The
cough
reflex elicitability (CRE),
cough
reflex strength (CRS) and Hering-Breuer inflation reflex (HBIR) were studied in 51 anaesthetized (
Pentobarbital
Spofa, 30 mg/kg, i.v.) female rabbits 24-h and 48-h after SO2 breathing. To provoke
cough
, the interior of the trachea and carina were stroked with a polyethylene catheter. To elicit the HBIR, the lungs were inflated to 1.0 kPa intratracheal pressure. Intrapleural and systemic blood pressures were recorded. The CRE, CRS and HBIR obtained 24-h and 48-h after SO2 breathing were compared with correspondent values of control animals. It was found, that CRE and HBIR were fully recovered 24-h after SO2 breathing, but the CRS was still decreased, however, there were no significant differences in CRE, CRS and HBIR between animals 48-h after SO2 breathing and control animals. It can be concluded, that decrease of the CRS 24-h after SO2 breathing is not due to slowly adapting stretch receptors block of airways.

In order to determine the possible involvement of GABA-ergic mechanisms in the modulation of the
cough
reflex, the effects of GABA antagonists on the pentobarbital-induced depression of respiration and
cough
were examined in a comparative study in rats. The
cough
reflex was induced by application of electrical stimulation to the tracheal mucosa by the puncture electrode-induced
cough
method. The 50% antitussive dose (AtD50) of pentobarbital was calculated by the "up and down" method.
Pentobarbital
(10 mg/kg, IP) caused a reduction of tidal volume, which was counteracted by pretreatment with picrotoxin (3 mg/kg, IP) or bicuculline (3 mg/kg, IP). However, neither picrotoxin nor bicuculline were able to counteract the reduction in frequency of respiration. The AtD50 of pentobarbital was 1.95 mg/kg when administered IP. The AtD50 of pentobarbital was not altered after pretreatment of rats with picrotoxin (1.85 mg/kg, IP) or with bicuculline (1.55 mg/kg). These results suggest that GABA-ergic mechanisms may not be involved in the
cough
-depressant effect of pentobarbital.

Too little is known of the mechanisms participating in the regulation of defence reflexes of the organs of respiration. The activity of respiratory tract--in particular lung stretch and
cough
receptors--may play an important role. In the present study the authors investigated the effect of short-term inflation of the lungs, during which lung stretch receptors are mainly stimulated, on the expiration reflex, which is one of the respiratory defence reflexes. The experiments were carried out on 18 rabbits (2.5-2.8 kg) under general anaesthesia (
Pentobarbital
Spofa, 30 mg.kg-1 i.v.). The VT, V', Ppl and systemic BP were recorded. The expiration reflex was elicited by stimulating the vocal folds with a silon fibre loop during resting respiration and during inflation of the lungs with pressures of 0.49, 0.98 and 1.47 kPa. The criterion of the intensity of the expiration reflex was the Ppl expressed in kPa. The authors found that the intensity of the expiration reflex during resting respiration was 0.29 +/- 0.02 kPa (S.E.M.). An inflation pressure of 0.49 kPa raised it to 0.69 +/- 0.06 kPa, a pressure of 0.98 kPa raised it still further to 1.12 +/- 0.06 kPa and a pressure of 1.47 kPa raised it yet again to 1.59 +/- 0.08 kPa. In the authors' opinion, the increase in the intensity of the reflex is due to augmented activity of lung stretch receptors.
