The failure of clearance mechanisms in the mare's uterus results in persistent inflammation and is considered a major cause of subfertility. Eighteen mares, of which three were susceptible to endometritis and four had been ovariectomised, underwent charcoal clearance testing to evaluate their clearance mechanisms. This consisted of installing 500 mg of charcoal (particle size 4 to 90 microm) added to 50 ml of phosphate-buffered saline (PBS) into the uterus. Forty-eight hours later the uterus was flushed out with 0.0012 per cent methylene blue in 50 ml of PBS for determination of the diluting factor by photospectrometry. Flush volume, pH, specific gravity and pellet size were all analysed. To investigate the effect of a beta2-adrenergic agonist on the ability of genitally healthy oestrous mares to eliminate a suspension of charcoal from the uterus, four genitally healthy mares with appropriate charcoal clearance were also subjected to clearance testing following intravenous administration of 0.8 microg/kg of clenbuterol every 12 hours and 1 microg/kg of clenbuterol every eight hours. All parameters were within their normal range following clenbuterol treatment. However, minimal but significant differences in pre- and post-treatment values regarding fluid volume and extinction rate were recorded.

Bisected bovine embryos were co-transferred with trophoblastic vesicles (TVs). These TVs were prepared by dissection of conceptuses that were collected by uterine flushing after culture for seven days in the uterus following transfer of embryos derived by in vitro fertilization (IVF). Pregnancy diagnoses were performed twice, between Day 26 and Day 43 and between Day 38 and Day 73 post-estrus by ultrasonography. The pregnancy rate was significantly increased at first pregnancy diagnosis when demi-embryos were transferred with TVs (66.7%, 16/24) compared with the control group (34.5%, 10/29) (P < 0.05). Three losses occurred in the co-transfer group between the first and second pregnancy diagnosis. The final pregnancy rates according to delivered offspring were 41.7% (10/24) and 27.6% (8/29), respectively. There were no statistically significant differences between the pregnant and non-pregnant groups with regard to the average diameter of the TVs measured before transfer at three points during the gestation period. The birth weight and gestation lengths of the offspring were almost the same for the co-transfer and control groups. In the co-transfer group, the genetic identities of calves from the separated embryos were not affected by the TVs, as confirmed by parental blood type testing. Delivered offspring in co-transferred groups showed normal morphology. In conclusion, the present study indicates that co-transfer of TVs prepared from conceptuses cultured in vivo following transfer of IVF embryos enhances the fertility of demi-embryos during the early stages of pregnancy, as has similarly been shown in previous research for those prepared from in vivo embryos.

High plasma urea nitrogen (PUN) concentrations are associated with decreased fertility in lactating dairy cows. Our objective was to evaluate the quality of embryos flushed from superovulated lactating cows having moderate or high PUN concentrations. Subsequent embryo survival was determined after transfer to recipient heifers with either low or high PUN. Lactating Holstein dairy cows (n = 23; 50-120 days in milk) were randomly assigned to one of two diets designed to result in moderate or high PUN concentrations (15.5 +/- 0.7 and 24.4 +/- 1.0 mg/dl, respectively; P < 0.001) and were fed for 30 days before embryo flushing and recovery. Embryos (n = 94) were evaluated morphologically, frozen and subsequently transferred into synchronized virgin heifers that were fed one of two diets designed to result in either low or high PUN concentrations (7.7 +/- 0.9 and 25.2 +/- 1.5 mg/dl, respectively; P < 0.001; 2 x 2 factorial design). The number, quality and stage of development of recovered embryos were similar for cows with moderate or high PUN. Transfer of embryos from moderate PUN donor cows resulted in a higher pregnancy rate (35%; P < 0.02) than the transfer of embryos from high PUN donor cows (11%). Pregnancy rate was not affected by either recipient diet or the interaction of donor and recipient diets (P > 0.05). These results indicate that high PUN concentrations in lactating dairy cows decrease embryo viability through effects exerted on the oocyte or embryo before recovery from the uterus 7 days after insemination.

