-31 deletion products of the DNA fragment containing the vnfH promoter and upstream region, when cloned in a transcriptional fusion vector and analyzed for vnfH expression in Azotobacter vinelandii, revealed that the upstream activator sequence of the vnfH promoter lies about 140 nucleotides upstream of the promoter. Subsequent substitution and deletion analysis by oligonucleotide-directed mutagenesis in the upstream region of the vnfH promoter showed that sequences 5'-GTACCATGCGGAAC-3' and 5'-GTACCTGCGGGTAC-3', located 170 and 140 nucleotides upstream of the vnfH promoter, respectively, are both required for vnfH expression. Addition of four nucleotides in the intervening sequence between the vnfH promoter and the putative VnfA (analog of NifA of the conventional
molybdenum
-dependent nitrogen-fixation pathway) binding site resulted in a drastic reduction of expression from the vnfH promoter in Azotobacter vinelandii, whereas addition of 10 nucleotides in the intervening sequence did not affect the expression. Therefore, the face of the helix-dependent contact appeared to be important. DNA bending seemed to play a crucial role in expression from vnfH promoter. The intervening sequence exhibited characteristics of sequence-dependent intrinsically curved DNA, as shown by anomalous low gel mobility with polyacrylamide gel electrophoresis, electron microscopy, and computer simulated curvature analysis. Distamycin at very low concentrations significantly reduced the anomaly in electrophoretic mobility of the intervening DNA sequence.

-containing hydroxylase that produces superoxide and uric acid from purine substrates and molecular oxygen, is involved in the oxidative stress underlying several human pathologies including lung diseases. An enzymatic activity similar to xanthine oxidase was previously reported in bronchoalveolar lavage fluid of patients with chronic obstructive pulmonary disease (COPD-
BAL
), by fluorometric analysis of DNA unwinding and cytochrome c reduction kinetics. Here we report the detection of xanthine oxidase activity products by electron paramagnetic resonance (EPR) in presence of the spin-trap 5,5-dimethyl-1-pyrroline N-oxide (DMPO) and reversed-phase high-performance liquid chromatography (RP-HPLC) in COPD-
BAL
(n = 14, average age of patients 65 years, range 38-81) and
BAL
from healthy nonsmoker controls (n = 6, average age 64 years, range 44-73). Superoxide DMPO adducts were detected in COPD-
BAL
and in an in vitro system containing xanthine and xanthine oxidase (XA/XO), but not in
BAL
controls and when superoxide dismutase (SOD, 1000 I.U./ml) was added to COPD-
BAL
. The HPLC analyses after addition of xanthine showed production of uric acid in COPD-
BAL
and in the XA/XO system but not in
BAL
controls. These results support the involvement of xanthine oxidase in the mechanisms of superoxide production by
BAL
supernatant, which increases oxidative stress in chronic obstructive pulmonary disease.

This article outlines symptoms and therapeutic options in chronic cobalt poisoning including a case report in which metallosis caused by a ceramic-metal articular pairing led to almost complete loss of sight and hearing after revision of a total hip prosthesis. At primary revision the firmly incorporated stem was left in place. For a better offset only the head was exchanged from a ceramic to a metal model which articulated with a socket containing a ceramic inlay. Postoperatively, movement of the hip joint became increasingly uncomfortable and painful. After 2 years, the patient started complaining about increasing impairment of his eyesight followed by a gradual loss of hearing. In a second revision, examination of the explanted material showed almost complete deterioration of the metal femoral head and a partially fractured ceramic inlay with extensive contamination of the bone and surrounding soft tissue by metal debris. At the time of revision increasing concentrations of the alloy elements cobalt, chromium, and
molybdenum
were measured in the serum and liquor. The concentration of cobalt, in particular, was remarkably high. Treatment options in cases with chronic cobalt poisoning include chelation therapy with EDTA or
BAL
/DMPS.
