In this report the authors present the results about biochemical analyses in human ejaculate. The following parameters were determined ATP, ADP, pyruvate, lactate, alpha-ketoglutarate, fructose, myokinase, phosphoglucoseisomerase, phosphoglycerate-kinase, ATPase, acid phosphatase. It was found a relationship between age and fructose content, this was equally for pyruvate, whilst the converse was observed for lactate. No age dependend relationship was observed for the other substrates. For ATP and ADP distinctly higher values can be demonstrated in normozoospermia. A special study of the extent to which the encyme activities in sperm plasma are associated with the respective pH values measured shows that pH values above 7.2 correlate with distinctly lower activities for acid phosphatase, PGI and MK.

Seven spleens and two peripheral blood specimens from eight patients with hairy cell leukemia were examined with enzyme cytochemical and histochemical methods. Hairy cells consistently exhibited acid phosphatase and tartrate-resistant acid phosphatase. However, nonspecific esterases characteristic of monocytes and histiocytes were consistently absent or very weak. beta-glucuronidase and cytoplasmic membrane-bound ATPase were positive in four cases, suggesting a possible relationship to the B-lymphocytic series. Fundamental splenic changes were accumulation of hairy cells and benign macrophages within the pulp cords, with resulting extreme expansion of the cords. Abnormally well developed ellipsoids were identified around the sheathed arteries within the cords. Sinuses, specifically delineated with the NASDA reaction, were atrophic and often destroyed. No cytogeneologic relationship was found between sinus endothelial cells and hairy cells. The pulp cords are the primary site of involvement of the spleen in hairy cell leukemia. A simultaneous proliferation of neoplastic cells, histiocytes and reticulum fibers accounts for the splenomegaly and clinical hypersplenism characteristic of the disease.

Renal functional abnormalities constituting the syndrome of postobstructive diuresis imply both altered tubular and glomerular membrane properties. To determine the morphologic and ultrastructural correlates of this disorder a rat model was developed and 32 postobstructed kidneys were studied by light and electron microscopy at the midpoint of diuresis and compared to 22 controls. The abnormal morphology was: dilated distal tubules and collecting ducts, isolated proximal and distal tubule cells that allowed free access of luminal contents to the basement membrane, widened terminal bars and intercellular spaces, thickening of the glomerular basement membrane and, depending upon the portion of nephron, normal or reduced adenosine triphosphatase and acid phosphatase content. In order to confirm the functional nature of the nephrons studied as well as to assess glomerular and tubular permeability, horseradish peroxidase and cytochrome c were infused. These tracers, normally permeable to the glomerular basement membrane, were found in the intercellular spaces and to a lesser extent within cell organelles in the postobstructed diuretic animals whereas controls demonstrated a retarded filtration of horseradish peroxidase, no tracer in the intercellular spaces and large amounts of tracer contained within cell organelles. Absence of enzyme activity in the medulla and reduced dark to light cell ratios in the cortical collecting ducts correlated with prior observations made by others of diminished concentration and acidification processes, respectively. An increase in adenosine triphosphatase activity and renin granules within the juxtaglomerular cells indicated increased renin activity. These observations suggest that the renal functional abnormalities of postobstructive diuresis are attributable to altered glomerular and tubular permeabilities as well as with changes in metabolic activity.

In the bilaterally growing DBD sensitive Yoshida tumours deformed nuclear divisions necrosis of the majority of the tumour and appearance of giant cells can be observed due to the single administration of the LD50 of the preparation. The histochemical activity of the LDH the activity alcalyc Adenosine triphosphatase and the nonspecific alcalyc phosphatase become negative while the acidic phosphatase's activity does increase after the administration of the LD25 of the preparation appearance of giant cells are very marked more than 60% of tumours cells are polynuction. The activity of the alcalyc ATP-ase and nonspecific phosphatase decrose's after a transitory increase and become negative while the acid phosphatase's activity increases. In the case of the DBD resistant tumours the morphological and histochemical alternations due to LD50 of the preparation are much slighter and their timecourse is shorter. No morphological and histochemical changes are observed after the administration of LD25 of the preparation.

