Weanling Wistar rats of both sexes were fed diets containing 0 (control), 1% and 5% ground sclerotia of Sclerotinia sclerotiorum derived from infected rapeseed (Brassica napus). Body weight, feed consumption and clinical appearance were monitored over an 84-day period. Blood samples were collected on days 41 and 84 and necropsies performed on day 84. Weight gain and feed consumption were similar in the control and 1% groups. In the 5% group, weight gain was depressed, feed wastage was greater and at termination more than half the rats were in poor body condition with alopecia and hyperkeratosis of the tail. These effects were probably nutritional and due to unpalatability of the diet. Blood urea nitrogen and blood glucose concentrations did not vary consistently among the groups. Serum glutamic-pyruvic transaminase activity was significantly depressed (p less than 0.001) by consumption of sclerotia. This depression was dose-related and consistent on days 41 and 84. There were no significant differences (p greater than 0.05) between groups in the ratios of liver weight and kidney weight to body weight.

Oral contraceptive (OC) use has been associated with 50 different metabolic changes but few women require increased amounts of nutrients to prevent deficiencies. Plasma triglyceride levels are markedly increased by OCs, but no consistent changes have been found in plasma cholesterol, fatty acids, or phospholipids. Small elevations in blood glucose and plasma insulin levels result from OC use, and plasma albumin is decreased and the alpha and beta globulins and fibrinogen are increased. Women on the pill show slight increases in the urinary excretion of some of the amino acids and decreases in some of the blood amino acids. Tryptophan metabolism is altered by OC use; changes in parameters of Vitamin-B6 metabolism are seen and Vitamin-B6 is used as a cofactor for several enzymes in the tryptophan pathway. At the beginning of OC use the retention of dietary nitrogen increases, and weight gain may result. The estrogens in OCs reduce plasma calcium, phosphorus, and magnesium. Most studies demonstrate an increase in serum iron and copper and a decrease in plasma zinc. Studies have also found an increase in plasma levels of Vitamin-A and a decrease of carotene, Vitamin-E, ascorbic acid, folacin, Vitamin-B12, and Vitamin-B6. 20% of OC users have enlarged cervical and vaginal cells as a result of abnormal folacin metabolism. The abnormality is corrected by oral folacin supplementation. Some women respond to OC treatment with biochemical signs of Vitamin-B6 deficiency and depression. These women should receive 20-40 mg Vitamin-B6 as a supplement.

Several pivaloyl and pivaloyloxy esters and amides of dopamine were synthesized for possible antiparkinson activity. The compounds were synthesized by select O- and N-acylation and N-methylation procedures. The tert-butoxycarbonyl function is an effective and easily removed nitrogen-protecting group for dopamine. Preliminary biological testing results showed that all compounds tested elicited a hypothermic response in mice, while only O,O-dipivaloyl-N,N-dimethyldopamine reversed reserpine-induced motor depression in mice. However, it is difficult to conclude from the preliminary data that the observed biological effects were due to central dopaminergic receptor stimulation.

The phenotypes of Azotobacter vinelandii ATCC 12837 strains defective in nitrogen fixation (Nif-) were characterized by intrageneric transformation with known Nif- strains of A. vinelandii OP. These former mutant strains were used as recipients for intergeneric transformation by deoxyribonucleic acid (DNA) prepared from Rhizobium spp. to determine if the rhizobia would transform the Azotobacter Nif- phenotypes to Nif+. The frequency of Nif+ transformants using Rhizobium DNA was always less than the frequency using Azotobacter wild-type DNA but was greater than the spontaneous reversion frequency. The Azotobacter Nif+ recombinants also were stable. DNA from all of the Rhizobium spp. transformed to Nif+ Azotobacter mutants defective in the nitrogenase component I (molybdoferredoxin); however, some recombinants had a lower nitrogenase activity and a delayed nitrogenase depression time. Mutants defective in the pleiotrophic transcriptional control of both nitrogenase components were transformed to Nif+ by the asymbiotic nitrogen fixing Rhizobium sp. 32H1 and 41A1, but not the symbiotic nitrogen-fixing species. The significance of these results and the possible future applications of this system are discussed.

Unanesthetized rats put in the hermetic chamber breathed with a gas mixture containing 10.5% of oxygen in nitrogen during 30 and 60 min (moderate hypoxia), and 3.5% of oxygen in nitrogen for 30 min (severe hypoxia). The circulating blood volume increased in moderate hypoxia but was rather reduced in severe hypoxia. Mobilization of blood from the liver occurred in moderate hypoxia after 30 min, becoming less obvious after 60 min or in severe hypoxia. Redistribution of blood in the myocardium, skeletal muscles of the head and neck occurred in moderate hypoxia. Severe hypoxia led to depression of the regional circulating blod redistribution and the signs of pathological accumulation of the blood in the liver, small intestine, colon and testicles.

