Experiments were conducted on the isolated perfused lungs of albino rats; there was shown a dependency of serotonin (ST) absorption on the Na+ and K+ concentration in the perfusate. Which high Na+ concentrations in the perfusate ST was absorbed by the lung tissue cells in great quantities, but when Na+ concentrations were low - ST absorption showed a sharp reduction. K+ concentration in the perfusate had a lesser influence on the ST absorption; the maximal absorption was seen with the K+ concentrations of from 5 to 20 mM. A reduction or an increase in the K+ concentration inhibited the ST absorption. Strophanthin K, a powerful Na, K-dependent ATPase inhibitor, markedly inhibited the ST absorption in a concentration of 10(-4)-- 10(-3) M. It is supposed that there was an association of the ST transport through the cell membrane with the Na+ transport.

In order to examine the question, whether improved digitalis tolerance by Spirolactone may be partially a result of antagonism on myocardial potassium balance, 6 patients without clinical signs of heart failure were given 400 mg, 3 were given 200 or 300 mg Spirolactone orally daily and 6 patients received placebo during a 5 to 7 days period. During cardiac catheterization hemodynamics and serum potassium concentrations were determined repeatedly prior to and following intravenous administration of 0,375 to 0,625 mg Strophanthin. Injection of Strophanthin resulted in a significant drop in left ventricular enddiastolic pressure and a rise in dp/dt max. In the control group a significant increase in arterial and coronary sinus potassium concentration was observed. Myocardial potassium balance was definitely negative from the third to the eighth minute. Values in the group receiving Spirolactone did not differ significantly from the placebo group. It is suggested that therapeutic doses of Strophanthin resllt in a loss of potassium from the myocardium by inhibition of the Na+, K+ membrane ATPase not influenced by pretreatment with Spirolactone.

Astragalus membranaceus saponins (AMS) 50-200 micrograms/ml had a positive inotropic action on the isolated working heart of rats, but in the case of 30 micrograms/ml the inotropic action turned negative. The contractility recovered after washing out the AMS. Strophanthin K performed in a similar manner as AMS. AMS 500 micrograms/ml decreased the resting potential of cultured rat heart cells by 10 mV. The results suggest that AMS plays its inotropic role through influencing the Na-K-ATPase.
