Although water vapor is one of the most important and certainly the most variable minor constituent of the atmosphere, accurate measurements of p(H20) with high time resolution are difficult, particularly in the cold upper troposphere/lower stratosphere. This work demonstrates that a diode laser-based photoacoustic (PA) water vapor detector is a viable alternative to current water vapor sensors for airborne measurements. The PA system was compared with a high-quality frost point hygrometer (FPH) and with a Lyman-alpha hygrometer in the pressure range of 1000-100 hPa at frost point temperatures between 202 and 216 K. These conditions were simulated in a large environmental chamberfor 14 h. Simultaneous measurements with the three instruments agreed within 6%. Nitric acid vapor interferes with the FPH measurements at low frost point temperatures but does not affect the other instruments. The sensitivity of the PA system is already sufficient for measurements in the upper troposphere, and straightforward improvements can extend its useful range above the tropopause. Rugged construction, extreme simplicity, small size, and potential for long-term automatic operation make the PA system potentially suitable for airborne measurements.

This study was conducted to evaluate the efficacy of desensitizing agents for the treatment of dentin hypersensitivity. Seventy-five hypersensitive teeth in 10 patients were studied. These were divided into five groups of 15 hypersensitive teeth: group I, control (triple distilled water); group II, 10% strontium chloride; group III, 5% potassium nitrate; group IV, 2% sodium fluoride; and group V, 40% formalin. The stimulus used to test thermal sensitivity was cold water at 15 degrees C, 10 degrees C, 5 degrees C, and 0 degrees C applied on every alternate day for 10 days. Results showed that 10% strontium chloride solution, 2% sodium fluoride solution, and 40% formalin solution significantly reduced dentin hypersensitivity, whereas a 5% solution of potassium nitrate did not.

Nitric oxide (NO) has high affinity to heme and by interaction with oxyhemoglobin (HbO2) is converted into nitrate to form methemoglobin (MetHb) as a side product. In combining with deoxy-Hb NO yields a stable molecule of nitrosyl-hemoglobin (HbFe(II)NO) that can further be converted into nitrate and hemoglobin (Hb). In addition, Hb was shown to transport NO in a form of S-nitrosohemoglobin (SNO-Hb). These features of the Hb and NO interaction are important for blood oxygen transport including hemoglobin-oxygen affinity (HOA). The present investigation was aimed to study the blood oxygen transport indices (pO2, pCO2, pH, HOA, etc.) in rats under hypothermia combined with a modification of L-arginine-NO pathway. To modify the L-arginine-NO pathway, rats were administered with N(G)-nitro-L-arginine methyl ester (L-NAME), L-arginine, or sodium nitroprusside (SNP) intravenously before cooling. A substantial impairment of oxygen delivery and development of hypoxia, with an important contribution of HOA into the latter accompanied the deep hypothermia in rats. All the experimental groups developed metabolic acidosis, less pronounced in rats treated with L-arginine only. In the experiments with a modification of the L-arginine-NO pathway, an enhanced cold resistance, attenuated oxygen deficiency, and a weaker oxyhemoglobin dissociation curve (ODC) shift leftwards were observed only after the administration of L-arginine. Neither SNP nor L-NAME had not any protective effects. L-Arginine lowered the value of standard P50 (pO2, corresponding to 50% Hb saturation with oxygen at 37 degrees C, pH 7.4, and pCO2 = 40 mmHg). The actual P50 (at actual pH, pCO2 and temperature) decreased by approximately 15 mmHg and was significantly higher than that under hypothermia without the drug treatment (21.03 +/- 0.35 vs 17.45 +/- 0.60 mmHg). NO also can contribute to this system through different mechanisms (HOA modification, vascular tone regulation, peroxynitrite formation, and effects).

