The contractile, histochemical, morphological and electrophysiological properties of two rat laryngeal muscles, the cricothyroid and posterior cricoarytenoid, have been measured. Both muscles act during respiration to maintain upper airway patency and an even distribution of air in the lungs. The large areas of T-tubule membrane are reflected in the average specific membrane capacities of 6.5 muF cm-2 to 10.5 muF cm-2, which are high considering the small diameter of the fibres (20-30 micron). Of the two muscles, the posterior cricoarytenoid has the faster contraction time and the more abundant sarcoplasmic reticulum content. In addition, the posterior cricoarytenoid is less resistant to fatigue and demonstrates lower
succinic dehydrogenase
activity. The fatigability of this muscle, coupled with its general lack of functional reserve, suggest that its failure may contribute to upper airway obstruction during respiratory distress.

Predominantly fast skeletal muscles of rabbits [tibialis anterior (TA), extensor digitorum longus (EDL)] were stimulated at a frequency naturally occurring in nerves to slow muscles (10 Hz continuously) for 8 h/day for 2--4 days. Such stimulation is known to convert all glycolytic fibers to oxidative and to increase capillary density. Our aim was to study early stages of conversion to investigate the factors responsible for the changes. Staining of quick-frozen sections for

, and alkaline phosphatase was used to study the distribution of different fiber types and to measure fiber cross-sectional areas, capillaries per square millimeter, and capillary-to-fiber ratios in each fiber category. TA but not EDL showed conversion of fast glycolytic to fast oxidative fibers after 2 days, more after 4 days of stimulation. In both muscles, the largest fast glycolytic fibers were diminished in number after stimulation. There was significant increase in total capillaries per square millimeter after 4 days and some increase after 2 days of stimulation. The increase in capillaries per square millimeter exceeded the increase in the number of fibers per square millimeter, and since there was no change in mean fiber area, the increase is attributed to capillary growth. In EDL, there was an increase in the number of capillaries supplying both fast glycolytic and fast oxidative fibers, suggesting that capillary growth precedes fiber type conversion. In TA, the number of capillaries supplying fast oxidative fibers was increased but that to fast glycolytic fibers, was not. This is consistent with capillary growth simultaneous with or following fiber conversion. In both TA and EDL the number of capillaries perfused after contraction was higher in stimulated muscles, suggesting that increased capillary flow contributed to capillary growth.

Myocardial preservation during open heart surgery in children was studied using biopsies of the right ventricle taken at the beginning and end of a bypass with a Tru-Cut biopsy needle. Three assessments were used: (1) semi-quantitative cytochemical grading of Baker's acid haematein reaction, and the distribution of
succinate dehydrogenase
and
myosin ATPase
; (2) microdensitometry of the
succinate dehydrogenase
activity; and (3) quantitative birefringence measurements to assess the response of the myocardial fibres to ATP and calcium. For each assessment, the values at the beginning and end of the bypass were compared. In a series of 42 children, two died in low cardiac output and three others required substantial inotropic support. No patient showed deterioration in the overall cytochemical assessment. Two patients showed deterioration in birefringence, one died and one had low cardiac output. The remaining three patients who had post-operative problems had low birefringence values at the beginning of bypass. Thirty-two patients were used for the microdensitometric assays, one died and three required substantial inotropic support. Succinate dehydrogenase activity decreased significantly during bypass in only two patients. One of these died and the other required substantial inotropic support.

, light; alpha GPDH, moderate to dark. The third class of fibers had the following histochemical characteristics: mATPase, light;

, moderate to dark; alpha GPDH, light. These fibers were considered to be either slow twitch, or tonic, and oxidative (S/O). Pyruvate kinase (PK), alpha GPDH, and citrate synthase (CS) activities were measured in homogenates of the same muscles studied histochemically. There was a positive relationship between both PK and alpha GPDH activities and the percentage of glycolytic fiber types within a muscle. Likewise, CS activities were greater in muscles high in FOG and S/O content. Based on CS activities, Varanus S/O fibers were eight-fold more oxidative than FG fibers within the same muscle. PK/CS ratios suggested that FG fibers possess high anaerobic capacity, similar to the iguanid lizard Dipsosaurus. The fiber type composition of the gastrocnemius muscle, relative to that of other lizard species, suggests that varanid lizards may possess a greater proportion of FOG and S/O fibers than other lizards.

The percentage of these types was determined in the musculus deltoideus, m triceps brachii caput longum, m gluteus medius, m semitendinosis, m biceps femoris and m vastus lateralis of the thoroughbred, Shetland pony, pony, heavy hunter and donkey. In addition the m gluteus medius was examined in the arab and American racing quarterhorse. High
myosin ATPase
activity fibres varied from a mean of 93.2 per cent in the m gluteus medius of the quarterhorse to 58.2 per cent in the m vastus lateralis of the donkey. In the m gluteus medius it was found that the percentage of high mycosin ATPase (pH 9.4) fibres varied significantly among breeds and these differences were related to the sprinting speed of the breed.

