We have transformed two new MC-induced tumours, a
sarcoma
(MCB31-SC), into ascites form. When transplanted s.c. these ascites tumours grow as solid, quite undifferentiated tumours, (AS = ascites solid tumours). We compared the metastasizibility of the AS tumours with that of the parent tumours. In doing so, we used both the tail and the hind leg as transplantation sites. The tumours can be radically removed from both sites by amputation, which prolongs the survival time of the animals and permits metastases to grow into detectable sizes. As registered grossly and by microscopy, the AS tumours have a greater tendency of spread than the parent (SS/SC) tumours. MCB-21 AS grows quicker than 21-SS and gives rise to more lymph node metastases. When transplanted to the tail the AS tumour also gives a higher incidence of lung metastases. We detected no such difference by leg-transplanted tumours. MCB31-SC did not produce any detectable metastases at all, while 31-AS, particularly from the tail, gave rise to numerous lymph node and lung metastases. There were no differences in tumour size or growth rate to account for this difference. Thus ascites conversion has changed the
carcinoma
MCB31-SC into an undifferentiated, metastasizing tumour, as detected by our procedure. The design of test protocols to detect metastasizibility is discussed.

Nineteen patients resistant to an adriamycin-containing regimen were treated with 4 days of amphotericin B and the same adriamycin-containing chemotherapy regimen that had been given without amphotericin B. Of the 14 evaluable patients (11 with breast
carcinoma
and three with
sarcoma
), one patient with breast
carcinoma
achieved a partial remission and a second patient with breast
carcinoma
remained stable. All other patients developed progressive disease. Both hematologic and nonhematologic toxicity were significantly increased when amphotericin B was added to chemotherapy.

A water-soluble nitrosourea, 3-[(4-amino-2-methyl-5-pyrimidinyl)methyl]-1-(2-chloroethyl)-1-nitrosourea hydrochloride (ACNU, NSC-245382), was tested for its antitumor activity against some kinds of transplantable mouse tumors. The compound was markedly active against myeloid leukemia C1498, plasmacytoma X5563, Ehrlich ascites
carcinoma
, and mammary tumor FM3A43, and moderately active against mammary tumor MM102 and meningeal
sarcoma
MS147. It appears that ACNU has a broad antitumor spectrum.

Carcinomas
of the extremities are relatively rare neoplasms when compared to
Sarcomas
. After a review of the literature about carcinomas in chronic fistulae and other bone lesions, two cases are reported in which a
carcinoma
originated in scar-tissue, 31 years after a frost-bite lesion of the foot. The problems of etiology, pathogenesis and therapy are pointed out.

and normal cells are surrounded in vitro by thick, transparent zones (approximately equal to 9 micron thick) that spleen cells and a variety of other cells and particles cannot penetrate. Seven lymphoblastoid cell lines did not possess such halos. The presence of these halos around adherent fibrosarcoma cells appeared to protect them from lymphocyte-mediated cytolysis. Hyaluronidase treatment, which destroyed the halo and allowed lymphocytes to approach the tumor cell membrane, enhanced the cytotoxic action of immune but not of normal spleen cells. These observations, in addition to highlighting a little-known feature of the cell surface, may also be of general relevance to the in vitro and in vivo killing of tumor cells by immune effector cells.

A case of spindle-cell
carcinoma
(pleomorphic
carcinoma
), a rare polypoid tumor of the tongue, is reported. The characteristic clinical, gross, and microscopic features of this peculiar lesion and its common sites of location are presented. Origin and pathogenesis of the
sarcoma
-like elements of the lesion are discussed and the literature on the subject is reviewed. Sites of apparent transition between the spindle cell elements of the tumor and the overlying epithelium were observed in light microscopic sections, and the electron micrographs of the spindle-cell portion of the lesion demonstrated the presence of tonofilaments and desmosomes in many tumor cells. These findings support the concept that this group of lesions are pleomorphic variants of squamous-cell
carcinoma
with predominantly spindle-cell pattern. Current evidence in the literature also suggests that metaplastic transformation of the tumor cells into mesenchymal elements may take place in some of these lesions, and that "metaplastic carcinoma" may probably be a more proper designation for them.

