to the two pathways. Results showed that the components of the PI3K/AKT/mTOR signal pathway were definitely decreased in heart, liver, spleen, muscle, vein and artery tissues in the Se deficient group. Expression IGF and IGFBP2 of the insulin pathway were differentially increased in the heart, liver, and spleen in Se deficient group samples and decreased in muscle and artery. In conclusion, 5 proteins, namely PI3K, AKT, mTOR, IGF, and IGFBP2, were differentially expressed, which could be potentially useful Se deficient biomarkers. In the present study, proteomic profiling was used to elucidate protein biomarkers that distinguished Se deficient samples from the controls, which might provide a new direction for the diagnosis and targeted treatment induced by Se deficiency in chickens.
