Two sporulating bacterial strains designated CECAP06(T) and CECAP16 were isolated from the rhizosphere of the legume Cicer arietinum in Argentina. Almost-complete 16S rRNA gene sequences identified the isolates as a Paenibacillus species. It was most closely related to Paenibacillus cineris LMG 18439(T) (99.6 % sequence similarity), Paenibacillus favisporus LMG 20987(T) (99.4 % sequence similarity) and Paenibacillus azoreducens DSM 13822(T) (97.7 % sequence similarity). The cells of this novel species were motile, sporulating, rod-shaped, Gram-positive and strictly aerobic. The predominant fatty acids were anteiso-C(15 : 0), C(16 : 0) and iso-C(16 : 0). The DNA G+C content of strains CECAP06(T) and CECAP16 was 51.3 and 50.9 mol%, respectively. Growth was observed from many carbohydrates, but gas production was not observed from glucose. Catalase and oxidase activities were present. The isolates produced beta-galactosidase and hydrolysed aesculin. Gelatinase, caseinase and urease were not produced. The results of DNA-DNA hybridization showed that the strains from this study constitute a novel species of the genus Paenibacillus, for which the name Paenibacillus rhizosphaerae sp. nov. is proposed. The type strain is CECAP06(T) (=LMG 21955(T) = CECT 5831(T)).

Bacterial strain PETP02(T) was isolated from nodules of Trifolium pratense growing in a Spanish soil. Phylogenetic analysis of the 16S rRNA gene sequence showed that this strain represents a member of the genus Phyllobacterium. However, divergence found with the 16S rRNA gene sequence of the single recognized species of this genus, Phyllobacterium myrsinacearum, indicated that strain PETP02(T) belongs to a different species. The results of DNA-DNA hybridization, phenotypic tests and fatty acid analyses confirmed that this strain represents a novel species of the genus Phyllobacterium, for which the name Phyllobacterium trifolii sp. nov. is proposed. The type strain is PETP02(T) (=LMG 22712(T)=CECT 7015(T)). This strain was strictly aerobic and used several carbohydrates as carbon source. It was not able to reduce nitrate. Aesculin hydrolysis was negative. It did not produce urease, arginine dihydrolase, gelatinase or beta-galactosidase. The DNA G+C content was 56.4 mol%. The nodD gene of this strain showed a sequence closely related to those of strains able to nodulate Lupinus. Infectivity tests showed that this strain is able to produce nodules in both Trifolium repens and Lupinus albus.

Three strains (N002, N069 and PT31(T)) of a novel thermotolerant methylotrophic yeast species belonging to the genus Pichia were isolated from soil collected in Thailand by three consecutive enrichments in methanol broth at room temperature. They were categorized as thermotolerant strains on the basis of their good growth below 20 degrees C and up to a high temperature (37 degrees C). The major characteristics of the three strains included the following and placed them in the genus Pichia: the formation of four helmet-/hat-shaped ascospores in a deliquescent ascus that might be unconjugated or produced by conjugation between a cell and its bud or between independent cells; multilateral budding; the presence of ubiquinone Q-7; negative for Diazonium blue B colour and urease reactions; and the absence of arthrospores and ballistospores. The three strains differed by one to three nucleotide substitutions in the sequences of the D1/D2 domain of the large-subunit rDNA sequence. Phylogenetic analysis revealed that their closest species was Pichia dorogensis, but with 11-13 nucleotide substitutions in 554 nt. The phenotypic characteristics of the three strains were the same. The strains could be distinguished from P. dorogensis by a number of phenotypic characteristics. On the basis of the above findings, these three strains were assigned to a single novel species of Pichia, for which the name Pichia thermomethanolica sp. nov. is proposed. The type strain is PT31(T) (=BCC 16875(T)=JCM 12984(T)=CBS 10098(T)).

