In unilaterally vagotomized rabbits, changes in single vagal nerve fibers from the lung stretch and irritant receptors were studied during the cough reflex following ammonia vapour administered into the lungs. The discharge rates of slowly adapting pulmonary stretch receptors (SARs) before the onset of cough and at the onset of cough did not change significantly. In contrast, the occurrence of cough after ammonia inhalation was usually preceded by vigorous stimulation of the rapidly adapting pulmonary stretch receptors (RARs). These results suggest that the genesis of cough provoked by ammonia is mainly mediated by afferent input from RARs.

The cough reflex in anaesthetized rabbits was elicited by irritation of the larynx with ammonia, inhalation of ammonia into the lungs, and mechanical stimulation of the tracheobronchial tree with a catheter. After block of lung stretch receptors with sulphur dioxide and virtual abolition of the Breuer-Hering inflation reflex, the cough response to all three stimuli was far less frequent and, when it occurred, was weaker especially with respect to the expiratory efforts. It is concluded that the Breuer-Hering reflex enhances the cough reflex, although modification of the sensitivity of tracheobronchial cough and irritant receptors by sulphur dioxide may also influence the cough.

The water, ethanol and ether extracts from Cynanchum glaucescems administrated orally showed significant antitussive effect in ammonia-induced cough model in mice. The water and ethanol extracts had obvious expectorant effect. The filtered solution of water decoction injected intraperitoneally could effectively prevent guinea pigs from asthma induced by acetulcholine and histamine mixture, and also inhibit the ear inflammation in mice caused by croton oil. All these effects showed close dose-effect relationship.

We conducted an assessment of respiratory health status including questionnaire and spirographic measurements in 54 male swine producers age 36.3 +/- 11.1 years (mean +/- SD) who worked an average of 10.7 +/- 6.3 years in the industry and spent 4.7 +/- 2.1 hours per day in the swine barns, and we also measured atmospheric contaminants including carbon dioxide, ammonia, total dust, respirable dust, and airborne endotoxin. Mean atmospheric dust contaminant levels were as follows: carbon dioxide, 2632 +/- 807 ppm; ammonia, 11.3 +/- 4.2 ppm; total dust, 2.93 +/- 0.92 mg/m3; respirable dust, 0.13 +/- 0.05 mg/m3; and endotoxin, 11,332 +/- 13,492 endotoxin units/m3. Of these, endotoxin related to forced vital capacity (P < .05) and endotoxin x hours per day was related to forced vital capacity (P < .05) and to forced expiratory volume in 1 second (P = .06). Respiratory symptoms and lung function studies did not relate to categories of low, medium, and high exposure to respirable dust. However, categories of endotoxin (available on 46 workers) related to respiratory symptoms (cough, P = .02; chronic bronchitis, P = .06; and to forced vital capacity, P < .01). These data suggest that respiratory health status relates to endotoxin levels but not to dust level exposures in the presence of low dust levels and indicates that control measures should include endotoxin as well as dust control.

Bronchopulmonary damage caused by heavy exposure to ammonia may result in chronic obstructive bronchitis, bronchial hyperresponsiveness, in rare cases also in a combined ventilation pattern. Exposure to ammonia for several years, as it occurs in swine confinements in pig-fattening farms is associated with a shift-related decrease of FEV1, FEF25-75 and PEF. Several studies show that ammonia at the TLV (20 ppm) inconstantly causes severe cough. In this connection, patients frequently cannot perform maximum forced ventilation manoeuvres. This leads to reduced spirometric values which usually do not represent bronchial obstruction. This interpretation is supported by normal values obtained by body plethysmography. Nevertheless, corresponding disorders may be associated with significant health impairment. Altogether, the results point predominantly to long-term effects. Obviously, continuous inflammation of the airways favours the development of chronic bronchitis. On this basis, susceptible subjects respond with airway symptoms, especially cough, upon re-exposure.

