We used in situ gastrocnemius muscle of anaesthetized dogs to test the hypothesis that O2 radical production during muscle contraction contributes to
fatigue
. Muscle tension was measured with a force transducer and blood flow was monitored with an electromagnetic flow probe. Muscle contractions were produced by stimulating the nerve for 15 min at 20 Hz, 12 trains/min, and a duty cycle of 0.25. Three groups of seven animals were given an infusion of 0.2 mL.min-1 of either saline, low-dose oxygen radical scavengers (250 IU.mL-1 superoxide dismutase, 640 IU.mL-1
polyethylene glycol
(
PEG
)-catalase, 0.25 mg.mL-1 deferoxamine, and 0.1 mg.mL-1 oxypurinol), or high-dose oxygen radical scavengers (3300 IU.mL-1 superoxide dismutase, 6600 IU.mL-1
PEG
-catalase, 2.5 mg.mL-1 deferoxamine, and 0.1 mg.mL-1 oxypurinol). Blood flow and vascular resistance of the gastrocnemius muscle during stimulation did not differ among groups. After 15 min of stimulation, the developed tension (represented as a percentage of initial tension developed) was 66 +/- 7% in the saline treated group, 70 +/- 6% in the low-dose group, and 70 +/- 4% in the high-dose group. The change in tension during recovery was not significant in the control or low-dose groups. However, there was partial recovery in the high-dose group. In conclusion, in this preparation, oxygen radical scavengers did not delay the development of decreased muscle tension.

The purpose of this study was to evaluate the usefulness of ratings of perceived exertion (RPE) as an indicator of exercise intensity in patients with chronic obstructive pulmonary disease (COPD). The subjects were ten male patients with COPD, whose mean forced expiratory volume in 1 s was 1.09 1, SD 0.41, and ten healthy middle-aged men.
Ramp
incremental exercise on a cycle-ergometer was performed and RPE was determined by the Borg 15-point scale. The absolute oxygen uptake at each RPE was significantly greater in the healthy subjects than in the patients with COPD. However, oxygen uptake calculated as a percentage of maximal at any RPE did not differ significantly between the two groups. Arterial blood lactate concentration at points 15 to 19 on the RPE scale was increased in healthy subjects (P less than 0.05-P less than 0.001), while the dyspnoea index at points 11 to 19 on the RPE scale was higher in patients with COPD (P less than 0.05-P less than 0.001). The main complaints on stopping exercise were dyspnoea in the patients with COPD and
fatigue
in the healthy subjects. Although the nature of RPE may have been different in the two groups, RPE could be a possible indicator of exercise intensity when physicians prescribe exercise to patients with COPD.

polyethylene glycol
conjugate of L-asparaginase (PEGLA) was administered to 21 patients with refractory non-Hodgkin's lymphoma. The dose given was 2,000 mu/m2 intramuscularly every 2 weeks. Eligibility required at least one prior trial of chemotherapy and ambulatory performance status. At entry, all patients had measurable lesions and documented disease progression. The median age of the patients was 61 years; 18 (86%) were ambulatory with minimal symptoms, 12 patients (57%) had 3 or more prior regimens, and 13 (62%) had stage IV disease. Histologic subtype was low grade in 11 patients (52%), intermediate in 7 (33%), high grade in 2 (10%) and unclassifiable in one (5%). There were two partial responses (11%) noted (95% confidence interval of response of 1-30%). Eleven patients (52%) were removed from study due to disease progression. Nine patients (43%), required removal for toxicity (7 for protracted nausea and vomiting and 2 for confusion). One patient died of sepsis while on study but this was not considered drug related. Almost one third of patients complained of
fatigue
or loss of appetite. Nausea and vomiting occurred in approximately half the patients and was moderate to severe in 9. Diarrhea and abdominal pain were also noted in one-third of those treated. Changes in the partial thromboplastin time and fibrinogen were noted in most patients but resulted in no bleeding complications. In this trial, PEGLA displayed modest activity in a heterogenous group of patients with progressive non-Hodgkin's lymphoma.