A strain GZ6 that can biodegrade LAS (Linear Alkylbenzene Sulphonate) is identified. It is aerobic gram-negative rod or short-rod (0.5 to 0.8 by 1.0 to 2.0 Mm). It is mobile with a single polar flagellum. Optimum growth occurred at 30 degrees C and pH7.0. It is catalase positive, urease positive, and arginine decarboxylase positive. All the other physiological and biochemical tests performed were negative. It utilizes the xenobiotic compounds chloridazon, antipyrin and LAS as sole carbon sources. Most sugars, alcohols, and carboxylic acids are not utilized. It has Q-10 as the major quinone. The main cell fatty acids are Sum7, C16:0 and Sum4. The DNA G + C mol % content is 70.10. A phylogenic tree was constructed on the basis of 16S rDNA sequences. It showed that the previously known member of the genus Phenylobacterium, Phenylobacterium mobile DSM1986T, is the nearest neighbor to strain GZ6. The level of binary sequence similarity between them is 97.49%. And the DNA-DNA relatedness is 40%. These genetic analysis and their morphological difference show that they are different species of Phenylobacterium. A new species, Phenylobacterium mobile sp. nov., has been proposed.

Strain CP2CT was isolated from biological soil crusts in the Colorado Plateau, USA. The isolate was aerobic, facultatively fermentative, Gram-negative, non-motile and red-pigmented (due to the presence of carotenoids), but did not contain bacteriochlorophyll a. The strain tested positive for catalase, oxidase and urease and was negative for lysine and ornithine decarboxylases and arginine dihydrolase. The major fatty acids present were C(18 : 1)omega7c and C(16 : 0). It had a high DNA G+C content of 75 mol%. Comparisons of 16S rRNA gene sequences identified bacteriochlorophyll a-producing strains of Paracraurococcus ruber (94.9 %), Craurococcus roseus (92.2 %) and Roseococcus thiosulfatophilus (92.3 %), as well as non-bacteriochlorophyll a-producing bacteria Muricoccus roseus (94.9 %), Roseomonas gilardii (94.2 %) and Roseomonas mucosa (93.8 %), as the bacteria most closely related to strain CP2CT. Phylogenetically, CP2CT was placed roughly equidistantly from the above organisms. Based on its phylogenetic placement and morphological and physiological characteristics, strain CP2CT is assigned to a new genus in the alpha-1 subgroup of the Proteobacteria, for which the name Belnapia gen. nov. is proposed. Strain CP2CT (= ATCC BAA-1043T = DSM 16746T) is proposed as the type strain of the type species of this genus, with the name Belnapia moabensis gen. nov., sp. nov.

A novel aerobic, chemoheterotrophic, bacteriochlorophyll-containing bacterium, strain OCh 323T, was isolated from sand at Monkey Mia, Shark Bay, located on the west coast of Australia. The cells were Gram-negative, non-motile rods of variable length; one or both cell poles was narrower. Bacteriochlorophyll a was synthesized under aerobic conditions. Catalase, oxidase and urease were produced. The ONPG reaction was positive. The major component of the cellular fatty acid was octadecenoic acid (18:1). The DNA G+C content was 68.1 mol%. The results of 16S rRNA gene sequence comparisons revealed that strain OCh 323T formed a novel, separate line of descent within the alpha-3 group of the Alphaproteobacteria. The similarity value for the 16S rRNA gene sequence of strain OCh 323T and that of the most closely related species, Rhodovulum sulfidophilum, was 91.4%. It is concluded that strain OCh 323T (=JCM 11220T=CIP 107377T) should be placed in a novel genus, Roseibacterium gen. nov., as the type strain of a novel species Roseibacterium elongatum sp. nov.

Four isolates from freshly caught fish samples obtained from the South Atlantic Ocean off the South African coastline were shown to represent a novel species in the genus Chryseobacterium by means of a polyphasic taxonomic study. The four isolates had virtually identical whole-cell protein profiles, fatty acid profiles and biochemical properties. Analysis of the 16S rRNA sequence of strain LMG 23089(T) revealed 99.3 and 98.9 % similarity to the 16S rRNA sequences of the type strains of Chryseobacterium balustinum and Chryseobacterium scophthalmum, respectively. Strain LMG 23089(T) and the C. balustinum and C. scophthalmum type strains formed a stable lineage supported by a bootstrap value of 100 %. The levels of DNA-DNA hybridization towards these nearest phylogenetic neighbours were below 57 %. The absence of growth on MacConkey agar or at 37 degrees C (on nutrient agar), the capacity to grow in the presence of 5 % NaCl and the production of urease activity differentiate this novel taxon from C. balustinum and C. scophthalmum. The four isolates are formally classified as Chryseobacterium piscium sp. nov., with strain LMG 23089(T) (=CCUG 51923(T)) as the type strain. Its DNA G + C content is 33.6 mol%.