The effects of the pesticides, amitrol, a derivative of amitrol (viz. 3,5-diamino-1,2,4-triazole), diquat, paraquat, linuron, MCPA, malathion, and monuron, were studied on the nitrogen-fixing algae, Anabaena cylindrica, Aulosira sp., Calothrix elenkenii. Chlorogloeae frischii, Cylindrospermum muscicola, Nostoc sp. from Collema tenax, Nostoc muscorum Tolypothrix tenuis, and Westiellopsis sp. In general, two types of response were discernible; an initial period of depression succeeded by an increased activity and an initial period of depression followed by a distinct decrease on prolonged incubation. The results indicate that some pesticidal compounds can severely limit the nitrogen-fixing capacities of blue-green algae, thereby affecting the overall nitrogen economy of soils in general.

Various nitrogen analogs of delta6a,10a-tetrahydrocannabinol were synthesized by a general procedure described in an earlier communication. Minimum effective doses (MED50's) and lethal doses (LD50's) were determined by a modified Irwin mouse screen after iv administration of compounds in PEG 200. The most potent compounds were the propargyl (5t), allyl (5m), and chloroallyl (5o-q) derivatives. Overt behavioral effects (CNS depression, static ataxia, and hypersensitivity) of 5t and Roger Adams' carbocyclic analog (III) were found to be similar in the mouse, cat, dog, and monkey. Dichloroisoproterenol prevented and reversed many of the depressant effects of both III and 5t but had no effect on the ataxia produced by these compounds. In antinociceptive tests, 5t was active in the phenylquinone and Eddy hot-plate tests but was inactive in the tail-flick test.

The present study was designed to develop an animal model applicable to the clinical patient in the investigation of the pathogenesis of septic shock. The model currently described is a lightly anesthetized, unrestrained monkey, carefully monitored during a 24 hour observation period. Varying doses of live Excherichia coli organisms were infused intravenously during a 30 minute period, and a variety of hemodynamic, respiratory and metabolic parameters were monitored. Doses of organisms varied between 7.6X10(9) and 3.0X10(11) organisms per kilogram of body weight, and there was no obvious correlation between size of dose and survival time. Two of nine experimental monkeys survived the Excherichia coli, while times of death of the remaining monkeys varied between three and 27 hours. Two control monkeys, not administered organisms, survived the 24 hour period with minimal changes in all measured parameters. Results reveal two patterns in response to organism administration. These were early acute death, after three to four hours, and prolonged life, death after 20 to 27 hours. The acute response was characterized by marked systemic hypotension, hypoglycemia, hypoinsulinemia, increased lactate level, decreased pH or respiratory depression. The other type of response involved profound sustained hypotension with hypoglycemia and hypoinsulinemia in most monkeys and elevations in lactate, blood urea nitrogen potassium creatinine, serum glutamicoxalacetic, lactic dehydrogenase and fractionatedlactic dehydrogenase levels. Depressions in respiration were not evident in the group which survived a longer period of time. Renal fibrin thrombi, prominent in baboons administered Escherichia coli, were absent in the rhesus monkey regardless of the size of the dose of organisms. The results of this study suggest the operation of a multifactiorial mechanism in septic shock with interactions between hemodynamic and metabolic factors varying within the species.

Trans-1,2-dichloroethylene (t-DCE), an industrial solvent, proved to be moderately toxic when studied in small laboratory animals. In adult female rats brief (8 h) and prolonged (8 h daily, on 5 consecutive days a week, for more than 16 weeks) inhalation of 200 ppm--the current TLV/MAC in various countries--produced histological evidence of slight to severe fatty degeneration of the liver lobules and Kupffer cells. In addition marked pulmonary hyperaemia and alveolar septal distention were noted. Fibrous swelling of the cardiac muscle (with striation) just barely maintained) and hyperaemia remained detectable for as long as 14 h post-exposure, but only occurred at 3000 ppm/8 h. A concentration of 1000 ppm/8 h was required to produce a fall in blood albumin, urea nitrogen, alkaline phosphatase activities and erythrocyte count. The cited concentrations failed to produce prenarcotic symptoms of narcosis (central nervous system (CNS) depression). The LD50 was found to be 6.0 ml/kg i.p. and 1.0 ml/kg p.o. for female rats, and 3.2 ml/kg i;p. for female mice. In some of the rats killed in these experiments the organ changes were found to be identical to those observed after inhalation.
