This paper is concerned with optimization of the fermentation conditions for preparation of alkaline pectin lyase (poly-galacturonic acid trans-eliminase, PATE) by Erwinia carotovora IFO3830 (E.C.IFO3830). The orthogonal matrix method was adopted as the experimental design method for media. The effects of media composition on the growth rate of E.C.IFO3830 are in the order of pectin > KH(2)PO(4)/Na(2)HPO(4) > MgSO(4) > (NH(4))(2)SO(4) > glucose > L-glutamate sodium, and those on PATE activity are in the order of glucose > pectin > L-glutamate sodium > KH(2)PO(4)/Na(2)HPO(4) > MgSO(4) > (NH(4))(2)SO(4). The strain of E.C.IFO3830 grows quickly and easily near an initial pH of 7.0, and especially at the alkaline range of pH 7.0-8.0. The growth rate profiles indicate that E.C.IFO3830 grows very quickly; its generation time t(1/2) is about 0.2 h. The appropriate fermentation period is about 10-20 h for a high level productivity of biomass and 25-35 h for high productivity of PATE enzyme.

Purinergic signaling represents a major non-synaptic signaling mechanism in the normal and pathological nervous system. The expression of the purinergic ligand gated ion channel P2X7 receptor (P2rx7) has been described on nerve terminals as well as in non-neuronal cells, such as astrocytes and microglia. The activation of P2rx7s results in Ca(2+) influx and increased transmitter release in the brain. P2rx7s previously suggested having a pivotal role in different pain modalities, including neuropathic pain. Here we investigated whether the activation of P2rx7 leads to increased glutamate release from the spinal cord in an experimental model of neuropathic pain (partial nerve ligation of the sciatic nerve, PNL). One week after surgery, we studied the effects of PNL on tactile allodynia using aesthesiometry, in parallel with the in vitro release of [(3)H]glutamate from lumbar spinal cord slices. The observed allodynia in wild-type (P2rx7+/+) mice one week after PNL surgery was lower that was observed in P2rx7 deficient (P2rx7-/-) animals. Perfusion of spinal cord slices with ATP (10mM) elicited [(3)H]glutamate release in both sham operated and neuropathic P2rx7+/+ animals. The ATP-induced [(3)H]glutamate release was absent in P2rx7-/- mice. Electrically evoked release of [(3)H]glutamate from spinal cord slices was not significantly altered in PNL animals and in P2rx7-/- mice. The results suggest that activation of P2rx7 by ATP releases glutamate in the spinal cord, which might contribute to mechanical allodynia following PNL. On the other hand, this release does not contribute to glutamate efflux evoked by conventional neuronal activity, which is consistent with the idea that P2X7 receptors are either extrasynaptic or expressed on non-neuronal cells. We characterized two newly isolated myoviruses, Bp8p-C and Bp8p-T, infecting the ginger rhizome rot disease pathogen Bacillus pumilus GR8. The plaque of Bp8p-T exhibited a clear center with a turbid rim, suggesting that Bp8p-T could transform into latent phage. Lysogeny assays showed that both the two phages could form latent states, while Bp8p-T could form latent phage at a higher frequency and stability than Bp8p-C. The genomes of Bp8p-C and Bp8p-T were 151,417 and 151,419 bp, respectively; both encoded 212 putative proteins, and only differed by three nucleotides. Moreover, owing to this difference, Bp8p-C encoded a truncated, putative actin-like plasmid segregation protein Gp27-C. Functional analysis of protein Gp27 showed that Gp27-T encoded by Bp8p-T exhibited higher ATPase activity and assembly ability than Gp27-C. The results indicate that the difference in Gp27 affected the phage lysogenic ability. Structural proteome analysis of Bp8p-C virion resulted in the identification of 14 structural proteins, among which a pectin lyase-like protein, a putative poly-gamma-glutamate hydrolase, and three proteins with unknown function, were firstly identified as components of the phage virion. Both phages exhibited specific lytic ability to the host strain GR8. Bp8p-C showed better control effect on the pathogen in ginger rhizome slices than Bp8p-T, suggesting that Bp8p-C has a potential application in bio-control of ginger rhizome rot disease.
