Properties of HCO--3-stimulated
ATPase
from rat heart muscle nuclei were studied. The maximal activity of HCO--3-
ATPase
was observed at concentration of bicarbonate 25 mM. The enzyme had a pH optimum at pH 8.0-8.5. Bicarbonate stimulated the
ATPase
activity only in presence of Mg2+, Mn2+ and Zn2+, Co2+,
Cd2+
and Ca2+ were ineffective. NaCO3 and Na2SO3 at concentration 30 mM stimulated the nuclear
ATPase
activity by 20% and 81%, respectively. Anions N3--, scn--, clO--4, and I-- inhibited both Mg2+-ATPase and HCO--3-
ATPase
. HSO--3 and SO2--4 ions did not affect the nuclear
ATPase
activity.

, Cu2+, Zn2+ and Pb2+. The degree of activation is dependent on the radius of the ion rather than on its nature, in contrast with the dissociation constant of the enzyme--metal ion complex.

We have demonstrated that repetitive injections of
cadmium
into rats leads to the abrupt development of Fanconi syndrome after the renal cortical concentration of
cadmium
has increased approximately eighty-fold. The syndrome is temporally associated with a fall in both Na-K-
ATPase
activity and ATP levels, suggesting a possible causal relationship between altered proximal tubular transport and inhibition of the ATP-Na-K-
ATPase
transport system.

) was purified from brown adipose tissue mitochondria. It had a specific activity of 50 units/mg which could be increased up to 85 units/mg by KHCO3. The isolated enzyme represented less than 0.5% of the initial membrane proteins.2. The enzyme had a molecular weight equal to beef heart
ATPase
and was composed of five subunits with molecular weights of 56 200, 54 300, 33 500, 13 400 and 9500 respectively. 3. Isolated
ATPase
was labile while cold and was activated by the divalent cations Mn2+, Mg2+, Co2+ and
Cd2+
. The optimum ATP/Mg2+ ratio found was 1.58 and the enzyme had a maximum activity at pH 8.5; the Km was 220 micrometer. 4. The
ATPase
activity was 55% inhibited by aurovertin. The isolated enzyme enhanced the fluorescence of aurovertin, quenched by ATP and Mg2+ and enhanced by ADP. 5. Oligomycin sensitivity and cold stability of isolated
ATPase
was restored by its reconstitution with both brown adipose tissue and beef heart particles depleted of
ATPase
. 6. The results presented demonstrate that the low
ATPase
activity of brown adipose tissue mitochondria is due to a reduced content of
ATPase

The properties of anion-sensitive
ATPase
of rat heart mitochondria were studied. Na2CO3, NaHCO3 and Na2SO3 stimualted
ATPase
activity by 69, 41 and 110%, respectively. Azide, tiocinate and perchlorate inhibited bicarbonate-stimulated
ATPase
. Bivalent cations increased
ATPase
activity in such a sequence: Zn2+ greater than or equal to
Cd2+
greater than or equal to Co2+ greater than or equal to Mg2+ greater than or equal to Mn2+ greater than Ni2+. In the presence of bicarbonate and sulfite.
ATPase
activity was maximally stimulated with magnesium. Ni2+ and Ca2+-ions inhibited Mg2+-dependent activity of bicarbonate-stimulated
ATPase
. AMP uninhibited
ATPase
activity. The 4 mM concentration of ADP inhibited activity of HCO-3-
ATPase
. Activity of ATPases decreased at lower temperatures. The properties of anion-sensitive
ATPase
of rat heart mitochondria and that of HCO-2-
ATPase
of other cells are discussed.

The effect of
Cd2+
on the respiration of rat liver mitochondria was investigated. The uncoupling effect of
Cd2+
was partially restored by the addition of Mg2+. The influence of
Cd2+
on adenine nucleotide concentrations in the reaction mixture consisting of mitochondria and ATP was also studied using high performance liquid chromatography. In the presence of added Mg2+, a two-fold increase in AMP concentration was brought about by the addition of
Cd2+
. There was a concomitant decrease in ATP. In the prence of added ADP, an increase in AMP concentration was also brought about by addition of
Cd2+
. The results are discussed in relation to
ATPase
and adenylate kinase activity in mitochondria.

