pH, Pco2, and Po2 measurements have been made over a three-week period on General Diagnostics Blood GAS Control Ampoules and on a tonometered buffer system. The latter seems adequate as a precision and accuracy control of pH, Pco2, and Po2 measurements in a single solution. The former seems adequate as a precision control of these measurements, but, on the particular batch provided, mean values need to be determined within the customer's laboratory.

The 36 basic "sentences" in the experiment were six-syllable nonsense sequences of the form DAS a LAS a GAS a or a DAS a BAS a LAS. Either (a) one vowel in the sentence was lengthened or shortened by about 50, 90, or 130 ms by computer-editing routines, or (b) the sentence was left intact (as spoken). The resulting perceptual impression after the vowel change was a change of tempo within the sentence. Vowel changes occurred systematically throughout the sentences, in one of syllables one through five. Reaction time (RT) was recorded to assigned target segments /b, d, or g/ in one of syllables one through six, and RT was compared to targets in tempo-changed versus intact sentences (these were acoustically identical except for the distorted vowel). The listeners responded to over 2000 versions of the sentences. The results were: (a) Tempo changes generally increased segment target RT. (b) Tempo-change effects were ubiquitous; for instance, vowel changes in the first syllable increased RT to targets in later syllables, and the reach of effects spanned four syllables. Both vowel shortening and lengthening increased target RT. (c) Effects attributed to precessing time decreased, whereas effects attributed to stimulus expectancy increased, with time into the sentence. (d) Tempo-change effects persisted throughout the experiment despite practice and familiarity with stimuli. The conclusions were: (a) The effects of time distortion of the stimulus on target RT were produced mainly by changes in stimulus-induced expectancy, not changes in processing time. (b) The expected input to perception is the acoustically intact utterance in both its rhythmic and segmental aspects; these aspects are not perceived independently.

Closing volume (CV) was ?EASURED WITH THE RESIDENT GAS TECHNIQUE IN 12 HEALTHY SEATED SUBJECTS AGE 22-70 YR, AND IN 8 SUBJECTS WITH THE BOLUS TECHNIQUE. Various volumes were inspired (Vi range: 20-100% vital capacity) from residual volume and CV was assessed on the subsequent recording of expired volume versus gas concentration. The results indicate that the resident gas technique may erroneously underestimate CV at reduced Vi in conformity with calculations which predict that during expiration, after a certain reduced VI, the nitrogen concentration is identical in the most basal lung region and at the mouth. CV obtained with the bolus technique decreased linearly with reduced Vi and the effect appeared to be age dependent according to the equation CV50/CV100=0.0078 X age +1.18, where CV50 and CV100 denote the bolus CV corresponding to Vi=50% and 100% of vital capacity. Therefore, in older subjects, during tidal breathing, airways appear to close at substantially lower lung volume than previously considered.

A number of studies on throat carriage of beta hemolytic streptococci (BHS) carried out during the years 1972-90 in urban and rural school children from low socioeconomic groups in the age group of 5-15 years in and around Delhi showed an overall carriage rate of BHS varying from 12.2%-64.3% depending upon the season and number of swabs taken. Group A was found to be the most predominant serological group (31.1%-62.6%). The T-typability was found to be 98.2%. The most prevalent T-patterns observed during 1972-78 study were 3/13/B3264 followed by 5/11/12/27/44. A significant difference was observed in the prevalence of T-patterns during the study of 2,034 children from 1979-83 and 3,094 children from 1984-90. When the most prevalent T-patterns were found to be 5/11/12/27/44 followed by 3/13/B3264. The study of the school children from 1972-90 showed the isolation of BHS as well as significant predominance of GAS (p < 0.001) in winter months than summer months. There was no difference in the distribution of carriage of BHS and GAS amongst rural or urban school children. Since RF/RHD are illnesses which were often encountered in school children among socially and economically disadvantaged populations stronger support for streptococcal surveillance programs should be encouraged.

The gastrin gene is expressed in fetal pancreatic islet cells, but in the adult is expressed mainly in the gastric antrum. To study the regulation of the gastrin promoter, we created several transgenes containing the human and rat gastrin 5' flanking regions joined to the coding sequences of the human gastrin gene. The human gastrin transgene contained 1,300 bp of 5' flanking DNA, while the rat gastrin transgene contained 450 bp of 5' flanking DNA. The human gastrin transgene was expressed in fetal islets, but was not expressed in adult gastric antrum. In contrast, the rat gastrin transgene was expressed in adult antral G cells, but no expression was observed in fetal islets. To study the possible role of gastrin as an islet growth factor, a chimeric insulin-gastrin (INS-GAS) transgene was created, in which the expression of the human gastrin gene is driven from the rat insulin I promoter. These INS-GAS mice were mated with mice overexpressing TGF alpha, transcribed from a mouse metallothionein-transforming growth factor alpha (MT-TGF alpha) transgene. While overexpression of gastrin or TGF alpha alone had no effect on islet mass, overexpression of both transgenes resulted in a twofold increase in islet mass. In conclusion, these data indicate that (1) gastrin can interact synergistically with TGF alpha to stimulate islet growth; (2) the human gastrin transgene contains the islet specific enhancer; (3) the rat gastrin transgene contains the antral specific enhancer.

