The article deals with oxidation of different substrates, intensity of glycolytic and glycogenolytic processes in mitochondria and homogenates of dog liver with its 2-hour exclusion from circulation under conditions of endotracheal ether-oxygen narcosis. It was established that already 30-60-minute ischemia causes a decrease in intensity of succinate, alpha-ketoglutarate oxidation and acceptor respiration, inhibiton in the activity of the citrate cycle enzymes; succinate dehydrogenase, alpha-ketoglutarate dehydrogenase, isocytrate dehydrogenase. The activity of NAD-dependent malate dehydrogenasedehydrogenase and Mg2+-ATPase as well as intensity of NADN oxidation in mitochondria increase. After 2-hour ischemia the activity of Mg2+-ATPase, cytochrome oxidase and peroxidase lowers. A sharply developed glycogenolysis is accompanied by inhibition of phosphorylase activity and a two-fold stimulation of the glycolytic reactions. Peculiarities in regulation of enzymatic reactions under conditions of ischemia and their role in origin of metabolism disturbances in the liver are under discussion.

Endothal and cantharidic acid were administered intraperitoneally to mice at 75 and 10 mg/kg, respectively, to compare their acute toxicity on liver tissue in vivo. Within 45 min both treatments caused extreme liver enlargement and congestion. Hepatic glycogenolysis was increased as evidenced by elevations in blood glucose and hepatic glycogen phosphorylase levels and by corresponding reductions in hepatic glycogen content and glycogen synthase activity. Endothal decreased hepatic ATP concentrations, although neither compound altered mitochondrial Mg2+-ATPase activity. Microsomal Mg2+-ATPase levels, however, were reduced by both treatments. There were no indications that reactive intermediates were involved in the toxicity of either compound. The results show that endothal and cantharidic acid act directly and cause similar biochemical changes in mouse liver in vivo.

Studies have been made on the possibility of realization of cAMP regulatory action on carbohydrate and calcium metabolism of skeletal muscles in chick embryos. It was shown that during embryonic period cAMP does not activate phosphorylase system and does not inhibit glycogen synthetase. It was found that the absence of any influence of cAMP on these enzymes of carbohydrate metabolism in embryonic muscles does not depend on the state of cAMP-dependent protein kinase, since this enzyme is functionally active. In contrast to the enzymes of carbohydrate metabolism, Ca2+, Mg2+-ATPase of sarcoplasmic reticulum is sensitive to stimulation by cAMP at least from the 2nd week of embryonic development. It is concluded that embryonic skeletal muscles of chicks exhibit all the conditions which are necessary for realization of the regulatory effect of cAMP. Yet the realization of any effect of the nucleotide depends on the maturity of the given effector link.
