The influence of soil contamination by inorganic and organic arsenic compounds on uptake, accumulation, and transformation of arsenic in pepper (
Capsicum
annum L.) was investigated in greenhouse pot experiments under controlled conditions. Pepper plants were cultivated in substrate amended by aqueous solutions of arsenite, arsenate, methylarsonic acid (MA), and dimethylarsinic acid (DMA) applied individually into cultivation substrate at concentrations of 15 mg As per kg of substrate. The plant availability of the arsenicals increased in the order arsenite = arsenate < MA < DMA. The highest arsenic concentrations were found in roots followed by stems, leaves, and fruits regardless of arsenic compound applied. In the control samples of pepper fruits, As(III), As(V), and DMA were present (25%, 37%, and 39% of the
water
-extractable arsenic). In control stems + leaves and roots, As(V) was the major compound (63% and 53% in a phosphate buffer extract) followed by As(III) representing 33% and 42%. Additionally, low concentrations (not exceeding 5%) of DMA and MA were detected as well. In all the soils analyzed after the first harvest of pepper fruits, arsenate was the dominating compound followed by arsenite. Methylarsonic acid, methylarsonous acid, and DMA were present at varying concentrations depending on the individual soil treatments. In the treated plants, the arsenic compounds in plant tissues reflected predominantly the extractable portions of arsenic compounds present in soil after amendment, and this pattern was more significant in the first part of vegetation period. The results confirmed the ability of generative parts of plants to accumulate preferably organic arsenic compounds, whereas in the roots and aboveground biomass, mainly inorganic arsenic species are present. Evidently, the source of soil arsenic contamination affects significantly the extractable portions of arsenic compounds in soil and subsequently the distribution of arsenic compounds within the plants.

The U-box motif is a conserved domain found in the diverse isoforms of E3 ubiquitin ligase in eukaryotes. From

-stressed hot pepper (
Capsicum
annuum L. cv Pukang) plants, we isolated C. annuum putative U-box protein 1 (CaPUB1), which encodes a protein containing a single U-box motif in its N-terminal region. In vitro ubiquitination and site-directed mutagenesis assays revealed that CaPUB1 possessed E3 ubiquitin ligase activity and that the U-box motif was indeed essential for its enzyme activity. RNA gel-blot analysis showed that CaPUB1 mRNA was induced rapidly by a broad spectrum of abiotic stresses, including drought, high salinity, cold temperature, and mechanical wounding, but not in response to ethylene, abscisic acid, or a bacterial pathogen, suggesting its role in the early events in the abiotic-related defense response. Because transgenic work was extremely difficult in hot pepper, in this study we overexpressed CaPUB1 in Arabidopsis (Arabidopsis thaliana) to provide cellular information on the function of this gene in the development and plant responses to abiotic stresses. Transgenic Arabidopsis plants that constitutively expressed the CaPUB1 gene under the control of the cauliflower mosaic virus 35S promoter had markedly longer hypocotyls and roots and grew more rapidly than the wild type, leading to an early bolting phenotype. Microscopic analysis showed that 35S::CaPUB1 roots had increased numbers of small-sized cells, resulting in disordered, highly populated cell layers in the cortex, endodermis, and stele. In addition, CaPUB1-overexpressing plants displayed increased sensitivity to
water
stress and mild salinity. These results indicate that CaPUB1 is functional in Arabidopsis cells, thereby effectively altering cell and tissue growth and also the response to abiotic stresses. Comparative proteomic analysis showed that the level of RPN6 protein, a non-ATPase subunit of the 26S proteasome complex, was significantly reduced in 35SCaPUB1 seedlings as compared to the wild type. Pull-down and ubiquitination assays demonstrated that RPN6 interacted physically with CaPUB1 and was ubiquitinated in a CaPUB1-dependent manner in vitro. Although the physiological function of CaPUB1 is not yet clear, there are several possibilities for its involvement in a subset of physiological responses to counteract dehydration and high-salinity stresses in transgenic Arabidopsis seedlings.

annuum L.), the excess or deficiency of the main cations involved in plant nutrition (K(+), Mg(2+), Ca(2+)) and two different degrees of salinity were related to the activity of plasma membrane H(+)-ATPase, the pH of the xylem sap, nutrient flux into the xylem (J(s)) and to a number of parameters related to
water
relations, such as root hydraulic conductance (L(0)), stomatal conductance (g(s)) and aquaporin activity. Excess of K(+), Ca(+) and NaCl produced a toxic effect on L(0) while Mg(2+) starvation produced a positive effect, which was in agreement with aquaporin functionality, but not with ATPase activity. The xylem pH was altered only by Ca treatments. The results obtained with each treatment could suggest that detection of the quality of the nutrient supply being received by roots can be related to aquaporins functionality, but also that each cation stress triggers specific responses that have to be assessed individually.

It has been previously shown that certain herbicides or plant extracts inhibited the viral infection. The goal of this study was to investigate the effect of Obuda pepper virus (ObPV) infection and herbicide or plant extract treatments on the photosynthetic processes of the host plants to get informations about the interactions of these factors. In
Capsicum
annuum-ObPV host-virus relations the virus infection slightly increased the activity of photosystem II (PSII), as it was supposed from fluorescence induction parameters. Chlorophyll content of leaves was also elevated probably due to virus-induced growth inhibition. The herbicide Stomp (active ingredient: pendimethalin) incorporated into the soil one week before planting (preplant treatment) together with virus infection even strengthened these effects in agreement with previous observations that this herbicide always did not prevent virus infection or reduce virus concentration in hosts. In ObPV-infected Nicotiana tabacum the structural changes showed similar tendency like in ObPV-infected C. annuum, but PSII efficiency did not significantly differ from that of the control. However, non-photochemical quenching (NPQ) increased because of the strongly decreasing CO2 fixation activity. Though simultaneous application of a
water
extract of Cirsium arvense shoot caused a little stronger inhibition of CO2 fixation, little loss in production was obtained due to significant reduction in virus concentration. In Solanum nigrum-ObPV relation the slightly increasing tendency of the values of actual PSII quantum efficiency could be related to the probably elevated ratio of reaction centre components (increased chlorophyll a/b ratio) in the thylakoids. Application of the herbicide Fusilade S (active ingredient: fluazifop-P-butyl) at 4-6 leaf stage as a post-emergence treatment practically prevented systemic virus infection and the virus-induced changes of photosynthesis are probably due to inhibiting the virus infection/replication process.

