Bacterial endotoxins alter the permeability of endothelium, but little is known of their effect on epithelium. We exposed specific pathogen-free rabbits to aerosolized Pseudomonas aeruginosa LPS or saline and performed serial measurements of RL, Cdyn, BP, WBC count and differential, and platelet counts. Pulmonary 99mTc-DTPA half-life was measured 4, 6, or 8 h after exposure. The animals were sacrificed and BAL performed. Background and PMA-stimulated superoxide production was measured from individual AM using electrooptical determination of reduction of NBT. Lung tissue was processed for light microscopy and ratio of wet to dry weight. 99mTc-DTPA half-life was significantly shorter in LPS-exposed animals at 6 h (p < 0.05) and 8 h (p < 0.001). There were no differences in Cdyn, RL, BP, WBC, differential, platelet, or BAL cell count at any time between groups. No histologic changes or differences in lung wet to dry weight ratios were found. PMA-stimulated AM superoxide production was significantly increased (p < 0.01) in LPS-exposed animals. This effect was time dependent and could be duplicated in AM from control animals following a 4-h incubation with LPS, lavage fluid from LPS-exposed animals, or recombinant murine TNF. These results demonstrate that aerosolized Pseudomonas LPS increases pulmonary epithelial permeability and primes AM superoxide production.

Male mice given different oral doses (0.05, 0.03 or 0.02 x LD50/animal/day) of styrene (LD50 = 1 g/kg) daily for 5 days did not incite any overt toxicity in lymphoid organs or on hematologic parameters. At the tested dose levels styrene produced a mild reduction in the organ weight of adrenal and spleen and slight reduction in the cellular viability of lymph nodes. There was a dose-dependent suppression in the humoral immune response (IgM-producing PFCs of spleen and serum anti-SRBC HA titre) to SRBC. The proliferative response to the B-cell mitogen, LPS however revealed a significant increase in the incorporation of 3HT with middle and lowest doses of styrene. The results of cell-mediated immunity appeared somewhat unexpected and more complex as exposure resulted in a dose-dependent enhancement in the cutaneous DTH reaction to SRBC together with increased blastogenic response of splenic lymphocytes to phytohaemagglutinin (PHA). Additionally, there was significant impairment in the functional activity (NBT reduction, attachment and phagocytic indices) of nonadherent and adherent peritoneal exudate cells. Based on the present data the study identifies the immunotoxic potential of styrene and which acts differently on various arms of the rodent's immune system.

Pathomorphological and immunological alterations caused by a mixture of styrene and dioctyl phthalate were studied in albino mice following oral administration of 0.02, 0.03, 0.05 x LD50 of the mixture. The chemicals were mixed together proportionate to their respective LD50 values and fed in ground nut oil, 5 d/wk for 4 weeks. Histological examination of spleen revealed considerable depletion of cellular population of lymphoid follicles which corresponded to the dose dependent decrease in splenic mononuclear cell population count. The thymic lobules revealed slight atrophy but accompanied by a significant increase in thymocyte population. Correspondingly few significant histological changes were observed in mesenteric and peripheral lymph nodes. The treatment caused impairment of primary humoral immune response to SRBC (IgM) but there was a significant increase in response of splenocytes to B-cell mitogen LPS. There was a suppression of cutaneous delayed type hypersensitivity and increase in splenic lymphocyte response to T-cell mitogen PHA. Simultaneously, indirect immunity represented by decreased phagocytosis and enhanced metabolic function of reducing NBT by peritoneal exudate cells was observed. The in vitro exposure of vero cells to the mixture caused dose dependent protective effect. The results of present study indicate that subchronic exposure to low doses of mixture of styrene and dioctyl phthalate under certain conditions may modulate some of the immune functions as compared to exposure to either chemicals alone.

