Given the recent findings regarding the association between alexithymia and Autism Spectrum Disorder (ASD) and the accumulating evidence for the presence of the Broader Autism Phenotype (BAP) in relatives of individuals with ASD, we further explored the construct of alexithymia in parents of children with ASD as a potential part of the BAP. We hypothesized that (a) parents of children with ASD will demonstrate higher impairment in their emotion processing when compared to controls, and (b) high impairment in emotion processing in parents will be associated with severity of symptoms in children with ASD. Psychometric and diagnostic data were collected on 188 children with a diagnosis of ASD. The Toronto Alexithymia Scale (TAS-20) was completed by 439 parents of children with ASD and a control group of 45 parents of children with Prader Willi syndrome (PW). Results show that ASD parents score higher than controls on the TAS-20 total score. Within the ASD group, children of fathers with high alexithymia score higher on repetitive behaviour symptoms compared to children of fathers with low alexithymia. The alexithymia trait appears to be one of the many building blocks that make up the BAP.

Mothers of children aged 2-12 years completed an exhaustive questionnaire assessing feeding and eating behaviors for both themselves and their children with autism, and typically developing siblings of children with Autism Spectrum Disorder (where available), or typically developing children with no sibling with a disability. Results indicate that children with autism were only marginally more likely to exhibit picky eating behavior (overall style) than their siblings or matched typically developing children. Rates of ritualistic feeding behaviors were equivalent in all groups of children although children with autism were more likely to be currently exhibiting problematic eating and feeding behaviors. The implications of these results for the treatment of feeding difficulties exhibited by children with autism will be discussed.

Studies of amniotic testosterone in humans suggest that fetal testosterone (fT) is related to specific (but not all) sexually dimorphic aspects of cognition and behaviour. It has also been suggested that autism may be an extreme manifestation of some male-typical traits, both in terms of cognition and neuroanatomy. In this paper, we examine the possibility of a link between autistic traits and fT levels measured in amniotic fluid during routine amniocentesis. Two instruments measuring number of autistic traits (the Childhood Autism Spectrum Test (CAST) and the Child Autism Spectrum Quotient (AQ-Child)) were completed by these women about their children (N=235), ages 6-10 years. Intelligence Quotient (IQ) was measured in a subset of these children (N=74). fT levels were positively associated with higher scores on the CAST and AQ-Child. This relationship was seen within sex as well as when the sexes were combined, suggesting this is an effect of fT rather than of sex per se. No relationships were found between overall IQ and the predictor variables, or between IQ and CAST or AQ-Child. These findings are consistent with the hypothesis that prenatal androgen exposure is related to children exhibiting more autistic traits. These results need to be followed up in a much larger sample to test if clinical cases of ASC have elevated fT.

There is a lack of instruments validated for screening of autism spectrum disorders (ASD) in general populations and primary care settings. The Autism Spectrum Screening Questionnaire (ASSQ) has previously been shown to have good screening properties in clinical settings. We used the ASSQ to screen a total population of 7-9 year-olds (N = 9430) for ASD in the Bergen Child Study. Parents and teachers filled in the ASSQ, and high-scorers were invited for clinical assessment, along with a large group of screen negative children. We found that the ASSQ was well suited as a general population screen. Combining parent and teacher ASSQ and using cut-off score of > or =17 provided the most efficient screen with sensitivity of 0.91 and specificity of 0.86.

Of particular interest to studying the etiology of Autistic Spectrum Disorders (ASDs) is the potential for multiple risk factors to combine through non-random mechanisms-assortative mating. Both genetic influences and a high-testosterone prenatal environment have been implicated in the etiology of ASDs, and given that waist-hip ratio (WHR) is indicative of a woman's circulating testosterone level, a man attracted to higher-than-average WHR women is likely to have a higher-than-average prenatal testosterone exposure for their offspring. We show that whereas fathers of children without ASD show a statistically reliable preference for WHRs at the low end of the normal range, indicative of women with low testosterone levels, fathers of children diagnosed with ASD do not consistently show this preference.

