Malignant Balb/c mouse lung cell clones related to alveologenic carcinoma exhibited low levels of epidermal growth factor (EGF) receptor activity compared to nonmalignant cell clones. Immunoprecipitation of cell homogenates and immunohistochemistry on urethane-induced lung tumors suggest that the absence of activity reflects decreased amounts of EGF receptor protein. Low levels of EGF receptor alone cannot cause neoplastic transformation, since a nonneoplastic cell cone, B5D3, exhibited low levels of EGF receptor despite its nontransformed phenotype. The reduced levels of EGF receptor in malignant clones have been mimicked by long-term (12 h) treatment of a nontransformed cell clone with 200 nM phorbol dibutyrate. The detection of mutated ras oncogene in the transformed cell lines, taken together with the EGF receptor findings, suggests that more than one alteration in the signal transduction pathway may be necessary for transformation in alveologenic
adenoma
and carcinoma cell systems. A further phenotypic feature of transformation, reduced expression of the extracellular matrix proteins
fibronectin
and laminin, may be mediated at the transcriptional level.

Thirteen cases of pleomorphic
adenoma
were studied by both immunohistochemical and other histochemical methods. The exocrine cells and myoepithelial cells appear to produce similar cell products as their normal salivary gland counterparts. Keratin was found in both exocrine cells and myoepithelial cells. CEA, secretory component, and lactoferrin were detected only in the tumor exocrine cells with adenoid differentiation. S-100 protein, ferritin,
fibronectin
, laminin and elastin were detected only in the myoepithelial cells. The residual sugars glucosyl, mannosyl, galactosyl and fucosyl were identified in both cell types, in variably detectable amounts.

10 pleomorphic adenomas of the human parotid gland were transplanted on several groups of nude mice. For comparative reasons, 10 other pleomorphic adenomas, a neurinoma and a chordoma and transplants of squamous cell carcinomas and of normal salivary gland tissue were also analysed. In the primary tumours and in the transplants, the presence of keratin, carcinoembryonic antigen, tissue polypeptide antigen, lactoferrin, lysozyme, immunoglobulins, secretory component, amylase,

and of several lectin-receptors (PNA, WGA, HPA, Ulex europaeus) was sought. The immunohistological observations show that many of the features of a pleomorphic
adenoma
are constant under the conditions of transplantation. In the transplanted tumour, the same heterogeneity as in the primary tumours can be observed. Autoradiographic studies show little labelling with 3-H thymidine, which is in good accordance with the biological behaviour of the tumour. The distribution of

shows an interesting association with myoepithelial-like cells. Our results support the hypothesis that the histogenetic origin of the pleomorphic

and 19 cases of adenoid-cystic carcinoma of the salivary glands, and one case in the mammary location, were investigated regarding immunohistochemical reactivity for Tissue Polypeptide Antigen (TPA), Pre-Keratins, Vimentin, S-100 Protein, and their arrangement pattern of
fibronectin
. As a whole, the results support the hypothesis of morpho-structural and mainly, onto-histogenetic similarities between these tumours, but they also underline the need for great care in outlining their morpho-functional features, in relation to their different prognoses.

Malignant cell lines related to mouse lung alveologenic carcinoma have been established from urethane-induced tumors and after in vitro spontaneous transformation of preneoplastic cell lines. Both the chemically and spontaneously transformed cell lines formed invasive, poorly differentiated carcinomas with secondary lung deposits when implanted subcutaneously in immune-suppressed mice. They differed from the related preneoplastic cell line in coordinately exhibiting anchorage-independent growth, reduced epidermal growth factor receptor activity and absence of pericellular
fibronectin
. These data suggest that similar molecular events may occur in type 2 pneumocyte-related cells in order to generate mouse lung alveologenic adenomas and carcinomas by both spontaneous and chemical carcinogen induction mechanisms. A reduced level of pericellular
fibronectin
was also demonstrated in an in situ compressive urethane-induced mouse lung
adenoma
. Loss of pericellular
fibronectin
may therefore be an early and persistent phenotypic alteration during transformation to the alveologenic

To elucidate the stromal characteristics in gastric carcinoma, the localization of chondroitin sulfate proteoglycans (PGs),
fibronectin
(FN), and type IV collagen was observed by immunohistochemical methods in 46 surgically resected stomachs including 41 carcinomas, 2 adenomas, and 3 ulcers. Chondroitin 4-sulfate PG revealed by 9A2 antibody and FN were distributed abundantly in the stroma of gastric carcinoma. On the other hand chondroitin 6-sulfate PG revealed by 3B3 antibody and type IV collagen were located more broadly in the stroma of undifferentiated carcinoma, especially scirrhous carcinoma than differentiated carcinoma. Type IV collagen was ultrastructurally localized around smooth muscle cells, myofibroblasts, and capillaries. And Bromodeoxyuridine was labelled in the stromal cells including endothelial cells and smooth muscle cells whose proliferative activities were suggested. In addition to carcinoma, muscularis mucosae of
adenoma
and ulcer were stained by 3B3 antibody. In gastric carcinoma, extracellular matrix was more strikingly distributed compared with noncarcinomatous region and in particular, stromal changes may be promoted by cancer cells in scirrhous carcinoma. These results suggest that specific stromal changes in gastric carcinoma may be related with the histologic types and growth patterns.

