In the longitudinal muscle strip of guinea pig ileum phenoxybenzamine (POB) produces a maximum parallel shift of 0.7 log units in the dose-response curve to histamine. In the presence of sodium thiosulfate in the wash fluid the parallel shift whith retention of maximum response increases to about 2 log units, and a similar value is obtained for Nethyl-N-(2-bromoethyl)-1-naphthylamine. The The agent N-ethyl-N- (2-chloroethyl)benzylamine produces a significantly smaller shift of dose-response curve of 1.53 log units before the maximum response becomes depressed. The receptor-specific depression of maximum response produced by higher doses of POB is reversed by sodium thiosulfate and by bovine serum albumin, while the parallel shift in dose-response curve is unaffected by both treatments. These findings may be explained by a hypothesis involving interaction of 2-haloalkylamines at two sites.

Mice pretreated with Trichinella spiralis extract (TsE), or infected with the parasite, rejected primary skin allografts in 18-23 days and secondary allografts in 12-16 days. Mice pretreated with saline or with bovine serum albumin (BSA) rejected the primary allografts in 12-18 days and did not accept the secondary grafts. Inoculation of increasing doses of parental spleen cells from mice pretreated with saline or with BSA in F1 hybrids produced proportionately stronger graft-versus-host reactions (GvHR) whereas increasing doses of cells from TsE pretreated mice reduced proportionately the capacity of the inoculum to induce a GvHR. Immunodepression of the parental cells was obtained with 7 and with 4, but not with 2, daily injections of TsE. The depression waned rapidly after the treatment with TsE but a significant degree still remained after 3 days. Immunodepression by TsE cannot be solely explained by antigenic competition and although our results are consistent with the induction of suppressor cells, it is probable that other mechanisms are also involved.

Forty-six of sixty-eight rural Nigerian adult patients with sputum-positive pulmonary tuberculosis (P.T.B.) had skin reactions of less than 10 mm. in response to 4 or 5 tuberculin units of purified protein derivative (P.P.D.). The size of the reaction correlated significantly with serum albumin and transferrin concentrations. The diminished reaction in these patients seems to be part of a generalized depression of cell-mediated immunity associated with undernutrition rather than a specific depression of P.P.D. reactivity associated with overwhelming antigen load. It is concluded that the tuberculin test is of no value in the diagnosis of P.T.B. in this community, either in individual cases or epidemiological surveys, unless combined with a biochemical assessment of the person's nutritional status.

Following an intradermal immunization of guinea pigs with dinitrophenylated bovine gamma globulin (DNP23-BGG), human serum albumin (HSA) and trinitrophenylated sheep red blood cells (TNP-SRBC) emulsified in complete or in incomplete Freund's adjuvans the effectivity of 6-mercaptopurine (6-MP) on the formation of specific antibodies was investigated. By application of 10 mg 6-MP/kf/day--in guinea pigs a significant depression of antibody formation against BGG could be demonstrated if treatment is started on the day of antigen injection and continued for seven days. The production of anti-SRBC agglutinating antibodies was only suppressed if the antigen was injected together with complete Freund's adjuvans. The anti-DNP and anti-HSA antibody formation was not influenced by this kind of immunosuppressive treatment.

Mammary tumor virus (MTV) infection has been shown to be associated with a diminished hypersensitive reaction to methylated bovine serum albumin. Since methylated bovine serum albumin-induced hypersensitivity appears to be a mixed [humoral versus cell-mediated immunity (CMI)] reaction, the deficit in reactivity could be caused by, among other things, a direct depression of CMI or an increase in a humoral, blocking component. Assay of oxazolone-induced contact sensitivity and phytohemagglutinin-induced lymphocyte stimulation revealed normal or greater than normal CMI in MTV-positive animals. Treatment of MTV-positive and -negative animals with a regimen of cytosine arabinoside designed to inhibit only humoral immunity and leave CMI intact, corrected the deficit in methylated bovine serum albumin reactivity in MTV-positive mice. Thus, it is suggested that MTV infection may facilitate the production of interfering or blocking humoral immunity.

Chicks which had been inoculated with infectious bursal disease virus (IBDV) at 1 day of age had a severe depression of bursa-dependent humoral immune functions by day 42. Antibody responses against rabbit red blood cells or to immunization with bovine serum albumin were significantly suppressed. In contrast, chicks inoculated with IBDV at 21 days of age produced near normal antibody responses as compared with the responses in noninfected control chicks. The IBDV had no significant effect on the thymus-dependent cellular responses as measured by skin graft rejection or delayed type hypersensitivity reactions to tuberculin.