Streptococcus pyogenes (group A streptococci; GAS) expresses important virulence factors like the antiphagocytic M protein, the complement factor-inactivating C5a peptidase and the immunoglobulin-Fc-binding proteins on its surface. The corresponding emm, scpA, and emm-related (fcrA, ennX) genes are adjacently encoded on the genome. They are coordinately in trans regulated by the positive regulatory VirR factor. The responsible virR gene is also located within this segment of the genome which was called vir-regulon. There are at least three different types of organization of the vir-regulon. A frequently encountered type is the "Large vir-regulon". It comprises from 5' to 3' the following genes: virR, fcrA, a relatively small emm, ennX, and a 4.6 kb version of scpA. Another common type is the "Small vir-regulon", which contains a virR deviating in its 3'-region, a relatively large emm, and a 3.5 kb version of scpA. The "Unusual vir-regulon" is less frequently detected. It closely resembles the small one, but harbors an additional 3 to 4 kb DNA fragment between emm and scpA, occasionally encoding an emm-related gene. The type of vir-regulon encoded by a GAS strain correlates to its serotype, its M class, and its expression of serum opacity factor. The structural genes of the vir-regulon are expressed at a high level during growth in exponential phase, under anaerobiosis, and at body temperature. The sensor molecule which modulates VirR activity according to these environmental conditions has not yet been detected.(ABSTRACT TRUNCATED AT 250 WORDS)

The 9/27 and GBP mRNAs are both inducible by Interferon-gamma (IFN-gamma). The promoters of both genes contain an Interferon Stimulation Response Element (ISRE), but while the GBP gene is strongly induced transcriptionally by IFN-gamma the response of the 9/27 promoter is very weak. We investigated the molecular basis for this difference. The different IFN-gamma-responsiveness was found to have more than one reason. First, 9/27 promoter DNA was unable to bind the Gamma Interferon Activation Factor (GAF) with a single high affinity site. It efficiently competed for the association of the GAF with the GBP promoter but this competition was due to the presence of two low affinity sites, the ISRE and an ISRE-like sequence, suggesting that the GAS and ISRE, though both having clear preferences for specific proteins, may nevertheless share a certain degree of structural homology. Second, the 9/27 and GBP ISREs differed markedly in their affinities for regulatory proteins (ISGFs 1,2,3) and the GBP ISRE was more potent in mediating IFN-gamma-induced promoter activity in transient transfection. Third and most importantly, however, the strong difference between the IFN-gamma response of the two promoters was mainly due to the sequences surrounding the ISRE: the positive-acting GAS on one side and sequences with silencing properties 5' and 3' of the 9/27 ISRE on the other side. The data thus show mechanisms to both up- and down-regulate the activity of the ISRE.

A controlled clinical study on the outcome of family intervention in chronic schizophrenics is being carried out in Athens. All subjects participating in the study reside with their families and attend a vocational rehabilitation unit. Those belonging to families characterized by high "Expressed Emotion" (EE) are randomly assigned to either the experimental or the control condition. The former receive family intervention combined with individual treatment, the latter individual treatment alone. 75 subjects have entered the study, and 121 relatives were interviewed with the Camberwell Family Interview. 40 patients (53.3%) were found to belong to high EE families. The present article presents the results of the baseline assessment. It was found that the EE status of the family was significantly associated with measures of psychopathology and social functioning (BPRS, GAS, DAS, number of residual symptoms). Further analyses revealed that these associations exist only when all key relatives express high EE. The implications of these findings are discussed.

Different expert-rating scales (GAS, BPRS, SANS, Ham-D) were applied to assess the psychopathological findings in 40 schizophrenic and 40 depressive patients (classified according to DSM III-R guidelines). The data demonstrated that the schizophrenic sample did not display acute psychotic or negative symptoms as a specific symptomatology, and that depressive mood was not a specific symptom of the depressive sample. Hence, these types of research tools should mainly be used as a global measurement for determining the severity of psychiatric disorders, or for monitoring the progress of individual patients in order to illustrate disparate psychopathological qualities in nosologically different groups.

Induction of anti-IgG during hyperimmunization of rabbit with Streptococcus pyogenes (group A streptococci; GAS) was previously shown to require the presence of IgG Fc receptors (FcR) in the vaccine strain. In the present work, we examined whether streptococcal FcR activity might also be of importance for heart and kidney deposition of IgG, known to occur in poststreptococcal sequelae as well as during experimental immunization of animals. Each of three IgG-binding (GAS types M1, M12 and M22) and two non-binding (GAS type T27 and S. agalactiae (GBS) type Ia) streptococcal strains were used for intravenous immunization of rabbits during two periods of eight and six weeks, respectively, separated by an interval of one month. Before use, vaccine strains were treated with KSCN and carefully washed in order to remove any surface-bound immunoglobulins. No deaths occurred among injected rabbits. No tissue deposition was elicited by the GAS type T27 or the GBS strain. In contrast, the strains of types M1, M12 and M22 all induced deposits of IgG in kidney and heart tissue, beginning during the first immunization period. In two tested animals, receiving GAS of types M1 or M22, circulating immune complexes containing anti-IgG antibodies were also detected. Finally, serum autoantibodies reacting with preparations of heart and kidney, but not lung or liver, were demonstrated in each of six animals receiving M1 or M22, reaching maximum levels during reimmunization; such antibodies were not evoked by the two strains not binding IgG. Our results suggest that, in GAS with capacity for non-immune binding of IgG, triggering of anti-IgG acted to enhance tissue deposition of IgG or immune complexes in immunized rabbits. Furthermore tissue-specific antibodies were elicited only by the IgG-binding strains and occurred comparatively late during immunization, suggesting that those antibodies might have been triggered due to the exposition of hidden kidney and heart determinants.