They could also alleviate the asthma induced by histamine and acetylcholine chloride in guinea pigs.

The objective of this experimental study was to determine the effects of aerial ammonia on disease development and bacterial colonization in weaned pigs inoculated with toxigenic Pasteurella multocida and Mycoplasma hyopneumoniae. Two groups of 10 pigs each were continuously exposed to 50 and 100 p.p.m. ammonia, respectively, and compared to a non-exposed control group of 20 pigs. Following aerosol inoculation with M. hyopneumoniae at day 9, all pigs were aerosol-inoculated with toxigenic P. multocida type A at days 28, 42 and 56. At day 63 they were euthanized. Clinical signs including coughing and respiratory distress were present in all groups following inoculation. No significant differences could be established in the extent or frequency of pneumonia between ammonia-exposed pigs and controls, or in the extent of conchal atrophy, the frequency of isolation of toxigenic P. multocida from conchae, tonsils, lungs and kidneys, or the average daily weight gain. The recovery of toxigenic P. multocida from nasal swabs following inoculation was significantly greater in pigs exposed to 50 p.p.m. ammonia or more as compared to the control group. In conclusion, high levels of ammonia combined with inoculations with M. hyopneumoniae and toxigenic P. multocida had no significant effect on disease development, but may have enhanced colonization by toxigenic P. multocida on the nasal turbinates.

The effects of ammonia on the respiratory system and somatic growth have been measured in piglets taking into account the interactions with endotoxins from gram-bacteria and dust. Dose-response curves were constructed for the inflammatory reactions induced in nasal cavities, the smooth muscle hyperresponsiveness in the trachea, the permeability of alveolo-capillary barrier, the pulmonary vasomotricity, the cough reflex and the systemic effects. An acceptable level of pollution was proposed.

The effect of ammonia on the cough response to citric acid and on substance P release from C-fibers involved in this reflex was assessed. For a period from one to four days, piglets were exposed, in an inhalation chamber, to ammonia at a concentration of 15 or 30 ppm. During exposure, cough induction tests were done every two days. Recovery of the cough reflex after ammonia exposure was also determined. In a separate group of piglets exposed for 2 days to 30 ppm ammonia, substance P content was determined in bronchial and tracheal lavage fluids and in the tracheal and bronchial mucosa. Ammonia (30 ppm) was found to inhibit coughing significantly (the cough frequency was reduced by 64%) after a two-day exposure. In animals exposed for 4 days to this ammonia concentration, the recovery ranged from 3 to 7 days (mean: 5 days). The same ammonia concentration also caused the substance P content to increase significantly in bronchoalveolar lavage fluid (to 432% of its initial value) and tracheal lavage fluid (to 149%) and to decrease significantly in the tracheal mucosa (-58%), however the content in bronchial mucosa was not significantly affected (-43%). Exposure to 15 ppm ammonia had no effect on the frequency of citric acid-induced coughing. In conclusion, ammonia inhibits citric acid-induced coughing in pigs at concentrations that can be detected in piggeries. This inhibitory effect may be related to substance-P depletion in C-fiber endings.

The effect of passive smoking on odor identification was studied in 10 children exposed to passive smoke at home. All had at least one parent who smoked at least one pack of cigarettes a day. The control group consisted of 10 children of nonsmoking parents. Ten odorants were tested: vinegar, ammonia, peppermint, roses, bleach, vanilla, cough drops, turpentine, licorice and mothballs. Each child was presented with five test trays containing all 10 odorants in random order. Of the total of 500 odors presented, the control group correctly identified 396 (79%) and the study group, 356 (71%) (p < 0.005). This work demonstrates that children exposed to passive smoke have difficulty identifying odors in comparison to children raised in relatively smoke-free environments. Since the study group tend to misidentify four of the 10 odorants tested--vanilla, roses, mothballs and cough drops--we suggest that these four odorants should suffice in testing odor identification in children.