annuum L.) suffers with a characteristic leaf curl symptoms due to the attack of mite, Polyphagotarsonemus latus (Banks) (Acari: Tarsonemidae) and thrips, Scirtothrips dorsalis Hood (Thysanoptera: Thripidae) or both. Experiments were conducted in the fields of College of Agriculture, Shimoga, India during kharif (September 2003 to January 2004) and summer (March-June) 2004. After proving its pathogenicity, the potential of the mycopathogen, Fusarium semitectum was evaluated under field conditions using the popular chilli variety "Byadagi". annuum L.) and evaluate their toxic activities against some yeast species. Initially, proteins from seed flour were extracted in phosphate buffer, pH 5.4, for 3 h at 4 degrees C and the pellet obtained at 90% saturation with ammonium sulfate was heated at 80 degrees C for 15 min. The resulting suspension was clarified by centrifugation and the supernatant was extensively dialyzed against

; the peptide-rich extract was then named F/0-90. Cation-exchange chromatography was performed to separate low molecular mass proteins. One of the resulting fractions, named F3, enriched with basic proteins of 6-16 kDa, was submitted to reverse-phase chromatography in a C2/C18 column by HPLC, resulting in four fractions denominated RP1, RP2, RP3 and RP4. When these fractions were submitted to N-terminal sequencing, the comparative analysis in databanks revealed homology for two of these peptides, isolated from fractions RP3 and RP4, with sequences of proteinase inhibitors and 2S albumins, respectively. The F3 fraction, rich in peptides, inhibited the growth of yeasts Saccharomyces cerevisiae, Candida albicans, Candida parapsilosis, Candida tropicalis, Pichia membranifaciens, Kluyveromyces marxiannus and Candida guilliermondii. The RP3 and RP4 fractions showed high inhibitory activity against the growth of the yeast S. cerevisiae. The F3 fraction was also able to inhibit glucose-stimulated acidification of the medium by yeast cells of S. cerevisiae and to cause several morphological changes in different yeasts, such as cell wall disorganization, bud formation as well as the formation of pseudohyphae.

Significant differences of nitrate N content in 0-20 cm soil layer were observed among different treatments. The control had the highest nitrate N content (50.33 mg . kg(-1)), followed by straw mulch (31.98 mg . kg(-1)) and straw + plastic film mulch (31.96 mg . kg(-1)), and plastic film mulch and applying

preserving agent. Compared with the control, soil covering could increase the nitrate N use efficiency of pepper, and decrease the accumulation of nitrate N in plough layer. In 0-20 cm soil layer, treatment plastic film mulch had the lowest available phosphorus content (0.72 mg . kg(-3)), and the second (0. 92 mg . kg(-1)) was the treatment straw + plastic film mulch. annuum L cv. California) or of plants treated with 50 mM NaCl, 10 mM CaCl(2) or 10 mM CaCl(2) + 50 mM NaCl. Under saline conditions, osmotic
water
permeability (P ( f )) values decreased in protoplasts and plasma membrane vesicles, and the same reduction was observed in the PIP1 aquaporin abundance, indicating inhibitory effects of NaCl on aquaporin functionality and protein abundance. The cytosolic Ca(2+) concentration, [Ca(2+)](cyt), was reduced by salinity, as observed by confocal microscope analysis. Two different actions of Ca(2+) were observed. On the one hand, increase in free cytosolic calcium concentrations associated with stress perception may lead to aquaporin closure. On the other hand, when critical requirements of Ca(2+) were reduced (by salinity), and extra-calcium would lead to an upregulation of aquaporins, indicating that a positive role of calcium at whole plant level combined with an inhibitory mechanism at aquaporin level may work in the regulation of pepper root
water
transport under salt stress. However, a link between these observations and other cell signalling in relation to water channel gating remains to be established.

ABSTRACT The first characterization of alterations in whole-plant photosynthetic rate and carbon assimilation of bell peppers associated with infection by Pythium aphanidermatum is described. Relationships of root disease caused by P. aphanidermatum to whole-plant net carbon exchange rate (NCER), total carbon accumulation, dark respiration rates,
water
loss, and destructive growth parameters were quantified in vegetative, hydroponically grown pepper plants (
Capsicum
annuum 'Cubico'). Inoculated plants displayed lower whole-plant NCER. This translated into a loss of 28% in cumulative C gain during 7 days after inoculation and occurred before visible shoot symptoms developed. Leaf area and dry weight of shoots and roots were significantly decreased and the shoot/root ratio was higher in inoculated plants than in noninoculated plants. We propose that reduced NCER in inoculated plants was mainly due to restricted development of leaf area, because no differences in NCER and evapotranspiration were observed between control and inoculated plants when expressed based on leaf area and root dry mass, respectively. These findings indicate that Pythium infection did not affect the photosynthetic apparatus directly and that the reductions in photosynthesis and growth were not caused by inefficient
water
transport by diseased roots. These results enlarge on the understanding of physiological responses of host plants to early stages of root disease.