Recent interest in gesture has led to an understanding of the development of gesture and speech in typically developing young children. Research suggests that initially gesture and speech form two independent systems which combine together temporally and semantically before children enter the two-word period of language development. However, little is known about gesture development in children's disordered speech. This paper presents two case studies of young children with autism. The children are under 3 years of age and attend an intervention programme to facilitate their social and communication development. Early indications suggest that whilst both gesture and speech development is delayed in children with Autistic Spectrum Disorders (ASD), the developmental trajectory is the same as for typically developing children.

Pragmatic language skill is regarded as an area of universal deficit in Autism Spectrum Disorder (ASD), but little is known about factors related to its development and how it in turn might contribute to skills needed to function in everyday contexts or to the expression of ASD-related symptoms. This study investigated these relationships in 37 high-functioning children with ASD. Multiple regression analyses revealed that structural language skills significantly predicted pragmatic language performance, but also that a significant portion of variance in pragmatic scores could not be accounted for by structural language or nonverbal cognition. Pragmatic language scores, in turn, accounted for significant variance in ADOS Communication and Socialization performance, but did not uniquely predict level of communicative or social adaptive functioning on the Vineland. These findings support the notion of pragmatic language impairment as integral to ASD but also highlight the need to measure pragmatic skills in everyday situations, to target adaptive skills in intervention and to intervene in functional, community-based contexts.

Homeobox protein engrailed-2 (EN-2), is a subtype of the homeoprotein transcription factors of EN family. It can be transported between cells in culture by unconventional secretion and internalization mechanisms. When internalized, it can activate the transcription or translation. EN-2 had been considered to be an important transcriptional factor during the embryonic development , it involved in the regulation of anteroposterior polarity. In recent years, much more functions of EN-2 are revealed. It controls synaptic choice as well as axon projections; and there are some relationships between EN-2 and Parkinson's disease. Furthermore, EN-2 is thought to be a candidate oncogene in human breast cancer and a candidate gene of Autism Spectrum Disorder. The structure, distribution, function and regulation of EN-2 as well as the relationships between EN-2 and some diseases are reviewed in the present article.

Although communication and social difficulties in autism have received a great deal of research attention, the other key diagnostic feature, extreme repetitive behaviour and unusual narrow interests, has been addressed less often. Also known as 'resistance to change' this may be related to atypical processing of infrequent, novel stimuli. This can be tested at sensory and neural levels. Our aims were to (i) examine auditory novelty detection and its neural basis in children with autism spectrum conditions (ASC) and (ii) test for brain activation patterns that correlate quantitatively with number of autistic traits as a test of the dimensional nature of ASC. The present study employed event-related fMRI during a novel auditory detection paradigm. Participants were twelve 10- to 15-year-old children with ASC and a group of 12 age-, IQ- and sex-matched typical controls. The ASC group responded faster to novel target stimuli. Group differences in brain activity mainly involved the right prefrontal-premotor and the left inferior parietal regions, which were more activated in the ASC group than in controls. In both groups, activation of prefrontal regions during target detection was positively correlated with Autism Spectrum Quotient scores measuring the number of autistic traits. These findings suggest that target detection in autism is associated not only with superior behavioural performance (shorter reaction time) but also with activation of a more widespread network of brain regions. This pattern also shows quantitative variation with number of autistic traits, in a continuum that extends to the normal population. This finding may shed light on the neurophysiological process underlying narrow interests and what clinically is called 'need for sameness'.

An increasing body of literature has indicated that social stories are an effective way to teach individuals diagnosed with autism appropriate social behavior. This study compared two formats of a social story targeting the improvement of social skills during game play using a pretest posttest repeated measures randomized control group design. A total of 45 children diagnosed with Autism Spectrum Disorder (ASD) ages 7-14 were randomly assigned to standard, directive, or control story conditions. Results demonstrated that the standard and directive story formats were equally as effective in eliciting, generalizing and maintaining the targeted social skills in participants who had prior game play experience and Verbal Comprehension Index (VCI) scores from the WISC-IV intelligence test in the borderline range or above.