A stable and homogeneous adenosine-5'-
triphosphatase

) has been solubilized from Rhodospirillum rubrum (R. rubrum) chromatophores by chloroform extraction. Purification of the Ca2+-dependent
ATPase
activity was 200-fold. Ca2+ can be replaced by Mg2+,
Cd2+
, and Mn2+. The Km for Ca-ATP (0.17 mM) is increased about 5-fold during solubilization of the enzyme, whereas the Km values for Mg-ATP (0.029 mM) and Cd-ATP (0.014 mM) are not affected. The chloroform-released
ATPase
has a molecular weight of 400,000 +/- 30,000 and consists of the following subunits (molecular weights in parenthesis): alpha(58,000), beta(53,500), gamma(39,000), delta(18,500), and epsilon(14,000). The amino acid composition and the fluorescence spectra are presented. Besides the chloroform-released
ATPase
complex three other Ca2+-dependent
ATPase
forms have been isolated from R. rubrum chromatophores by other methods for comparison. Ultrasonication of the membranes leads to the release of an
ATPase
complex which is mainly composed of alpha, beta, and gamma-subunits. From an acetone powder extract an
ATPase
complex could be purified by affinity chromatography which is composed of four kinds of subunits (alpha, beta, gamma, delta). The same acetone powder yields an
ATPase
consisting of only three different types of subunits (alpha, beta, gamma) if the final purification step is preparative disc electrophoresis on 6% polyacrylamide gels instead of affinity chromatography.

Carbamyl phosphate synthetase from Escherichia coli has been shown to use only the A isomer of adenosine-5'-[2-thiotriphosphate] in both the
ATPase
reaction (MgATP HCO3- leads to MgADP + Pi) and the carbamyl phosphate synthesis reaction (2MgATP + HCO3- + L-glutamine leads to 2MgADP + Pi + carbamyl-P + L-glutamate). The B isomer was less than 5% as reactive. In the reverse reaction, only the A isomer of adenosine-5'-[2-thiotriphosphate] is synthesized from adenosine-5'-[2-thiodiphosphate] and carbamyl-P as determined by 31P NMR and a coupled enzymatic assay with
Cd2+
- hexokinase. It is therefore proposed that carbamyl phosphate synthetase uses the same diastereomer of MgATP at both ATP sites.

of TFo-F1 was maximal at 70 degrees at pH 8.6 and was stable after monomerization in 4 M urea and 0.5% Triton X-100 at 25 degrees. The activity was dependent on Mg2+, Co2+, or Mn2+, and it became insensitive to dicyclohexylcarbodiimide when Ca2+ or

was added instead. 3. TFo-F1 required P-lipids of this bacterium contained branched fatty acyl groups but no unsaturated groups and were stable against oxidation and heat. 4. Studies by electron microscopy, gel electrophoresis, and use of anti-

indicated that the TFo-F1 complex was composed of an

Vesicles catalyzing 32Pi-ATP exchange and ATP-driven enhancement of fluorescence of anilinonaphthalene sulfonate were reconstituted by dialyzing pure TFo-F1 and P-lipids together, and were active even at 50-75 degrees. The vesicles reconstituted from TFo-F1 and bacterial P-lipids were more stable than those reconstituted from TFo-F1 and soybean P-lipids.

A patient with excessive industrial exposure to silicon and an elevated silicon content in his renal tissue was found to have a distinctive nephropathy, characterized pathologically by changes in the glomeruli and proximal tubules, and manifested clinically by albuminuria and hypertension. Proximal tubular function was intact. From a biochemical standpoint, this finding correlates with the demonstration in vitro that, in contrast to
cadmium
, a known cause of Fanconi syndrome, silicon does not inhibit renal cortical sodium-potassium-
adenosine triphosphatase
