Proteolytic enzymes seem to play important roles in the life cycles of all medically important protozoan parasites, including the organisms that cause malaria, trypanosomiasis, leishmaniasis, amebiasis, toxoplasmosis,
giardiasis
, cryptosporidiosis and
trichomoniasis
. Proteases from all four major proteolytic classes are utilized by protozoans for diverse functions, including the invasion of host cells and tissues, the degradation of mediators of the immune response and the hydrolysis of host proteins for nutritional purposes. The biochemical and molecular characterization of protozoan proteases is providing tools to improve our understanding of the functions of these enzymes. In addition, studies in multiple systems suggest that inhibitors of protozoan proteases have potent antiparasitic effects. This review will discuss recent advances in the identification and characterization of protozoan proteases, in the determination of the function of these enzymes, and in the evaluation of protease inhibitors as potential antiprotozoan drugs.

Whether purified antigens of Lamblia intestinalis trophozoites can be used to detect these antibodies by immunoassay. The drugs of immunodominant Lamblia antigens were prepared by anion-exchange chromatography of solubilized trophozoite components and they are mainly presented by proteins having molecular weights of 70, 56, and 49 kD. Immunoassay using these antigens revealed antibodies to Lamblia trophozoite antigens in sera of 87.6% of patients with
lambliasis
(its diagnosis was established on the basis of microscopic data on the duodenal content) and only in 16.2% of clinically healthy blood donors. Twenty six sera from patients with
trichomoniasis
having high levels of antibodies to trichomonad antigens were studied to evaluate the specificity of this method for detection of antibodies. It has been found that the proportion of subjects in this group who have also antibodies to Lamblia antigens does not greatly differ from that of healthy blood donors (19.2 and 16.2, respectively).

Secnidazole (hydroxy-2-propyl)-l-methyl-2-nitro-5-imidazole) is an antimicrobic agent. This drug has pharmacological activity against intestinal and hepatic amebiasis,
giardiasis
and vaginal
trichomoniasis
. This paper shows the physicochemical parameters of secnidazole determined during a preformulation study. The determination of the apparent partition coefficient and the profile of solubility in dependence of pH demonstrate the basic characteristic of the drug. The dissolution assay was performed to evaluate its behavior in water. For this purpose, a new spectrophotometric method, which was linear from 5 to 15 microg/ml, sensitive, precise, accurate and selective, was validated to assay the bulk drug. The evaluation of higroscopicity revealed that the drug is unstable above 54% of relative humidity. Rheological properties, such as porosity, tapped and bulk densities and percentage of compressibility were calculated. Results show a bad rheological characteristic for this drug. DSC curves do not show any physical interaction between the drug and the excipients in the compatibility studies.

Secnidazole (alpha,2-dimethyl-5-nitro-1H-imidazole-1-ethanol) is an antimicrobical drug, and it is particularly effective in the treatment of amebiasis,
giardiasis
,
trichomoniasis
, and bacterial vaginosis. Secnidazole crystallizes as a hemihydrate, which belongs to a monoclinic system having space group P2(1)/c, with a = 12.424 A, b = 12.187 A, c = 6.662 A, and beta = 100.9 degrees. The optimized geometries and total energies of different conformers of the secnidazole molecule have been determined by the method of density functional theory (DFT). For both geometry and total energy, it has been combined with B3LYP functionals having extended basis sets 4-31G, 6-31G, and 6-311++G(d,p) for each of the three stable conformers of secnidazole. Using this optimized structure, we have calculated the infrared and Raman wavenumbers and compared them with the experimental data. The calculated wavenumbers are in an excellent agreement with the experimental values. Based on these results, we have discussed the correlation between the vibrational modes and the crystalline structure of the most stable conformer of secnidazole. A complete assignment is provided for the observed Raman and IR spectra.

Metronidazole has been used for the treatment of infections for >45 years and is still successfully used for the treatment of
trichomoniasis
, amoebiasis, and
giardiasis
. Anaerobic bacterial infections caused by Bacteroides species, fusobacteria, and clostridia respond favorably to metronidazole therapy. Good clinical results in the treatment of vaginosis due to Gardnerella vaginalis have also been reported. Rates of resistance to metronidazole are still generally low; however, several studies have reported decreased susceptibility among Bacteroides species, as well as different mechanisms of resistance. Metronidazole-resistant Helicobacter pylori strains have been described, but combination therapy (eg, metronidazole, amoxicillin, or clarithromycin plus omeprazole) is still recommended for eradication of this pathogen in patients with gastroduodenal ulcers. Metronidazole is considered to be a cost-effective drug because of its low cost, good activity against pathogenic anaerobic bacteria, favorable pharmacokinetic and pharmacodynamic properties, and minor adverse effects. Metronidazole is still the criterion standard for therapy of anaerobic infections, as was described by Tally and colleagues 35 years ago.

The cellular plasma membrane contains enzymes whose active sites face the external medium rather than the cytoplasm. The activities of these enzymes, referred to as ecto-enzymes, can be measured using living cells. Ecto-phosphatases are ecto-enzymes that presumably hydrolyze extracellular phosphorylated substrates, releasing free inorganic phosphate. Although, several alternative functions have been suggested for these enzymes, such as participation in proliferation, differentiation, adhesion, virulence, and infection, little is known about the physiological roles of these enzymes in protozoa parasites. In this review, we discuss the principal features of ecto-phosphatases in protozoan parasites that are causative agents of important diseases such as Chagas' disease, leishmaniasis, amoebiasis,

Metronidazole is a treatment of choice for several types of infections, but coexisting conditions or concomitant medications may preclude its use. Although tinidazole, a newer nitroimidazole, may be an option in cases where drug interactions make the use of metronidazole inadvisable, similar absolute contraindications exist. Parasitic protozoa are among the most important pathogens worldwide. Diseases such as malaria, leishmaniasis, amoebiasis,
giardiasis

trichomoniasis
, and trypanosomiasis affect millions of people. Humans are constantly threatened by infections caused by these pathogens. Parasites engage a plethora of surface and secreted molecules to attach to and enter mammalian cells. The secretion of lytic enzymes by parasites into host organs mediates critical interactions because of the invasion and destruction of interstitial tissues, enabling parasite migration to other sites within the hosts. Extracellular matrix is a complex, cross-linked structure that holds cells together in an organized assembly and that forms the basement membrane lining (basal lamina). The extracellular matrix represents a major barrier to parasites. Therefore, the evolution of mechanisms for connective-tissue degradation may be of great importance for parasite survival. Recent advances have been achieved in our understanding of the biochemistry and molecular biology of proteases from parasitic protozoa. The focus of this paper is to discuss the role of protozoan parasitic proteases in the degradation of host ECM proteins and the participation of these molecules as virulence factors. We divide the paper into two sections, extracellular and intracellular protozoa.