The present work is a study of the effects of soft segment molecular weight and chemical structure on the
fatigue
crack propagation of model copoly (ether-urethane-urea)s (PEUU). The PEUU were synthesized using polypropylene glycol (PPG), polytetramethylene glycol (PTMG), and
polyethylene glycol
(
PEG
) as the soft segment component. The number average molecular weights of the polyethers were within the range of 1000-2000. Methylene bis (4-phenylisocyanate) (MDI) and ethylene diamine were used as the diisocyanate and the chain extender, respectively. The cyclic loading experiments were carried out using a computerized film stretcher that can conduct sinusoidal operation at a constant strain amplitude, strain rate, and frequency. crack propagation (FCP) rate were selected to characterize the
fatigue
behavior of the model polyurethanes. An empirical equation was applied to define the
fatigue
properties of model polyurethanes and to evaluate the
fatigue
resistance. To investigate the effect of molecular variables on the FCP, the morphological changes caused by structural differences and cyclic stress were determined using dynamic viscoelastometer (Rheovibron), Small Angle X-ray Scattering (SAXS), and Fourier-Transform Infrared (FT-IR) spectroscopy. Mooney-Rivlin plot was used to determine the crosslink density variation. In addition the orientation behavior at the crack tip was characterized by IR dichroism technique using a polarized FT-IR microscope. The results indicated a reasonable relationship between the FCP rates and the hard segment content, crosslink density, and deformation property at the crack tip. However, the initial stage of phase separation and domain disruption behavior did not show a good correlation with the FCP properties of model polyurethanes. Among the model polyurethanes tested, the PEUU with PTMG (Mn = 1000) exhibited the best

. Much of this work has been done, however, using in vitro muscle preparations; the purpose of the present study was to assess the effect of scavengers on in vivo diaphragm contractile function. To accomplish this, we compared the rate of development of
fatigue
of the electrically stimulated diaphragm in four groups of dogs: (1) animals given intravenous
polyethylene glycol
adsorbed superoxide dismutase (
PEG
-SOD, 2,000 units/kg) 1 h before a
fatigue
trial; (2) a group given intravenous dimethylsulfoxide (DMSO, 0.5 ml/kg of a 50% solution) before
fatigue
; (3) a group given saline before
fatigue
; and (4) a group treated with denatured
PEG
-SOD (2,000 units/kg) before
fatigue
. We measured diaphragmatic concentrations of thiobarbituric acid reactive substances (TBAR), a marker of free radical-mediated lipid peroxidation, on muscle samples taken at the conclusion of

Sixteen normal, non-smoking first year medical students underwent four standard exercise tests, while cardiovascular (heart rate and blood pressure) and pulmonary (respiratory rate, tidal volume and oxygen consumption) adjustments were being monitored. Maximal exercise level for all tests were defined following the Jones scale. No variation was noted in respiratory rate among the four exercise tests. The cycle ergometry (CE), hand ergometry (HE), and treadmill (TM) exercise tests produced progressive increases of the various parameters as expected, although target maximal heart rates were not reached in most cases secondary to muscle
fatigue
. The step test (ST) approximated the physiological parameter changes noted in these three tests during the early stages of exercise, but levelled off after 3 minutes, probably due to lack of incremental load. The ST took the longest time to reach maximal level of exercise parameters, and recorded the lowest tidal volume increase. The same subjects underwent a modified step test (MST1) by adding 1 kg sandbags every 3 min to backpacks worn by the subjects, to provide incremental load. This modification provided a slight increase in the parameters measured, but still plateaued after 3 min. Another set of 18 normal, non-smoking medical students underwent bicycle ergometry and two other modified exercise tests. The modified step test two (MST2) was similar to our initial MST1, except that the sandbags added every 3 min were not fixed at 1 kg but were also incremental (1, 2, 3 and 4 kg). The other test (

test or RT) required the subjects to walk up and down a ramp, which entailed not only the addition of incremental weights of sandbags every 3 min, but also increasing the elevation of the ramp at each stage. Data analysis showed that these two tests showed almost similar changes in cardiovascular and respiratory adjustments in the subjects, compared to the standard bicycle ergometry. These pilot studies showed that low-cost modified step tests may be utilized to approximate the expensive standard treadmill and bicycle exercise tests. More studies on larger populations have to be done to validate these results.