The dangerous for health nitrate and nitrite can penetrate with food human organism for this reason in study the influence of thermal processing and storage on the level of these compounds in the vegetables was determined. The content of nitrates and nitrites was determined in such vegetables as carrot, parsley-root, celery and potatoes growing by farmers in the Podlasie province. Nitrates and nitrites were assessed in fresh and boiled vegetables as well as in the stock and in carrot juice. These compounds were also determined after storage of vegetables at +4 degrees C for 2 weeks and at -15 degrees C for 1 and 3 months. Nitrates and nitrites concentrations were assayed colorimetrically by the Griess method modified to food investigation. Boiling reduced nitrate content in the vegetables. Considerable part (about 50%) of these compounds passed into stock during boiling. Storage of these vegetables at +4 degrees C resulted in slight lowering of nitrate content in carrot and parsley-root and marked elevation in celery. Cold storage of vegetables through 1 and 3 months had no important influence on nitrate content. Boiling decreased the content of nitrites similarly as nitrates. Storage of the vegetables at +4 degrees C through 2 weeks led to an increase in nitrite content in carrot and parsley-root while in celery the content of nitrites was reduced. Storage of vegetables at -15 degrees C resulted in lowering content of nitrite. The study revealed that the vegetable-roots (carrot, parsley and celery) and potatoes cultivated in the Podlasie province as well as in the other provinces were excessively contaminated by nitrates and nitrites. Moreover, it has been shown that thermal processing such as boiling considerably reduced the content of nitrates and nitrites in these vegetables while freezing changed mainly nitrite content.

Meteorological conditions during the period of plant vegetation make the greatest contribution to the formation of the plant nitrate pool. Nitrate nitrogen is accumulated at high concentrations during sunless, cold years. The weather conditions of the last five to ten days of plant vegetation determine nitrate accumulation. The meteorological conditions during the entire period of vegetation determine the increase in the content of nitrates due to nitrogen fertilizers. The effect of meteorological conditions is especially pronounced in low fertility soils and upon introduction of high doses of nitrogen fertilizers. The reaction of plants to unfavorable meteorological conditions greatly varies with reference to their biological and genotypical specificity.

The functional potential of bacteria isolated from the rhizosphere of barley (Hordeum vulgare L.) in May, July, and August and cultivated on nutrient-rich substrate (1/10 TSBA) and nutrient-poor substrate (cold soil extract agar) was determined. There was no significant difference in numbers of CFU when counted on nutrient rich or poor substrate. Bacterial numbers increased approximately 3-fold in the rhizosphere soil from May to August but was unchanged in bulk soil over the same period. A total of 4474 randomly isolated bacteria were screened for enzymatic activities involved in carbon turnover (amylase, cellulase, mannanase, xylanase, and chitinase), nitrogen turnover (protease, nitrate and nitrite reductase), and phosphate turnover (phosphatase). In the rhizosphere soil, bacteria carrying C and P turnover enzymes were not stimulated by the growing plant whereas protease and nitrate and nitrite reductase were stimulated by the growing plant. No changes were observed in the bulk soil. Two taxonomic groups were followed: Cytophaga-like bacteria (CLB) and fluorescent pseudomonads, the latter being abundant in the rhizosphere and important contributors to the cycling of organic matter in soil. Unexpectedly in the spring samples, CLB were around 25% of all bacteria isolated, whereas fluorescent pseudomonads made up less than 10%. The relative proportion of these bacterial groups then decreased during the plant growth season but at all times showing a clear rhizosphere effect. Furthermore, up to 70% of the isolates carrying enzymes involved in the turnover of carbon, in the May sample, were identified as CLB, indicating the importance of this group in early colonization of the rhizosphere. The fluorescent pseudomonad group contributed less than 3%.

Nitrogen is an essential nutrient in the biogeochemistry of forested ecosystems. The influence of canopy lichens on the winter biogeochemistry of nitrate in broadleaved deciduous forests is examined and it is hypothesized that nitrate sequestration will not differ between winter precipitation events. Rejection of this hypothesis would mean that meteorological conditions of winter precipitation events have a detectable influence on nitrate sequestration by canopy lichens and nitrate input to the forest floor. Canopy lichens of the genus Parmelia were found to influence winter nitrate stemflow inputs to forest soils differentially. Epiphytic lichens on an individual Carya glabra Mill. (pignut hickory) canopy tree, centrally located within the stand of an open deciduous forest, actively sequestered nitrate leached from the tree's woody frame, lowering aqueous stemflow inputs at the tree base. The quantities of nitrate sequestered by corticolous lichens during the 2 February 1999 mixed-precipitation event were significantly greater than those during all other precipitation events examined. Greater rates of nitrate uptake during the 2 February 1999 event may be attributed to (1) its intermediate rain intensity, which would have soaked the lichen thalli in a nutrient-rich bath, and (2) an air temperature range between -2 degrees C and 8 degrees C that would have increased viscosity and surface tension of stemflow drainage, thereby decreasing stemflow velocity and increasing the contact time of stemflow water on the lichen thalli. Other precipitation events were either too cold to promote metabolic activity by canopy lichens or too warm and intense for an optimal contact time of stemflow with lichen thalli, resulting in lower quantities of nitrate sequestered. Meteorological conditions of winter precipitation events have been documented to influence sequestration of nitrate by corticolous lichens and decrease aqueous stemflow inputs to the forest floor of broadleaved deciduous forests.