In view of great species differences in biology of polymorphonuclear cells, and non-availability of basic data on buffalo PMN cells for assessing their functional activity, the present work on the immuno-defence system involving protein synthesis and O2- production was undertaken to highlight the immunomodulatory role of thyroxine. Digitonin, LPS and Con-A activation generated superoxide, which was monitored by NBT reduction. The study suggested that concanvalin A (Con-A) and T4 were able to synergetically increase the production of superoxide and H2O2. The likely involvement of thyroxine in activation was studied by [125I]thyroxine incorporation, which was significantly increased due to activation. In contrast, aflatoxin B1 together with Con-A caused a significant decrease (P < 0.05) in incorporation of [125I]T4. Optimum time dependence in [14C]leucine incorporation by buffalo PMN cells was found to be 30 min and the factors like T4 (7.7 ng/ml) and glutathione (400 micrograms/ml) significantly enhanced the incorporation. In contrast, antiinflammatory agent, indomethacin (40 micrograms/ml) inhibited protein synthesis in PMN cells; while puramycin also significantly lowered the [14C]leucine incorporation. Total [14C]leucine incorporation in acid extractable cationic proteins and peptides, known for their antibacterial properties was found to be 30-40% when separated on AU-PAGE. The studies revealed the in vitro immunomodulatory role of T4 in O2-, H2O2 production and cationic protein synthesis by the activated PMN cells of buffalos.

The evaluation of peripheral blood neutrophils functional state in 27 patients with progressive bilateral and 7 patients with sudden unilateral sensorineural hearing loss (SNHL) before and in the first period of ubiquitine therapy was performed. The metabolic activity of cells in NBT reduction tests: spontaneous (NBT-sp) and stimulated with LPS E. coli (NBT-st) were examined. The results in mean percentage and absolute number of NBT-positive cells, relative coefficient of neutrophil metabolic activity (CNMA) and also calculated NBT index were presented. Significant percentage of patients with increased NBT-sp test values before treatment and in the second examination were noticed. Relative CNMA was 3-6 times higher than normal values. The neutrophils of patients had a remarkably decreased ability to response to LPS stimulation. In patients with progressive bilateral SNHL during treatment with TFX considerable increase of patients (from 37.9 to 76.5%) with normal values in NBT-st reduction tests was found. The increased values of NBT-sp reduction tests and relative CNMA may suggest the presence of inflammation process or infection. The decreased values of NBT index which were observed confirm the existence of disturbances in neutrophils metabolic activity and lack of functional reserve of these cells. Usefulness of NBT reduction tests in patients with SNHL is discussed.

Lipopolysaccharide fraction isolated from Ehrlich ascites carcinoma (E-LPS) was investigated as an antitumor agent against human leukemia cell ML-2. Marked cell growth inhibition was observed with ML-2 cell accompanied by inhibition of DNA synthesis and perturbation of cell cycle. Induction of differentiation in treated ML-2 cells was observed as indicated by morphological maturation, NBT reducing activity and indirect immunofluorescence.

Several in vitro studies were performed to study the cellular reaction of European sea bass (Dicentrarchus labrax L.) against Sphaerospora dicentrarchi. Head kidney phagocytes were obtained from parasitised (P) and non-parasitised (NP) fish. The production of superoxide anion (O2-), tested by the NBT method, was higher in P than in NP fish. The addition of increasing amounts of sea bass serum (SBS) produced a gradual increment of the respiratory burst with SBS from parasitised animals (P-SBS), whereas this increment reached a plateau at lower concentration with SBS from nonparasitised ones (NP-SBS). O2- production was higher when adding NP-SBS than with P-SBS or fetal bovine serum. Heat inactivation of NP-SBS and FBS reduced the respiratory burst significantly, whereas it did not change the effect of P-SBS. The number of NBT-positive cells after particulate stimulation was significantly higher using S. dicentrarchi spores than using SRBC, but lower than with phorbol myristate acetate. Phagocytes primed overnight with spore extracts produced higher amounts of O2- than those LPS-primed or non-primed ones. A similar percentage of phagocytosis was detected using glutaraldehyde fixed spores and SRBC. Most of the phagocytes engulfed three or more SRBC, whereas most of the phagocytes engulfed only one spore. Complement mediated opsonisation by NP-SBS may occur, as the phagocytic index was reduced when the serum was heat inactivated.

