Helminth infection has a potent systemic immunomodulatory effect on the host immune response, which also affects the development of autoimmune diseases. We investigated the dose-dependent influence of Trichinella spiralis infection on experimental autoimmune encephalomyelitis (EAE). Our model of concomitant T. spiralis infection and EAE demonstrates that established infection of Dark Agouti (DA) rats with the parasite causes amelioration of the clinical course of induced EAE in a dose-dependent way. Infection with T. spiralis L1 stage muscle larvae (TSL1) reduced the severity of the autoimmune disease as judged by lower maximal clinical score, cumulative index, duration of illness and degree of mononuclear cell infiltration in T. spiralis infected animals compared to control, EAE-induced group. This study provides a valuable model of worm infection to investigate helminth-induced regulatory mechanisms for optimal benefit to the host.

A seroepidemiology survey of nine zoonoses was carried out in 2007 on 90 healthy adult volunteers in Viljujsk, a northern city in the Republic of Sakha (Eastern Siberia). The seroprevalence of Lyme borreliosis was 3.3% by immunofluorescence. None of the subjects displayed a positive enzyme-linked immunosorbent assay/Western blot result for alveolar or cystic echinococcosis. The seroprevalence of toxocariasis by Western blot was 4.4%, and 8.9% of the subjects had anti-Toxoplasma IgG. By enzyme-linked immunosorbent assay, the seroprevalence of trichinellosis was 4.4%. Three subjects were simultaneously positive for tick-borne encephalitis and West Nile infection, so no clear diagnostic conclusion could be reached for these flavivirus diseases. Interestingly, Crimea-Congo hemorrhagic fever had an 11.1% seroprevalence rate, indicating that Viljujsk is the most northern focus of this infection. Additionally, this finding suggests a potential involvement of Crimea-Congo hemorrhagic fever agent, or of another member of the Bunyaviridae family, in the genesis of the so-called Viljujsk encephalomyelitis.

Helminths, as complex pathogens, possess a large number of different epitopes, some of which may be similar to the epitopes of the host. Besides being the cause for the activation of self-reactive immune cells, molecular mimicry may also be the cause for the expansion of regulatory T cells, crucial for the host tolerance of self-antigens. Amelioration of experimental autoimmune encephalomyelitis (EAE), animal model of multiple sclerosis (MS), caused by Trichinella spiralis infection or application of its muscle larvae excretory-secretory products (ES L1), was achieved through activation of Th2 and regulatory responses. The present study aimed to reveal whether the cause of observed immunomodulation could be the existence of shared epitopes between ES L1 antigens and auto-antigens. Serum samples from 92 MS patients were tested in Western blot for the reactivity toward components of ES L1. Immunoglobulins from the sera of MS patients recognized several ES L1 components, but 45, 49 and 58 kDa proteins dominated others by the frequency of interaction. According to the logistic regression analysis, these interactions were statistically significantly associated with MS, regardless of the disease phenotype or severity. Selected molecules might share homology with self-antigens and as such are worthy of further investigation in terms of potential immunomodulatory capacity and involvement in the parasite's provoked amelioration of EAE.