Male Wistar rats were made hyperthyroid by intraperitoneal injections on alternate days for 1-6 weeks, of 200 micrograms/kg triiodothyronine (T3). The effects of this treatment on the contractile properties of the soleus, a slow twitch and the extensor digitorum longus (EDL), a fast twitch skeletal muscle, were studied in vivo in the anaesthetized animal. Post mortem, serial frozen sections of both muscles were stained histochemically for
myosin ATPase
,
succinic dehydrogenase
and phosphorylase. Muscle fibres were classified as either slow twitch (SO), fast twitch oxidative glycolytic (FOG) or fast twitch glycolytic (FG). Elevation of plasma T3 levels is associated with progressive alterations in the muscle fibre populations of both muscles. In the soleus there is conversion of SO to FOG fibres while in the EDL the main effect is FG to FOG conversion. There are also marked changes, mainly confined to the soleus muscle, in contractile properties; progressive increases in isometric twitch and tetanic tension and in the rates of contraction and relaxation during both twitch and tetanus. The effect of T3 on slow muscle contractility may be related to its effect on muscle phenotype. However, changes in the former precede detectable fibre population changes. T3 may influence properties such as the CA2+ binding activity of sarcoplasmic reticulum of existing slow twitch fibres before the later changes associated with the interconversion of fibre types occur.

Lizard skeletal muscle fiber types were investigated in the iliofibularis (IF) muscle of the desert iguana (Dipsosaurus dorsalis). Three fiber types were identified based on histochemical staining for

), and alphaglycerophosphate dehydrogenase (alphaGPDH) activity. The pale region of the IF contains exclusively fast-twitch-glycolytic (FG) fibers, which stain dark for mATPase and alphaGPDH, light
SDH
. The red region of the IF contains fast-twitch-oxidative-glycolytic (FOG) fibers, which stain dark for all three enzymes, and tonic fibers, which stain light for mATPase, dark for
SDH
, and moderate for alphaGPDH. Enzymatic activities of myofibrillar ATPase, citrate synthase, and alphaGPDH confirm these histochemical interpretations. Lizard FG and FOG fibers possess twitch contraction times and resistance to fatigue comparable to analogous fibers in mammals, but are one-half as oxidative and several times as glycolytic as analogous fibers in rats. Lizard tonic fibers demonstrate the acetylcholine sensitivity common to other vertebrate tonic fibers.

In all muscles the greyhound had a significantly higher percentage of fibres with high

activity at pH 9.4 than the other breeds, with almost 100 per cent in most muscles examined. The activities of nine enzymes and glycogen concentration were determined in m gluteus medius and m semitendinosus of the greyhound and crossbred. Significantly higher levels of creatine kinase, aldolase, alanine aminotransferase and citrate synthase and significantly lower activities of 3-hydroxyacyl coenzyme A dehydrogenase and hexokinase were found in both muscles of the greyhound. The implications of these findings are discussed.

The histochemical and ultrastructural changes in the mature feline bulbocavernosus muscle (BCM) induced after 10 days of denervation or castration were compared. Control BCM, denervated BCM, and BCM from castrated cats reacted similarly for
myosin ATPase
(predominantly dark staining) and for
succinic dehydrogenase
(predominantly high). Staining for phosphorylase was decreased in the denervated BCM and BCM from castrated cats. Intrafiber lipid content was reduced after denervation or castration. The appearance of lysosome-like structures in BCM from both treatment groups was the most obvious ultrastructural alteration. Other ultrastructural changes were focal and infrequent.

Tenotomy of the rat soleus muscle is followed by a central degeneration of slow, fatigue-resistant muscle fibers. Previous experiments showed that fast, fatigable fibers of the gastrocnemius when transformed to slow, fatigue-resistant fibers by cross-reinnervation also develop lesions after tenotomy. The experiments described in this communication were carried out to discover whether the susceptibility of fibers to lesions was determined by their fiber type or the nature of their innervation. Rats were rendered hyperthyroid by the administration of sodium 3,3',5-triiodo-L-thyronine (T3) for 7 to 10 weeks. Tenotomy of the soleus muscles was then carried out and the experimental and contralateral muscles were removed and stained for
myosin ATPase
activity after a further 2 weeks. The hyperthyroid state of each animal was confirmed by the assay of
succinate dehydrogenase
activity of liver and the contralateral muscle. After acid preincubation, whole muscle fiber type counts of contralateral muscles showed a statistically significant change from a predominantly acid-stable population of fibers to acid-labile fibers. In addition, many fibers of intermediate staining properties were seen. When the experimental muscles were examined, all three varieties of fiber showed central degeneration. The nature of the fiber type change induced by T3 and the role that innervation might play in this is discussed. It was concluded that the susceptibility of fibers to the lesions that follow tenotomy is dependent on the nature of their innervation rather than their fiber type.