Twenty-six brain-metastases in black patients, appearing for most of them as primitive, were observed during a period of 11 years in the Neurological Clinic of the Fann U.H.C. in Dakar. They represent 13.9% of brain-tumours and 11.81% of intracranial space occupying lesions. Their etiology is very diversified: lung cancer (4, 15.4%); breast (0); liver (3, 11.5%); digestive tract (3, 11.5%); prostate gland (1); ovaries (1); parotid gland (1); thyroid gland (2); lympho-reticulosarcoma (2); melano-
sarcoma
(1); chorio
carcinoma
(3); undetermined (5, 11.5%). This series enables us to appreciate differences with African statistics, which are not numerous and with other foreign statistics. The difference of division of cancer in Africa, particularly in Senegal, explains some results such as the relative high frequency of liver-cancer in the etiology of this metastasis. But it is surprising to learn that lung-cancer is frequent and breast-cancer absent. Also to be marked is the non-negligible influence of the choriocarcinoma metastasis. Progress in approaching diagnosis, by developing scintigraphy, might facilitate extraction of solitary metastasis more often.

To check the suggestion that 3-methylcholanthrene-induced,
sarcoma
-associated, tumor-specific transplantation antigens (TSTA) could be modified H-2 antigens, reciprocal isoantigenic variants derived from TA3Ha/MSWBS hybrid cells were examined. This hybrid was produced by the fusion of the TA3Ha ascites
carcinoma
(H-2a) and the 3-methylcholanthrene-induced MSWBS ascites
sarcoma
(H-2s). MSWBS expresses a strong TSTA capable of inducing a rejection reaction in the syngeneic A.SW host. The genetic determinants of the H-2 complex are known to be localized on chromosome No. 17. TA3Ha contributes two normal, telocentric chromosomes No. 17 to the hybrid. In contrast, both chromosomes No. 17 of MSWBS are localized on readily identifiable translocations (17/1 and 17/M1). We have previously shown that the chromosomes No. 17 of one parental strain or the other (but not both) can be eliminated from the hybrid upon selective passage in the opposite parental strain. The present studies showed that the two strain A-compatible variants that have lost the
sarcoma
-derived 17-chromosomes still contained the same TSTA as the two reciprocal strain A.SW-compatible variants that have lost the mammary
carcinoma
-derived H-2 chromosomes. These findings argue against the possibility that methylcholanthrene-induced TSTA is a modified form of H-2 or that its structural determinants(s) is localized on chromosome 17.

Chest x-rays, whole lung tomograms, and operative findings were correlated in 152 patients with an extrathoracic malignancy who underwent 182 thoracotomies for the evaluation of pulmonary nodules. Among 25 patients with a normal chest x-ray but one or more pulmonary nodules demonstrated by whole lung tomography, 19 had metastatic disease documented at thoracotomy. Of 64 patients with unilateral nodules seen on conventional chest x-ray, 10 of 32 with
sarcoma
, 2 of 7 with melanoma, but only 2 of 25 with
carcinoma
had bilateral nodules demonstrated by tomography. At thoracotomy, 8 of 38 patients in whom a solitary nodule was present on both chest x-ray and on tomogram had multiple nodules. Similarly, 35 of 48 patients with more nodules seen on the tomograms than on the chest x-ray had even more extensive metastatic disease demonstrated by operation. Among patients who had resection of all metastatic pulmonary nodules, 50% of those with
carcinoma
and 32% of those with
sarcoma
who underwent resection of a solitary nodule approached that of patients with
carcinoma
. These findings suggest that whole lung tomograms contribute to the determination of the extent of pulmonary metastasis in patients with solid malignancies, even in the presence of a negative chest x-ray. The improved long term survival achieved by resection of these metastases may be partially attributed to this screening procedure.

The reactive properties of tumors of epithelial origin (
carcinoma
RS-1) and connective tissue origin (Jensen
sarcoma
) are not analogous. Vitamin A inhibits the growth of
carcinoma
RS-1 and does not change the growth parameters of Jensen
sarcoma
. Hydrocortisone and proserine increase the growth rate of
carcinoma
RS-1. Hydrocortisone was found to inhibit the growth of Jensen
sarcoma