A radioimmunoassay for cyclic AMP has been developed using protein A containing staphylococci as an immunoabsorbent. Protein A containing heat-killed staphylococci (Cowan I) are coated with rabbit antiserum raised against the 2'-O-succinyl derivative of cyclic AMP coupled to human serum albumin. After washing with a Tween 20 containing buffer, antibody coated staphylococci are diluted with heat-killed staphylococci devoid of protein A (staphylococcus epidermidis) and mixed with [125I]-2'-O-succinyl cyclic AMP tyrosine methyl ester, standards or unknowns. At the end of the incubation, separation of bound and free labelled antigen is achieved by bound and free labelled antigen is achieved by centrifugation. The results are comparable to those obtained with a precipitation assay using polyethylenglycol 6000. Acetylation prior to radioimmunoassay increases sensitivity about 80-fold. 50% depression of zero dose binding occurs at 15--16 femtomoles acetylated cyclic AMP. The crossreactivity with cyclic GMP, ATP, ADP, 5'-AMP and adenosine is extremely low. The present technique is an attractive alternative to the second antibody method or polyethylenglycol precipitation.

This study illustrates the specific immune response of chronically starved, undernourished adults after inoculation of live smallpox vaccine. It produced no adverse effect, and major vaccinial reaction was observed in all. 63% of undernourished individuals showed a fourfold or greater rise of the neutralizing antibody titre. In contrast, only 9% of normal healthy subjects could show similar response. However, the prevaccination titre was much lower in the undernourished group than in the control group, and the postvaccination titre also remained persistently lower in the former than in the latter group. Furthermore, whereas the specific humoral antibody response in the undernourished subjects was partially adequate, the development of specific cellular immunity against vaccinia was remarkably poor, indicating that smallpox vaccination in these subjects might be less effective against variola infection. This observed profound effect of chronic starvation and severe undernutrition on the immune apparatus was possibly multifactorial, protein depletion being the most important factor, as proved by the significantly low serum albumin level. The significantly low peripheral blood lymphocyte count and spectacular unresponsiveness to many antigens in these individuals suggested profound depression of the thymolymphatic system. Further, the significantly low level of neutralizing antibody in the malnourished subjects suggested that the formation of this protective antibody might necessitate the cooperation of T lymphocytes.

Toxicosis was induced in pregnant heifers by feeding 25,000 mg/head/day of FireMaster BP-6, a commercial blend of polybrominated biphenyls (PBB). The PBB feeding decreased dry matter intake approximately 50% by 4 days exposure. Emaciated animals became anorexic a few days prior to death at 33 to 66 days. Weight losses of heifers average 80 kg. Other clinical signs observed were dehydration, diarrhea, excessive salivation and lacrimation, fetal death, abortion, and general depression as evidenced by depressed heart and respiratory rates. Clinical signs were apparent after 10 days exposure and progressively intensified along with loss of condition until death. Clinicopathologic changes included significantly increased serum glutamic-oxaloacetic transaminase and decreased serum calcium by 30 days exposure. Lactate dehydrogenase, urea nitrogen, and bilirubin were elevated, and serum albumin decreased by 36 to 40 days. Principal urine changes were decreased specific gravity and moderate proteinuria. Pregnant heifers fed 0.25 or 250 mg/head/day for 60 days and nonpregnant heifers fed 250 mg/head/day for 180 days displayed neither clinical signs nor clinicopathologic changes indicating adverse effects from PBB exposure. Post-exposure, all heifers exposed to PBB for 60 days calved normally with zero calf mortality and were successfully rebred. Milk production was not different from control animals. Birth weights of calves from dams exposed to 250 mg PBB/head/day were significantly greater than calves of dams exposed to 0 mg or 0.25 mg/head/day. PBB exposure of dams produced no detrimental effects on calves as indicated by clinical signs, clinicopathologic changes, or performance.

Procainamide acetylation and hydrolysis, procainamide-derived p-amino-benzoic acid acetylation, and plasma hydrolysis of procaine were studied in normal volunteers and in 20 patients with chronic liver disease, Impairment of procainamide acetylation was evident in the patients, but no correlations were demonstrable between the degree of impairment and the severity of the disease. On the other hand, procainamide hydroylsis was diminished in liver disease, and as indicated by depression of serum albumin levels and plasma prothrombin activity this alteration did correlate with the degree of impairment of liver function. Procaine hydrolysis in plasma was also affected, the mean in vitro plasma half-life being prolonged in the patients with liver disease and correlating with the degree of hepatic impairment. A correlation of procainamide hydrolysis with procaine hydrolysis was also observed. Finally, acetylation of procainamide-derived p-aminobenzoic acid appeared to increase in patients with liver disease, the degree of acetylation increasing with decreasing procainamide hydrolysis capacity.
