A study of neutrophil functions has been performed in 17 patients with advanced cancer and in 21 controls. Their phagocytic activity, their index of reduction of
NBT
and their
myeloperoxidase
activity were examined. The reslts suggest that cancer patients might have a "factor" or "factors" in their plasma which interfereu (s) with phagocytosis and which promote (s) exocytosis of lysosomal enzymes ("reverse endocytosis") from from autologous polymorphonuclear leucocytes as well as from cells obtained from normal healthy donors.

A complete lack of
myeloperoxidase
(
MPO
) was demonstrated in a boy suffering from acute myeloic leukemia during the acute phase of the disease and after a remission was achieved. A partial defect of
MPO
was demonstrated in the patient's father, no further abnormalities were seen in other members of the family. The fine structure of the patient's neutrophils and monocytes appeared normal, no activity of
MPO
was demonstrated on the fine structural level. In the father's neutrophils transitional forms between cells exhibiting a normal
MPO
activity and those without activity were demonstrated. The neutrophil bactericidal activity was strongly inhibited in the patient and decreased in his father. Normal values were found in:
NBT
test, chemotaxis, serum-dependent phagocytosis, number of B and T lymphocytes, serum immunoglobulins, and complement. A possible connection between
MPO
deficiency and leukemia is discussed.

The dose-dependent effect of L-asparaginase (Crasnitin, Bayer) on the serum IgG, IgA and IgM content was studied in 14 children with acute lymphoblastic leukemia. This effect was less evident in the intracellular metabolism of peripheral blood granulocytes (studied by the

A total of 140 miners, divided into 4 groups were studied. The first group consisted of 25 practically healthy people, newly employed, examined at a health center of the mine, the second of 21 patients with anthracosilicosis, the third of 36 with anthracosilicosis confirmed by X-ray, the fourth included 58 facing the first stage of anthracosilicosis. Erythrocyte histidine and catecholamine levels,

and beta-glucuronidase activities, data of lysosomal cation (LC) and

tests in neutrophils were under study. The miners of groups 2, 3, 4 showed the increased beta-glucuronidase activity (by 47-73%) and
NBT
test values (37-96%), lowered levels of catecholamines (57-67%), histidine (48-60%), results of LC-test (29-32%) and
myeloperoxidase
activity (21-31%) in comparison with normal subjects. The findings will help diagnose the latent forms of anthracosilicosis comparatively early, when the structural changes cannot be detected by X-ray.

activity, amount of cationic proteins, expression of Fc receptors, spontaneous and induced luminol-dependent chemiluminescence, neutrophil bactericidal action against H-thymidine-labelled Staphylococcus aureus (strain 209), amount of blood immunoglobulins, and the modulating effect of serum of patients on the chemiluminescence of neutrophils of healthy donors. The control group consisted of 35 volunteers whose ages ranged from 18 to 50 years. Basic differences were revealed between the indices of neutrophil activity in patients with an uneventful postoperative period and those of patients with infectious complications in the preoperative period which became much graver in the early postoperative period. This suggests that neutrophilic dysfunctions play a significant role in the development of postoperative infectious complications in this category of patients.

1.25 (OH)2D3 is a potent inducer of differentiation of leukaemic cells into a monocytic direction. However, therapeutic application is difficult because of the development of hypercalcaemia. We examined a novel vitamin D analogue, MC 903, which is at least 100 times less effective on calcium metabolism in rats than 1.25 (OH)2D3. Using the HL-60 cell line, differentiation was measured with a comprehensive panel of qualitative and quantitative parameters. Development of monocytic cells was shown morphologically, immunophenotypically and functionally by increased capability of reducing
NBT
(vs cultures without MC 903, p less than 0.0001) and by qualitatively and quantitatively increased non-specific esterase activity. Furthermore, a concomitant decreased activity of
myeloperoxidase
and lactate dehydrogenase was noticed. In conclusion, MC 903 is a potent inducer of monocytic differentiation, comparable with 1.25 (OH)2D3 and will therefore be an interesting and potential therapeutic agent for studies in human acute leukaemia.

To avoid extensive manipulation for the purification of RNA from cells, several methods were evaluated for the direct release of RNA from influenza virus infected cells and supernatants using slot blot hybridization and non-radioactive probes. Treatment with an equal volume of 10 M aqueous guanidine hydrochloride produced the best hybridization signal. Less, but significant amounts of RNA were also released using the following treatments: dilute alkali (final concentration of 0.16 M NaOH) or 100 degrees C/5 min or RNA sample buffer containing formamide/formaldehyde, then heating at 65 degrees C/10 min. Despite the presence of large amounts of cell debris, RNA from guanidine hydrochloride treated whole cell extracts bound quantitatively to the positively charged nylon membranes. The sensitivity of RNA detection when whole cell extracts treated with guanidine hydrochloride were probed with a digoxigenin labelled cDNA probe was similar to the detection of RNA in highly purified, protein free samples. Three positively charged membranes were tested (from Amersham, ICN and Boehringer Mannheim) using two alkaline phosphatase substrates,

reaction with the TMB substrate was the most consistently sensitive, irrespective of which membrane was used. The ability to quantitatively detect RNA from whole cells without any purification will allow the rapid screening of large numbers of samples for specific RNA species in research or diagnostic laboratories.

test was highly sensitive to cytostatic therapy. Tumor dissemination and morphologic variant contributed to changes in the neutrophilic morphology and function.