This paper presents the successful use of a non-surgical, transcervical uterine lavage technique for the treatment of uterine infection-induced infertility in three female large cats. We developed a non-surgical uterine lavage technique, which allowed repeated flushing of the uterine lumen and installation of therapeutic antibiotics. The entire procedure was performed under general anaesthesia (duration of anesthesia ranged from 40 to 70 min). It was successfully applied in a Sumatran tiger (Panthera tigris sumatrae), a Corbett tiger (Panthera tigris corbetti) and an Amur leopard (Panthera pardus orientalis). The tigers were treated only once, whereas the leopard received four uterine treatments, due to re-infection after mating. Decisions to conduct uterine treatments were based on detection of uterine fluid during previous transrectal ultrasound examinations. The catheter was guided into the vagina, with the aid of an endoscope, passing the urethra, and then into the uterus, with the aid of transrectal ultrasonography. Both uterine horns were separately flushed with approximately 300 mL of cell medium M199, followed by an antibiotic infusion. Upon ultrasonographic re-examination, the topical uterine treatments resulted in an apparent decline in the inflammatory and/or degenerative processes. The Corbett tiger had the most severe uterine alterations, in addition to an aseptic pyometra. As a result, she was treated 1 month prior to ovariohysterectomy (in order to reduce the surgical risk). The Sumatran tiger was artificially inseminated twice after hormone-induced estrus, and the Amur leopard expressed a spontaneous estrus and re-initiated mating behaviour.

Endometriosis is an estrogen-dependent gynaecological disease associated with pain and infertility, which occurs in humans and menstruating primates. In this study, the marmoset monkey (Callithrix jacchus), which is a non-menstruating primate with high circulating estrogen levels, was used to test firstly the hypothesis that endometriosis is based on uterine shedding into the peritoneal cavity, secondly to study the pathogenesis of endometriosis due to its estrogenic situation. Female marmoset monkeys (n = 29) were exposed to two different experimental procedures (non-invasive versus invasive) for intrapelvic placement of endometrial cells by uterine flushing over an experimental period of 2-3 years. First endometriotic foci were detected by colour Doppler ultrasound at the bladder, the uterus and the ovaries at the earliest after 4 months of either treatments. However, invasive induction was more effective in terms of the time-course of induction and the number of resulting endometriotic foci. The analysis of the endometriotic foci by histology, immunohistochemistry and molecular techniques allowed a division into two distinct groups: an initial developing stage occurred, which under further treatment led to the second stage of established endometriosis. Both procedures showed a treatment-dependent increase of vascular supply to the endometriotic foci over the experimental period. The invasive method induced the final established stage of endometriosis more rapidly, with the expression of steroid receptors, aromatase, 17betaHSD1 and CD10. Altogether, 72% of the treated marmoset monkeys developed endometriosis under our endometrial reflux protocols. Our data support the theory that endometriosis can be induced artificially in a non-menstruating primate (C. jacchus) by endometrial shedding into the peritoneal cavity. Because the marmoset is a primate with very high peripheral estrogen levels, this offers an interesting model for studying the pathogenesis of this estrogen-dependent disease, as well as for therapeutic impacts on enzymes involved in steroid metabolism.

In two trials, eight attempts were made to collect fertilized ova from dairy goats by nonsurgical methods. In both trials the cervix of each doe was dilated by inserting a Laminaria japonica tent device into the cervical canal prior to flushing. In Trial 1, an attempt was made to collect embryos from four nonsuperovulated does by flushing phosphate-buffered saline (PBS) through a rigid pipette. Little fluid and no embryos were recovered from the does. All four donors were in estrus two days after the procedure. In the second trial, FSH-superovulated does were collected on day 5 following estrus. The donors were anesthetized, and a modified Foley catheter was passed through the cervical canal. In three does, a 24 ga. two-way Foley was stiffened with a size 10 (French) polypropylene catheter which penetrated the Foley, extending 7 cm beyond the tip. Recovery of flushing medium with this device was minimal, and laparotomy of one doe revealed a punctured uterus. Replacement of this device with a different catheter, through which a polypropylene catheter (size 5 Fr.) penetrated only 1 to 2 cm, resulted in satisfactory return of infused PBS, and recovery of two blastocysts and one degenerated ovum from this doe. Use of the same device on a second doe without laparotomy resulted in collection of seven blastocysts and three degenerated ova. Of three observed donors that received Laminaria tents (including one which was not flushed) two were in estrus three days after the procedure, while unused synchronized recipients showed normal cycle lengths. Surgical transfer of two blastocysts from each donor to each of two synchronized recipients resulted in the birth of twin kids from one recipient doe. The study demonstrates the feasibility of embryo collection from dairy goats by nonsurgical means.

