In 38 patients with chronic renal insufficiency of different degree of severity examinations of the stationary concentration of the adenine nucleotides in the erythrocytes were carried out. It was shown that in the red blood cells of uraemics a genuine increase of the concentration of these compounds occurs, in which case the adenosine triphosphate dominates absolutely as well as relatively. In individual cases erytho-cyctic ATP-values of more than 3 micron mol pro ml cells may be achieved. The increase of the ATP-concentration in the red blood cells correlates with the degree of severity of the renal insufficiency and the renal anaemia. The hyperphosphataemia occurring as a rule in renal insuficiency is of causal importance for the increase of ATP. By a consecutive increase of the intracellular phosphate level and by influence on different steps of enzymes (phosphofructokinase, aldolase, glycerin aldehyde phosphate dehydrogenase) and changed regulations it effected an activation of the glycolysis. The increase of the plasma adenine and plasma adenosine concentration plays apparantly an accessory role for the increase of the concentration of the adenine nucleotides existing in the erythrocytes. Together with an increased concentration of 2,3-diphosphogycerate (2,3-DPG) the increase of the ATP-level has an effect on the oxygen transport function function of haemoglobin in the sense of a facilitation of the O2-output. These processes explain the relative adaption of patients with chronic renal insufficiency to renal anaemias of partly high degree.

Exposure of foundry workers to mixtures of different heavy metals is a very important toxicological problem. In this paper the estimation of the effects of lead, zinc, and copper on erythrocyte metabolism is presented. Concentrations of copper and zinc at work posts of the group examined did not exceed TLV, while lead concentration was 1.5 to 4 times higher than TLV. Erythrocyte metabolism was measured through activities of such glycolytic pathway enzymes as PFK, PGI, PK, aldolase and G6-PD from the hexose monophosphate pathway. Additionally the free erythrocyte protoporphyrin (FEP) level, D-ALA activity, serum GSH level, 2,3 DPG level in erythrocytes and lactic acid production during a 2-h incubation of red blood cells (RBC) was estimated. The blood-lead level, FEP level, copper concentration in erythrocytes in exposed group were significantly higher than in control group while the zinc level in erythrocytes was significantly lower. Measuring erythrocyte metabolism we showed that the activity of PGI, PFK, aldolase, lactic production and 2,3 DPG levels was significantly higher in the exposed group, probably as a result of anaerobic glycolysis activation.

Red cell enzymes, 2,3-diphosphoglycerate (2,3-DPG) and adenosine triphosphate (ATP), were evaluated in a 23-mo-old boy with juvenile chronic myelocytic leukemia (JCML) at the onset of his illness and 6 mo later during the accelerated phase. The activities of the age-dependent red cell enzymes, hexokinase, aldolase, pyruvate kinase, and glucose-6-phosphate dehydrogenase, were elevated, as were the concentrations of red cell 2,3-DPG and ATP, consistent with a young red cell population metabolizing at an increased glycolytic rate. The activities of the non-age-dependent enzymes, glyceraldehyde-3-phosphate dehydrogenase (G3PD), phosphoglycerate kinase, and enolase, were also increased to levels similar to or greater than those observed in term infants. As the illness progressed, the activity of red cell G3PD increased further, and phosphoglucose isomerase activity increased markedly. These results are consistent with the prior suggestion that JCML represents a reversion to "fetal" erythropoiesis.

In investigating the influence of vibrational energy on the metabolism of the erythrocyte, it was hypothesized that under conditions of normal PaO2 and SaO2 in arterial blood, vibration induced vasoconstriction would decrease local blood flow and induce hypokinetic hypoxia. This decreased blood flow and therefore decreased delivery of oxygen to the tissue would markedly lower tissue PO2 (hypokinetic hypoxia), which would influence the energetics and metabolism of the erythrocyte. The metabolism of the red blood cell (RBC) was evaluated by measuring the enzymatic activities of PFK (2.7.1.11), PGI (5.3.1.9), PK (2.7.1.40), and aldolase (4.1.3.13) from the anaerobic glycolytic cycle and D-G-6-P (1.1.1.49) from the pentose cycle. Also measured were the levels of ATP and 2,3 DPG and the in-vitro production of lactic acid. In the group of workers showing early changes (vibration angioneurosis) associated with the vibration syndrome, changes in RBC metabolism were demonstrated. Statistically significant were increases of PFK, PK and the production of lactic acid, indicating the activation of anaerobic glycolysis. Furthermore statistically significant were the increased 2,3 DPG and decreased ATP levels.

