The complete amino acid sequence of the biotinyl subunit from the enzyme transcarboxylase of Propionibacterium shermanii has been determined from the structures of overlapping tryptic and cyanogen bromide peptides together with sequenator analysis on the whole subunit. The subunit contains 123 amino acid residues. Eleven of nineteen residues in the region of biotin attachment, when compared to pyruvate carboxylase from avian liver (Rylatt, D. B., Keech, D. B., and Wallace, J. C. (1977) Arch. Biochem. Biophys. 183, 113-122), were found to be in identical positions relative to biocytin. There was less homology with acetyl-CoA carboxylase from Escherichia coli (Sutton, M. R., Fall, R. R., Nervi, A. M., Alberts, A. W., Vagelos, P. R., and Bradshaw, R. A. (1977) J. Biol. Chem. 252, 3934-3940), but in all of these biotin enzymes there was an alanylmethionyl-biocytinyl-methionine sequence. The secondary structure of the biotinyl subunit has been estimated using the method of Chou and Fasman (Chou, P. Y., and Fasman, G. D. (1978) Adv. Enzymol. 47, 45-148) and considered in relationship to the role of the biotinyl subunit in the structure and function in transcarboxylase.

A method for the isolation of Leishmania donovani amastigotes from infected hamster spleen and liver tissues is described. Over 85% of the isolated amastigotes were viable as judged by acridine orange-ethidium bromide staining and in vitro transformation to the promastigote form. A comprehensive survey of the enzymes of carbohydrate metabolism in L. donovani amastigotes and promastigotes was conducted. Amastigotes and promastigotes possess all of the enzymes of the Embden-Meyerhof pathway, hexose monophosphate shunt, and tricarboxylic acid cycle. Cell-free extracts of both forms demonstrate an active glutamate dehydrogenase, thus linking activity which permits entry of pyruvate into the tricarboxylic acid cycle. Both forms demonstrate an active glutamate dehydrogenase, thus linking amino acid metabolism with carbohydrate metabolism. Pyruvate carboxylase, the enzyme responsible for replenishment of C4 acids by heterotrophic CO2 fixation into pyruvate, was also demonstrable in the tissue and insect forms. In general, activities of promastigote enzymes are higher than the amastigote enzymes. Differences between the vertebrate (amastigote) and invertebrate (promastigote) forms in their potential to utilize carbohydrates as substrates would appear to be quantitative rather than qualitative.

Pyruvate carboxylase from Pseudomonas citronellolis is composed of non-identical subunits which include a larger biotin-containing polypeptide (alpha) of Mr = 65,000, and a smaller biotin-free polypeptide (beta) of Mr = 54,000. We have investigated these two polypeptides by analyzing their amino acid composition, cyanogen bromide peptide maps, and immunochemistry. The results showed that the subunits of the enzyme have quite different properties. Antibodies prepared against the polypeptides were used as probes of the catalytic functions of the subunits. Immunotitration studies indicated that only anti-alpha inhibited enzyme activity. The antibiotin fraction of this antibody population was removed by passage through biotin-Sepharose (anti-alpha'). Titration curves using anti-alpha' showed identical inhibition when total pyruvate carboxylase activity, ATP/Pi exchange activity, and pyruvate/oxalacetate exchange activity were measured, suggesting that both active sites are located on the alpha polypeptide. The arrangement of the subunits in the quaternary structure was investigated by means of the surface probe carbonic anhydrase linked to toluene isocyanate, and by partial digestion experiments with trypsin, chymotrypsin, and pronase. The results indicated that the alpha polypeptides are on the outside of the molecule and the beta polypeptides are the internal subunits.