Levels and patterns of C1-C4/C9 organic nitrates were measured for the first time in Antarctica. The sampling was done by adsorptive enrichment on Tenax TA followed by thermodesorption cold-trap high resolution capillary gas chromatography with electron capture detection. 2-70 1 air on-column have been analyzed this way. C1-C9 alkyl mononitrates, C2-C4 alkyl dinitrates, C2-C4 hydroxy alkyl nitrates, and halocarbons could be identified in air samples collected near the German Neumayer Research Station, Antarctica, in February 1999. Volatile biogenic and anthropogenic halocarbons were used to assess the origin of the air parcels analyzed. The average concentration measured for sigmaC2-C6 alkyl nitrates was in the range of 9.2 +/- 1.8 ppt(v), while the sum of the mixing ratios of six C2-C4 hydroxy alkyl nitrates was in the range of 1.1 +/- 0.2 ppt(v). Moreover, C2-C4 alkyl dinitrates were found at levels near the detection limit of 0.1-0.5 ppt(v). The concentrations of organic nitrates found in Antarctic air represent ultimate baseline levels due to chemical and physical loss processes during long-range transport in the air. The South Atlantic and the Antarctic Ocean as a general secondary source for organic nitrates in terms of an air/sea exchange equilibrium has to be evaluated yet, but it seems logical. Our results confirm the common assumption that there are no biogenic marine sources of C2-C9 organonitrates. We have found a level of > 80 ppt(v) for methyl nitrate. This level if it can be confirmed in a systematic survey requires a strong biogenic source of methyl nitrate in the Antarctic Ocean.

The exogenous application of hormones has led to their implication in a number of processes within the plant. However, proof of their function in vivo depends on quantitative data demonstrating that the exogenous concentration used to elicit a response leads to tissue hormone levels within the physiological range. Such proof is often lacking in many investigations. We are using abscisic acid (ABA)-induced turion formation in Spirodela polyrrhiza L. to investigate the mechanism by which a hormone can trigger a morphogenic switch. In this paper, we demonstrate that the exogenous concentration of ABA used to induce turions leads to tissue concentrations of ABA within the physiological range, as quantified by both enzyme-linked immunosorbent assay and high-performance liquid chromatography/gas chromatography-electron capture detection analysis. These results are consistent with ABA having a physiological role in turion formation, and they provide an estimate of the changes in endogenous ABA concentration required if environmental effectors of turion formation (e.g. nitrate deficiency, cold) act via an increased level of ABA. In addition, we show that the (+)- and (-)-enantiomers of ABA are equally effective in inducing turions. Moreover, comparison of the ABA; levels attained after treatment with (+)-, (-)-, and ([plus or minus])-ABA and their effect on turion induction and comparison of the effectiveness of ABA on turion induction under different pH regimes suggest that ABA most likely interacts with a plasmalemma-located receptor system to induce turion formation.

A facultatively psychrophilic bacterium, strain 4-22T, was isolated from a cold current off the Monbetsu coast of the Okhotsk Sea in Hokkaido, Japan. The isolate was a rod-shaped facultative anaerobe that reduced nitrate to nitrite and hydrolysed starch, DNA and alginic acid, but not chitin or gelatin. The isolate grew at 0 degrees C, but not at 26 degrees C; the optimum growth temperature was 14-16 degrees C. NaCl was required for growth. The DNA G+C content was 43.5 mol%. The whole-cell fatty acids consisted of significant amounts of an unsaturated fatty acid, C16:1, and a saturated fatty acid, C16:0. A polyunsaturated fatty acid, docosahexaenoic acid (C22:6), was also detected (1.6%). Phylogenetic analysis based on 16S rRNA gene sequences showed that strain 4-22T was closely related to Psychromonas antarctica (95.7% similarity). DNA-DNA hybridization revealed a relatedness of 31% between strain 4-22T and P. antarctica. Based on physiological and biochemical characteristics and the phylogenetic position as determined by 16S rRNA gene analysis and DNA-DNA relatedness, it is concluded that the isolate represents a novel species, for which the name Psychromonas marina sp. nov. is proposed. The type strain is 4-22T (= JCM 10501T = IAM 14899T = NCIMB 13792T).