To allow for the nonsurgical collection of swine embryos, the uteri of sows (n=7) were surgically shortened. A section of each uterine horn was resected to facilitate a transcervical flushing procedure. All sows with a shortened uterus exhibited natural estrus at least once after the operation. Four to six days after insemination, embryos were collected with a two-way Foley catheter. Embryos were collected (n=55, 6.3+/-6.0: x +/-SD ) from every treated sow. Although treated sows often did not exhibit estrus beyond 1 to 9 natural estruses, those sows (n=27) with persistent corpora lutea (CL) over a 4 to 5 wk period were given prostaglandin F(2alpha) (PGF(2alpha)) and they returned to estrus in 5.2+/-1.1 d: x +/-SD .

Five cyclic primiparous sows were used to test a surgical procedure for in vivo transcervical collection of pig embryos. The procedure consisted of shortening the uterine horns. After surgery, all sows returned to estrus and embryos were recovered following artificial insemination. Transcervical uterine flushing was carried out in four sows. On average 3.6 +/- 1.5 (mean +/- SD) embryos were recovered from the five sows. The results indicate that it is possible to recover embryos transcervically from sows with a resectioned uterus.

The ovarian response of 25 buffalo-cows was visually assessed, and their oviducts and uteri separately flushed 3 to 6 d post superovulatory estrus at slaughter. Ten buffalo-cows slaughtered on Days 5 and 6 were examined per rectum for corpora lutea (CL) and follicles > 8 mm prior to slaughter, and the estimate was compared later with the actual ovarian response. Five out of the ten buffalo-cows were nonsurgically flushed in vivo on Day 5 of the estrous cycle, a day before slaughtering, and as a result, six ova/embryos were recovered. After the flushing of the reproductive tract at slaughter, one more ovum was recovered from the uterus of each of the three buffalo-cows. As a result of treatment of three groups of five buffalo with 3000 IU pregnant mare serum gonadotrophin (PMSG) on Days 6, 10 or 14 of the estrous cycle, 3.8, 6.2 and 3.4 CL on the average were recovered, respectively (Experiment I). A mean number of 8.8 and 9.0 CL, respectively, was obtained in two groups of five buffalo each, after treatment with 40 mg of follicle stimulating hormone (FSH) on Day 10 of the stage of the estrous cycle (Experiment II) and 3000 IU PMSG regardless of the stage of cycle (Experiment III). The percentage of ova/embryos recovered in the three experiments was 32.8, 20.4 and 22.2, respectively.

Beef cows were superovulated with follicle stimulating hormone (FSH) to compare two nonsurgical methods of embryo recovery from the uterus. In the first method each uterine horn was independently flushed with physiological saline solution (PSS) through a Foley catheter passed through the cervix and into the uterine horn. In the second method both uterine horns were simultaneously flushed with PSS by passing the catheter into the uterine body. In both methods, the numbers of ovulations were determined after embryo collection by counting the corpora lutea (CL) on both ovaries of each cow through a flank incision. Independent flushing (n = 19) averaged 6.4 embryos and 16.1 CL per cow for a recovery rate of 40%. Simultaneous flushings (n = 22) averaged 5.4 embryos and 17.7 CL per cow for a recovery rate of 31%. This difference between the recovery rates of the two flushing methods was not significant (P>0.05).