We tested fluorescent and light microscopic markers to improve recognition of pituitary adenomas at biopsy. The optimal reagent was 100 mg/L of fluoresceinated Ricinus communis agglutinin 120 (RCA 120) lectin plus 3 mg/L of propidium iodide. The refrigerated solution was immediately available for use on routine frozen sections. The sections were stained for one minute and viewed immediately after they were rinsed with saline and coverslips applied. Fluorescein-labeled RCA 120, which binds galactose, localized vascular stroma. Propidium iodide, which binds nucleic acids, stained nuclei. Stromal configuration, nuclear morphology, and cell to stroma ratio were illuminated and used to distinguish
adenoma
from adenohypophysis. We also describe a method utilizing peroxidase-conjugated RCA 120 that demonstrates the same features by light microscopy. Fluoresceinated RCA 120-stained vessels and stroma of routinely processed material more reliably than hematoxylin-eosin or labeled antibody to

Histochemical studies of
fibronectin
in cancer and
adenoma
of the large intestine were conducted by the immunoperoxidase technique. The
fibronectin
immunohistochemical reaction was present in frozen sections, whereas it was obviously suppressed in the formalin-fixed sections. In the normal mucosa of the large intestine, the reaction was strong in the basement membrane of epithelial cells and weaker around the interstitial cells of the lamina propria mucosa. The reaction in cancer tissues was negative in the basal part of the gland, while the interstitial connective tissue was strongly positive for
fibronectin

, there was a correlation between the intensity of the
fibronectin
reaction and the degree of epithelial cell atypism; that is, in a gland with severe atypism the reaction was very weak or absent, while in a gland with mild atypism a weak reaction still persisted. reaction has a close relationship with malignancy and with cellular atypism of the epithelium of the large intestine.

Alternative splicing of
fibronectin
pre-mRNA has been shown to be independently regulated at the EDA and EDB regions in a tissue and developmental stage-specific manner. In this study, RT-PCR approaches were developed for the detection of EDA and EDB FN mRNA isoforms in hepatocarcinoma cells (SK-Hep-I) grown in vitro and in human liver biopsies. While EDA+ and EDB+ isoforms were not present in control adult liver, they were detectable in the hepatocarcinoma cells and in fetal liver. The RT-PCR analysis, extended to biopsies of malignant and non-malignant hepatic tissues, showed that FN mRNAs containing the EDA and EDB sequences were present in the 14 hepatocellular carcinomas (HCCs) tested but absent in the non-tumorous liver tissues (i.e., normal parenchyma, non-specific reactive and chronic hepatitis, steatosis). The EDB+ FN mRNA isoforms were also detected in 3 cases of benign neoplasm (hepatocellular
adenoma
, HCA, I; nodular focal hyperplasia, NFH, 2), while the EDA+ was only detectable in I of the 2 cases of NFH. In addition, both EDA+ and EDB+ isoforms were expressed in 5 out of 9 cirrhotic livers surrounding the tumors. This molecular analysis, which can also be performed on small liver biopsies (2 mg), may therefore be a useful additional tool in the diagnosis of HCC.

The presence of the extracellular matrix glycoprotein tenascin was studied immunohistochemically in normal human salivary glands and in pleomorphic adenomas. Its expression was compared to that of
fibronectin
, and type IV collagen. In the normal salivary gland, tenascin was found with interruptions in periductal tissues, and continuously in blood vessels, fat cells and around nerve bundles. In pleomorphic
adenoma
, tenascin was detected surrounding the clusters of epithelioid cells, in areas with a myxoid and a chondroid matrix, and around some myoepithelial cells as a halo. As compared to
fibronectin
, there is a similar location of tenascin and
fibronectin
around tumor cell clusters but not in myxoid and chondroid matrices.

was found around the cells in chondroid matrix. In conclusion, tenascin is not only found in malignant tumors but also in benign tumors such as pleomorphic
adenoma
. The presence of tenascin as a halo around myoepithelial cells suggests a role of these cells in development of myxoid and chondroid matrices.