A Gram-negative, microaerophilic helical rod, isolated from the gastric mucosa of a dog and designated strain JKM4(T), was subjected to a polyphasic taxonomic study. The tightly coiled organism, measuring 10-18 mum long and up to 1 mum wide, was motile by means of multiple sheathed flagella located at both ends of the cell and by a periplasmic fibril running along the external side of the helix. Strain JKM4(T) grew preferably on biphasic culture plates or on very moist agar. Coccoid forms predominated in cultures older than 4 days as well as in growth obtained on dry agar plates. The strain grew at 30 and 37 degrees C, but not at 25 or 42 degrees C and exhibited urease, oxidase and catalase activities. On the basis of 16S rRNA gene sequence analysis, the novel isolate was identified as a member of the genus Helicobacter and showed > 97 % similarity to Helicobacter felis, Helicobacter bizzozeronii and Helicobacter salomonis, three species previously isolated from the canine gastric mucosa. Protein profiling of strain JKM4(T) using SDS-PAGE revealed a pattern different from those of other Helicobacter species of mammalian gastric origin and from Helicobacter canis. Additionally, the urease gene sequence of strain JKM4(T) was different from those of urease genes of H. felis, H. bizzozeronii, H. salomonis and "Candidatus Helicobacter heilmannii". It is thus proposed that strain JKM4(T) (=LMG 23188(T)) represents a novel species within this genus, Helicobacter cynogastricus sp. nov.

Forty-five mycobacterial strains isolated from 23 Colombian HIV-positive patients were identified as members of the Mycobacterium avium complex (MAC) and were characterized using different molecular approaches. Seven of the isolates showed characteristic features that allowed them to be differentiated from other members of the complex. The isolates had a novel 16S-23S rRNA internal transcribed spacer (ITS 1) gene sequence which is described as a new sequevar, MAC-X. All of the seven novel isolates gave a positive result with the MAC-specific AccuProbe (Gen-Probe), but tested negative for Mycobacterium avium and Mycobacterium intracellulare species-specific probes (64 and 100 % of the isolates, respectively). The novel isolates could be differentiated phenotypically from other members of the MAC on the basis of the production of urease and by a consistent mycolic acid pattern. The novel isolates shared some characteristics with M. avium, such as the avium variant I (av-I) pattern of the hsp65 gene as determined by PCR restriction analysis and a positive PCR result for the mig (macrophage-induced) gene. However, the novel isolates showed a unique 16S rRNA gene sequence. DNA-DNA relatedness values, from 24 to 44 %, confirmed the distinction of the novel isolates from other members of the MAC at the genetic level and their status as members of a separate species. The novel isolates are proposed as representatives of a novel species, Mycobacterium colombiense sp. nov., that is closely related to M. avium within the MAC. The type strain is 10B(T) (=CIP 108962(T)=CECT 3035(T)).

Two previously undescribed anamorphic yeasts, strains T-11(T) and T-26(T), recovered from wild rabbit faecal pellets collected in Muju, Korea, were identified using phenotypic and molecular taxonomic methods. The isolates were characterized by the proliferation of budding cells, positive diazonium blue B and urease reactions, the presence of Q-10 as the major ubiquinone, the presence of xylose in whole-cell hydrolysates and the inability to ferment sugars. Phylogenetic analyses based on 26S rRNA gene partial sequences revealed that strain T-11(T) was located in the Bulleromyces clade and was related to Sirobasidium intermedium, Tremella exigua, Cryptococcus cellulolyticus and Bullera pseudoalba. Strain T-26(T) was located in the Mesenterica clade and was closely related to Cryptococcus sp. F6 and Cryptococcus heveanensis CBS 8976. Sequence divergence values of more than 4 % from other described Cryptococcus species, together with the phenotypic differences, showed that the isolated yeasts represent previously unrecognized members of this genus. Therefore, two novel yeast species are proposed: Cryptococcus mujuensis sp. nov., with strain T-11(T) (=KCTC 17231(T)=CBS 10308(T)) as the type strain, and Cryptococcus cuniculi sp. nov., with strain T-26(T) (=KCTC 17232(T)=CBS 10309(T)) as the type strain.