The aim of the study was to evaluate the functional state of peripheral blood neutrophils in patients with chronic active viral hepatitis. Twenty-six patients with HBV, HCV or CMV in different clinical status were included in the analysis. In the study, the number of leukocytes and neutrophils was determined. The metabolic activity of neutrophils was examined in NBT reduction tests i.e. spontaneous (NBTsp) test and the one stimulated with LPS E. coli (NBTst). The results were analysed in relation to disease advancement and the type of viral infection. The data obtained from the affected patients were compared with the results from 46 healthy subjects. Most patients displayed neutropenia. It was found that the number of NBT positive cells and the coefficient of neutrophil metabolic activity (CNMA) in NBT sp test were highly significantly increased. The majority of patients had reduced values in NBTst test, which suggested lack of response to LPS E. coli by neutrophils. These findings may reflect the state of inflammation in the body. NBT reduction tests may be useful in monitoring metabolic activity of neutrophils in patients with chronic active viral hepatitis.

Immunosenescence is an age-associated dysregulation of the immune function, which contributes to increased susceptibility to disease in the elderly. Alveolar macrophages (AM) are known phagocytes that generate reactive oxygen species (ROS) and nitric oxide (NO), essential mediators for host defence. We studied phagocytosis, ROS and NO production in AM obtained from young, adult and senescent rats (1-2, 9-12 and 18-24 months old, respectively) after exposure to lipopolysaccharide (LPS, 0.1-10 microg mL(-1)), 12-O-tetradecanoylphorbol 13-acetate (TPA, 0.1 microg mL(-1)) or LPS + TPA in culture. Phagocytosis was significantly lower in control AM from adult rats than in AM from young animals. Nevertheless, AM from adult animals pretreated with LPS exhibited higher phagocytic capacity than AM from younger animals. ROS was identified by the NBT test at single cell level and quantified by automated image analysis. When TPA was added to all three populations, AM from adult and senescent animals responded more than AM from young animals. All LPS-stimulated AM produce more NO than controls. However, NO production increased three-, four- and two-fold in young, adult and senescent animals, respectively. Our results demonstrate that AM from young, adult and senescent animals display differential responsiveness to inflammatory mediators. Therefore, aging processes markedly affect AM metabolic functions and may further compromise the lung immune defence response, increasing adverse long-term health effects.

The nitric oxide synthase (NOS) activity in the haemocytes of shrimps Fenneropenaeus chinensis (Osbeck) and Marsupenaeus japonicus (Bate) was studied after white spot syndrome virus (WSSV) infection to determine its characteristics in response to virus infection. First, the NOS activity in haemocytes of shrimps was determined by the means of NBT reduction and changes in cell conformation. And the variations of NOS activity in shrimps after challenge with WSSV intramuscularly were evaluated through the analysis of L-citrulline and total nitrite/nitrate (both as NO derivates) concentrations. The result showed that NOS activity in the haemocytes of F. chinensis increased slightly from 0 to 12h postchallenge, indicated by the variations of l-citrulline (from 11.15+/-0.10 to 12.08+/-0.64 microM) and total nitrite/nitrate concentrations (from 10.45+/-0.65 to 12.67+/-0.52 microM). Then it decreased sharply till the end of the experiment (84 h postchallenge), the concentrations of l-citrulline and total nitrite/nitrate at 84 h were 1.58+/-0.24 and 2.69+/-0.70 microM, respectively. The LPS-stimulated NOS activity kept constant during the experiment. However, in M. japonicus, the NOS activity kept increasing during the first 72 h postchallenge, the concentrations of l-citrulline and total nitrite/nitrate increased from 7.82+/-0.77 at 0 h to 10.79+/-0.50 microM at 72 h, and from 8.98+/-0.43 at 0 h to 11.20+/-0.37 microM at 72 h, respectively. Then it decreased till the end of the experiment (216 h postchallenge), and the concentrations of l-citrulline and total nitrite/nitrate at 216 h were 5.66+/-0.27 and 4.68+/-0.16 microM, respectively. More importantly, an apparent increase of LPS-stimulated NOS activity was observed in M. japonicus at 48 h postchallenge, which was about 4 times higher than that in the control group of health shrimps. In correspondence with the difference of NOS activity between the two species of shrimps, the cumulative mortalities of the shrimps were also different. All shrimps of F. chinensis in the mortality experiment died in 66 h, much more quickly than M. japonicus, whose accumulative mortality reached 100% after 240 h. Data here reported let us hypothesize that NOS activity in the haemocytes of shrimps F. chinensis and M. japonicus responses to WSSV infection differently, and this might be one of the reasons for the different susceptibility of F. chinensis and M. japonicus to WSSV infection.