A simple screening procedure for the detection of glucose-phosphate isomerase (GPI), phosphofructokinase (PFK), aldolase (AL) and glyceraldehyde-3-phosphate dehydrogenase (GAPD) deficiencies in blood, is described. These enzymes catalyze the second, third, fourth, and sixth reactions in the Embden-Meyerhof pathway. The procedure is based on the conversion of glucose-6-phosphate to 1,3-diphosphoglycerate (1,3-DPG) which is catalyzed by the sequential action of the GPI, PFK, AL and GAPD. The presence of the enzyme activities is visually estimated by the reduction of NAD+ (non-fluorescent) to NADH (fluorescent) which occurs when 1,3-DPG is formed. Absence of fluorescence indicates the deficiency of anyone of the four enzymes, which are specified by using separately the PFK, AL and GAPD respective substrates.

The investigations were carried out on 20 men (aged 32-48), hospitalized after lower extremity fractures, in whom no systemic diseases or disorders were found. The activity of glucose-6-phosphate dehydrogenase (G6P-DH), aldolase (A1), lactate dehydrogenase (LDH) and the concentrations of pyruvate, lactate, nicotinamide-adenine dinucleotide (NAD), nicotinamideadenine dinucleotide phosphate (NADP), adenosine-5'-triphosphate (ATP), adenosine-5'-diphosphate (ADP), adenosine -5'-monophosphate (AMP) and 2,3-diphosphoglycerate (2,3-DPG) in erythrocytes taken from patients were estimated. After a two-week hypokinesia G6P-DH, A1, LDH activities and ATP, ADP, AMP concentrations decreased, whereas other estimated parameters did not differ from the control levels. During a 30-day hypokinesia of patients with fractures of the lower extremity a transient decrease of erythrocyte metabolic activity was observed after two weeks. off

In red blood cells, a modulation of the level of the allosteric effector of hemoglobin, 2,3-diphosphoglycerate (2,3-DPG) would have implications in the treatment of ischemia and sickle cell anemia. Its concentration is determined by the relative activities of the synthase and phosphatase reactions of the multifunctional bisphosphoglycerate mutase (BPGM). In this report we develop first a more direct synthase assay which uses glyceraldehyde phosphate to suppress the aldolase and triose phosphate isomerase reactions. Secondly we propose a radioactive phosphatase assay coupled to chromatographic separation and identification of the reaction products by paper electrophoresis. Such identification of these products allow us to show that the multifunctional BPGM expresses its mutase instead of its phosphatase activity in conditions of competition between the 3-phosphoglycerate and the 2-phosphoglycolate activator in the phosphatase reaction. These two more precise procedures could be used to study the effects of substrate and cofactor analogues regarding potential therapeutic approaches and could be used for clinical analyses to detect deficiency of BPGM.

Ergometrics, using both treadmill (T) and bicycle-ergometer (B), plays an important role in assessing physical work capacity. In addition to the other important parameters (blood pressure, heart rate, oxygen consumption, oxygen and carbon dioxyde-partial pressure in blood, etc.), it is of interest to follow, measure and compare values of certain metabolic products immediately after completion of exercise on T and B. The aim of this study was to define which test develops the most significant changes in metabolic parameters when the same workload intensity is maintained. The metabolic parameters measured were: lactate, pyruvate, 2.3-diphosphoglycerate (2.3-DPG), lactate dehydrogenase (LDH) and aldolase activity. The mean values obtained at the end of both tests were: lactate 3.17 mmol/l +/- 1.017 mmol/l (T) and 4.28 mmol/l +/- 1.2 mmol/l (B); pyruvate 78.17 mumol/l +/- 30.32 mumol/l (T) and 92.16 mumol/l +/- 32.73 mumol/l (B); 2.3-DPG 3.58 mmol/l +/- 1.31 mmol/l (T) and 3.93 mmol/l +/- 1.44 mmol/l (B); LDH activity 239.22 U/I +/- 38.24 U/I (T) and 260.5 U/I +/- 61.27 U/I (B); aldolase activity 4.93 U/I +/- 1.45 U/I (T) and 5.88 U/I +/- 2.31 U/I (B). The results obtained indicate that a much higher production and/or lower elimination of acid metabolic products occurs during the test on B. This is most likely due to increased muscle pressure in big muscle groups in the legs which, by compressing the local blood vessels, diminish oxygen supply and the elimination of acid metabolic products. According to the results of the study, the use of T can be recommended as a more appropriate method for physical work capacity assessment of untrained individuals in occupational medicine.