Bryostatin 1 is a protein kinase C partial agonist which has both antineoplastic and immune-stimulatory properties, including the induction of cytokine release and expansion of tumour-specific lymphocyte populations. In phase I studies, tumour responses have been observed in patients with malignant melanoma, lymphoma and ovarian carcinoma. The dose-limiting toxicity is myalgia. Sixteen patients (age 35-76 years, median 57 years) with malignant melanoma were treated. All had received prior chemotherapy. In each cycle of treatment, patients received bryostatin 25 degrees g m(-2) weekly for three courses followed by a rest week. The drug was given in PET diluent (10 microg bryostatin ml(-1) of 60%

, 30% ethanol, 10% Tween 80) and infused in normal saline over 1 h. The principal toxicities were myalgia (grade 2, eight patients and grade 3, six patients) and grade 2 phlebitis (four patients),
fatigue
(three patients) and vomiting (one patient). Of 15 patients evaluable for tumour response, 14 developed progressive disease. One patient developed stable disease for 9 months after bryostatin treatment. In conclusion, single-agent bryostatin appears ineffective in the treatment of metastatic melanoma in patients previously treated with chemotherapy. It should, however, be investigated further in previously untreated patients.

A microcapsule suspension, a substitute for animal blood in hemolysis tests, has been developed for evaluation of the absolute hemolytic properties of circulatory artificial organs. The microcapsule suspension was made by dispersing microcapsule slurry into an
ethylene glycol
sodium chloride solution. The microcapsule slurry was composed of a leuco dye solution and polyurethane membrane made by the reaction between aliphatic poly-isocyanate and polyamine by interfacial polycondensation. The microcapsule was a small particle containing dye inside. The microcapsule suspension was white; the diameter of the microcapsules was from 5 to 100 microns. The specific gravity of the suspension was 1.024, and the membrane was elastic. The fluid showed Newtonian characteristics, different from animal blood, and its viscosity was approximately 5.8 mPa.s. After the microcapsules were destroyed, the leuco dye was extracted with n-hexane from the suspension and was measured by spectroscopy after being colored with acid ethanol. Hemolysis can be regarded as a
fatigue
fracture of cell membranes rather than a static fracture. The destruction of microcapsules by a Potter type tissue grinder was observed at a low stroke number region and was compared to rat blood. Moreover, hemolysis tests of a commercially available centrifugal blood pump and the prototype of our centrifugal pump for mechanism checks were carried out with bovine blood. The hemolysis level of the prototype pump increased with time while the hemolysis level of the commercial blood pump did not change as much as that of the control when both pumps were tested with the microcapsule suspension. These results are similar to tests utilizing bovine blood. Therefore, hemolysis tests of circulatory artificial organs completed with microcapsule suspension are expected to provide results similar to tests with animal blood.

The mechanical stability of gels applied for entrapment and retention of biocatalysts in bioreactors is of crucial importance for successful scale-up applications. Gel abrasion in agitated reactors will depend on liquid shear, bubble shear, and wall shear, as well as collisions between the gel particles. As a simplified standardized model system, abrasion of gel beads was studied in 1-m-high bubble columns with controlled aeration, and quantified by measuring the loss of gel material into solution. Gel beads were also taken out to measure stress-strain response during controlled compression. More general rheological properties of different gels were studied by applying a variety of regimes of controlled compression of standardized gel cylinders: Gel strength was measured by recording the fracture properties and the Young's modulus. Viscoelastic properties were revealed by recording creep during compression as well as recovery after compression. Oscillation tests up to 1000 cyclic compressions were applied to compare the
fatigue
of different gels. Results obtained for Ca-alginate gels, gels of chemically modified polyvinyl alcohol with stilbazolium groups (PVA-SbQ) as well as mixed gels of Ca-alginate and PVA-SbQ are compared with previously published data for kappa-carrageenan, agar, and
polyethylene glycol

PEG
) gels. It is concluded that material
fatigue
rather than mechanical properties such as stiffness or fracture stress should be considered when selecting a suitable gel material on the basis of abrasion resistance. The very soft and superelastic PVA-SbQ gel showed no significant

in mechanical tests and no abrasion was detected in the standardized model system used. Ca-alginate gels, however, showed severe irreversible changes due to
fatigue
at oscillating loads and creep at constant load. Due to their similarities with kappa-carrageenan gels in mechanical tests, it is likely that Ca-alginate would also be sensitive to abrasion. Mixed gels of Ca-alginate and PVA-SbQ represent a complex system with intermediate properties, showing significant

and creep, but elastic properties from the PVA-SbQ gel make it less sensitive than the pure Ca-alginate gel.
