Twenty Thoroughbred 3 year old horses (10 stallions and 10 mares), trained and raced at the Warsaw Race-Course were studied from March through November. Blood was taken approximately every 8 weeks to determine the activities of aspartate and alanine transaminases, acid and alkaline phosphatases and
aldolase
. It was observed that the activities of aspartate aminotransferase and
alkaline phosphatase
reached their maxima in July and alanine transaminase in May. The activities of acid phosphatase and
aldolase
showed their minima in July. Comparing these data with the literature it was noted that the changes observed are mainly seasonally-dependent; but, training had some influence on the activity of the enzymes involved in energy metabolism.

Isozyme patterns of 23 different enzymes were compared in normal, benign, and malignant breast tissues; in MCF-7 cells; and in organoids of normal human breast tissue. Benign lesions generally showed isozyme patterns similar to those of normal tissues. Lactate dehydrogenase isozyme 5 was significantly increased in malignant tumors; MCF-7 cells had only lactate dehydrogenase (L-lactate:NAD oxidoreductase; EC 1.1.1.27). The mitochondrial form of malate dehydrogenase was also significantly increased in human malignant tumors; this was especially evident when comparing tumor and apparently uninvolved breast tissue from the same patient. The K4 isozyme of pyruvate kinase was the major form in most malignant breast tumors, but in only 41% of normal tissues, 30% of fibrocystic disease specimens, and 46% of fibroadenomas. A more anodal band of pyruvate kinase, probably a K3M or K3Kpm hybrid, predominated in most normal and benign tissues, but in only 63% of primary and 56% of secondary tumors. A4, and hexokinase I. Traces of
aldolase
A3C and of hexokinase II were observed in some tumors. None of the tumors had the Regan variant of
alkaline phosphatase
. The isozymes of lactate and malate dehydrogenases and of pyruvate kinase appear to be the most promising as putative tumor markers.

The ability of rat liver zinc-thionein to donate its metal to the apo-enzymes of the zinc enzymes horse liver alcohol dehydrogenase, yeast
aldolase
, thermolysin, Escherichia coli
alkaline phosphatase
and bovine erythrocyte carbonic anhydrase was investigated. Zinc-thionein was as good as, or better than, ZnSO(4), Zn(CH(3)CO(2))(2) or Zn(NO(3))(2) in donating its zinc to these apo-enzymes. Apo-(alcohol dehydrogenase) could not be reactivated by zinc salts or by zinc-thionein. Incubation of the other apo-enzymes with near-saturating amounts of zinc as ZnSO(4), Zn(CH(3)CO(2))(2), Zn(NO(3))(2), or zinc-thionein resulted in reactivation of the apo-enzymes. With apo-
aldolase
zinc-thionein gave 100% reactivation within 30min. Reactivation by ZnSO(4) and Zn(CH(3)CO(2))(2) was complete and instantaneous. Zinc-thionein was somewhat better than Zn(NO(3))(2) in completely reactivating apo-thermolysin. With apo-(
alkaline phosphatase
) 43% reactivation was obtained with Zn(CH(3)CO(2))(2) and 18% with zinc-thionein. With apo-(carbonic anhydrase) zinc-thionein was better than ZnSO(4), Zn(CH(3)CO(2))(2) or Zn(NO(3))(2), with a maximal reactivation of 54%. That zinc was really being transferred from zinc-thionein to apo-(carbonic anhydrase) was shown by the fact that 2,6-pyridine dicarboxylic acid and 1,10-phenanthroline had minimal effects on the reactivation of apo-(carbonic anhydrase) when added after the incubation {[apo-(carbonic anhydrase)+zinc thionein]+chelator}, but inhibited reactivation when added before the incubation {apo-(carbonic anhydrase)+[zinc-thionein+chelator]}. These observations support the idea that zinc-thionein can function in zinc homeostasis as a reservoir of zinc, releasing the metal to zinc-requiring metalloenzymes according to need.

