Ammoniacal silver solutions give striking impregnation of Alzheimer's disease (AD) lesions if sections are pretreated with copper sulfate and hydrogen peroxide. In contrast to most silver impregnation methods, no staining of normal neurites is obtained, and senile plaques (SPs), neurofibrillary tangles (NFTs), and neuropil threads (NTs) are strongly stained in black against a clear background. A sodium acetate wash interposed between the copper sulfate and hydrogen peroxide resulted in suppression of the staining of amyloid lesions. This variant of the basic procedure (CPS-II method), maintains the capacity of the latter (CPS-I method) to strongly impregnate NFTs and NTs. In addition, it clearly delineates the dystrophic neurites of SPs obscured by the strong argyrophilia of the amyloid deposits seen in CPS-I stains. NTs are strongly impregnated with both CPS-I and CPS-II methods and are unmasked from normal neurites, which remain unstained. The staining can be abolished by pretreatment with formic acid and erased with a brief wash in sulfochromic acid. Destained sections can be restained with either method or with immunoperoxidase procedures. CPS staining of previously immunostained tissues produces marked intensification of the diaminobenzidine reaction product. In AD brains, the immunostaining is markedly enhanced and selective when the silver procedure is preceded by formic acid treatment. The selectivity and high sensitivity of the procedure may be useful as a diagnostic tool and of value to study the biogenesis and natural evolution of the brain lesions of AD.(ABSTRACT TRUNCATED AT 250 WORDS)

The current study investigates the influence of three dentin hypersensitivity treating agents (Gluma CPS, MS Coat and Saforide) on bond strength to dentin of two luting agents (Panavia Fluoro Cement and Super-Bond C & B). Sixty bovine dentin substrates were divided into 12 combinations of four treatment conditions (Gluma CPS, MS Coat, Saforide and control) and three adhesive systems (AD Gel sodium hypochlorite + Panavia Fluoro Cement, Panavia Fluoro Cement without AD Gel and Super-Bond C & B). After bonding the treated teeth to steel rods, 24-h tensile bond strengths were determined, and average values (n=5) were compared by analysis of variance (ANOVA). Without application of the desensitizers, bond strengths of the two groups (Super-Bond C & B, 10.2 MPa; AD Gel + Panavia, 11.5 MPa) were comparable, and they were greater than the group bonded with the Panavia material with no AD Gel conditioning (7.1 MPa). Application of the Saforide ammoniated silver fluoride desensitizer reduced bond strength of both the Super-Bond and Panavia luting agents, whereas the MS Coat polymeric agent negatively affected bond strength of the Panavia cement only. The use of the Gluma desensitizer did not affect bond strength of any of the three adhesive systems, and the bond strength of the Panavia cement with the AD Gel conditioning was not reduced by application of any of the three desensitizers. The four combinations of two desensitizers (MS Coat and Gluma CPS) and two adhesive systems (Super-Bond C & B and AD Gel + Panavia) are deemed to be applicable to fixed prosthodontic treatment.

The aim of the present study was to investigate the effect of silver nanoparticles (AgNP) of different sizes toward two primary producer aquatic species. Thalassiosira pseudonana and Synechococcus sp. have been selected as representative models for the lower trophic organisms in marine and freshwater habitats, respectively. Time-dependent cellular growth was measured upon exposure to both AgNP and silver nitrate (AgNO(3)). In addition, AgNP behavior in freshwater and marine waters has been followed by CPS disc centrifuge, in the time frame of AgNP exposure studies, and the kinetic release of silver from AgNP of different sizes was measured by dialysis and inductively coupled plasma mass spectrometry (ICP-MS). The combination and interpretation of all these data suggest that a shared effect of AgNP and released silver was responsible for the toxicity in both organisms. Furthermore, the toxic effects induced by AgNP exposure in the present study seem to result from a mixture of parameters including aggregated state, size of the AgNP, stability of the preparation, and speciation of the released silver.

To understand the olfactory mechanisms of Holotrichia parallela antennae in detecting volatile compounds in the environment, protein profiles of H. parallela antennae were analyzed using two-dimensional electrophoresis followed by mass spectrometry and bioinformatics analyses. Approximately 1,100 protein spots in silver staining gel were detected. Quantitative image analysis revealed that in total 47 protein spots showed significant changes in different genders of adult antennae. Thirty-five differentially expressed proteins were identified by Matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF/TOF) tandem mass spectrometer, among which 65.7% are involved in carbohydrate and energy metabolism, antioxidant system, transport, and amino acid/nucleotide metabolism. Some proteins identified here have not been reported previously in insect antennae. Identified male-biased proteins included odorant-binding protein 4, pheromone-binding protein-related protein 2, odorant-binding protein 14, prophenoloxidase-I, acyl-CoA dehydrogenase, aldo-keto reductase-like, carbamoyl phosphate synthetase, etc. whereas some proteins are female biased, such as antennae-rich cytochrome P450, aldehyde dehydrogenase, and putative glutamine synthetase. Alterations in the levels of some proteins were further confirmed by real time polymerase chain reaction (RT-PCR). The proteomic resources displayed here are valuable for the discovery of proteins from H. parallela antennae.