A comparison of several serum tumor markers (lactate dehydrogenase (LDH), LDH isozyme, hydroxybutylrate dehydrogenase (HBD),
alkaline phosphatase
(
ALP
), cholinesterase (Choline-E),
aldolase
(
ALD
), leucine aminopeptidase (LAP), gamma-glutamyl transpeptidase (gamma-GTP), human chorionic gonadotropin (HCG), carcinoembryonic antigen (CEA) and alpha 1-fetoprotein (AFP)) was made in patients with carcinoma or benign tumor of the ovary and healthy control subjects. The greatest positive rates were obtained with the markers HBD (76.5%) and Choline-E (73.3%) for patients with carcinoma of the ovary, respectively. However, based on false positive results, Choline-E was also greatest (50.0%) for patients with benign tumor of the ovary. The lowest false positive rates were obtained with
ALD
, but the positive rates for patients with stage I and II diseases were 0.0%. The most suitable single marker for patients with stage I and II diseases was HBD (62.5%), followed by LDH (41.7%). Three of 4 patients with early cancer, who had normal serum LDH levels, showed abnormal LDH isozyme patterns (elevated LDH-4 and -5). A combination of LDH activity and LDH isozyme resulted in an increase in the positive results (41.7% to 70.0%), that is, the cancer patients were positive for one of the two markers. For CEA, AFP and HCG, the positive results were 26.9%, 19.0% and 7.1%, respectively. Positive and false positive rates for
ALP
were 36.7% and 7.1%, but the positive rates in the early stage were lower (14.3%), compared to those for LDH and HBD. HBD and LDH activities in the ovarian malignant tissues and ascitic fluids were significantly higher than those in the benign tumor tissues and ascitic fluids, resulting in a significant elevation of serum LDH and HBD levels in the patients. Moreover, it was suggested that inhibition test of
ALP
by the inhibitors might be able to identify the tissue origin of
ALP
in the cancer patients.

(AlkP), pseudocholinesterase (PsCHE) isocitrate dehydrogenase and gamma-glutamyltranspeptidase (gamma-GTP), was determined in 256 patients with progressing myodystrophy (PMD) (Duchenne's form in 125, Becker's form in 14, pelvicohumeral form in 36, humeroscapulofacial form in 19, ocular form in 10, other rare forms in 34, and nonidentified forms in 13 patients). In the control group (64 men, 56 women and 50 children), the activity of the enzymes was found to depend on the patients' sex and age. With regard to both parameters, i. e. the degree of the enzyme activity rise and the frequency of the pathological values the most informative were CK, then PK and

Dynamic examinations of the activity of glutamate-aspartate and glutamate-alanine aminotransferases (AST, ALT),
fructose diphosphate aldolase
and
alkaline phosphatase
in the cerebrospinal fluid (CSF) were carried out in 512 patients (14 groups) suffering from viral and bacterial meningitis in the acute period, as well as in reconvalescents. The activity of the CSF enzymes was also determined in 70 healthy subjects. It was found that in the acute period of meningitis the activity of the CSF enzymes (mostly of the aminotransferases) rose, this rise being greater in meningococcal and tuberculous meningitis than in the viral one. In reconvalescents the activity of the aminotransferases dropped, and that of
aldolase
and
alkaline phosphatase
got normal. The activity of the blood serum enzymes showed no substantial changes. The differences in the activity of the enzymes may serve as a criterion for diagnostic differentiation of meningitis.

The pathology and enzymology of the intestinal mucosae of lambs dosed daily with 2500 Trichostrongylus vitrinus larvae and killed at five, nine or 14 weeks were compared with worm-free animals. , leucine amino-peptidase, maltase and glycyl-L-leucine dipeptidase were all significantly depleted during infection, although the magnitude, time of onset and duration of the individual enzyme responses varied. Mucosal activities of the pancreatic enzymes, trypsin and to a lesser extent chymotrypsin were also markedly decreased particularly during the first nine weeks of infection. Specific acetylcholinesterase activity was significantly increased throughout the study, maximal levels being observed at five weeks. In contrast 'pseudo'-cholinesterase levels were consistently within the control range. ), creatine phosphokinase (CPK), guanase, and cholinesterase (CE). The calculation of the correlation coefficients and the regression lines revealed that GOT, GPT, LDH,

, and guanase raised their activity following treatment of the test animals up to a higher extent and at a higher rate. Lower rate changes were those in the activity of AP, leucinaminopeptidase, and CE. No changes whatever were found in the activity of CPK. On the basis of these results the diagnostic value is determined of both the individual enzymes and the enzyme constellation as a whole.