Ultrastructural and histochemical changes of skeletal muscle were studied in three patients affected with gas gangrene. There was complete lack of the phosphorylase, succinate dehydrogenase and adenosine triphosphatase activities in the affected muscles of all the patients. In unaffected muscles these enzymes showed weaker activities than in norm. The lactate dehydrogenase activity, especially the heart type isozyme (LDH-1 or H4) proved less sensitive to the effect of clostridial toxin. A general increase in the acid phosphatase activity was found both in affected and in unaffected muscles. On electron microscopic examination damage to sarcolemmal membrane and disintegration of myofilaments was seen. The mitochondria were swollen and their cristae distorted and fragmented.

The effect of dl-norgestrel (13 beta-ethyl-17 alpha-ethynyl-19-nortestosterone, Wy 3707) on the biochemical makeup of rabbit ovary and uterus was investigated. 20 adult, healthy virgin female rabbits either received olive oil only or olive oil plus 12 mcg of norgestrel/rabbit/day for 30 days. The animals were sacrificed 24 hours after the last treatment. In the ovary protein concentration and activities of glucose-6-phosphate dehydrogenase (G6PD), lactate dehydrogenase (LDH), and malate dehydrogenase (MDH) remained unaffected. However, in the uterus the level of protein was significantly elevated (p less than .01), the activities of G6PD and acid phosphatase were enhanced (p less than .05), and those of adenosine triphosphatase (ATPase) and alkaline phosphatase were diminished (p less than .05). LDH and MDH in the uterus remained unchanged. The effect of norgestrel at this antiovulatory dose in rabbits consisted of a disturbance in the biochemical constituents of the uterus leading to a lowering of the ATPase activity and an impairment of the anaerobic mechanism of the organ.

A method is described for the preparation of synaptosomes and synaptosomal membranes from chicken brain. Procedures for isolating rat synaptosomal membranes could not be used directly; several modifications of existing procedures are reported. Purity of the subcellular and subsynaptosomal fractions was monitored by electron microscopy and measurements of ferrocytochrome c: oxygen oxidoreductase (EC 1.9.3.)), monoamine: oxygen oxidoreductase (deaminating) EC 1.4.3.4), rotenone-insensitive NADH: cytochrome c oxidoreductase (EC 1.6.99.3), NADPH: cytochrome c oxidoreductase (EC 1.6.99.1), orthophosphoric monoester phosphohydrolase (EC 3.1.3.2), ATP phosphohydrolase (EC 3.6.1.4), and levels of RNA. Microsomes are the main contaminant of the synaptosomal membrane fraction. Mitochondrial and lysosomal enzymes occur in lesser amounts. No myelin contamination was observed. Marker enzymes for contaminants suggest that these synaptosomal membranes are as pure as membranes described by others, and the specific activity of a neuronal membrane marker, (Na+ -K+)-activated ATPase, is as high as other preparations. Levels of this enzyme in the membrane fraction are enriched 13-fold over homogenate ATPase levels.

Mycotic foci were studied histochemically on various experimental models of candidiasis. NAD-H, NADP-H-diaphorase, acid phosphatase and ATPase were revealed in the fungi, the activity of these enzymes depended on the state of the fungus. Diaphorase activity in the mucous membrane epithelium falls only if it is damaged by massive invasion of pseudo-mycelium. Inhibition of the enzyme activity in the visceral foci (kidney, liver, heart) occurs only in case of pronounced destruction and is not observed at the distance from the fungi. The results do not confirm the idea of fungal secretion of mycotoxins penetrating into the surrounding tissues and damaging them.

Mice infected with protoscoleces of Echinococcus granulosus for 12-14 months were treated ig with mebendazole (Meb) 25-50 mg.kg-1 x d-1 for 7-14 d, albendazole (Alb) 200 mg.kg-1 x d-1, cr praziquantel (Pra) 500 mg.kg-1 x d-1 for 14 d. The mice were killed 24 h after the last medication, and acid phosphatase (ACP), alkaline phosphatase (AKP), and adenosine triphosphatase (ATPase) including (Na, K, Mg)-ATPase, (Na, K)-ATPase, and (Mg)-ATPase were determined and compared with those of untreated control group. The results showed that ACP activities of cyst wall in treated groups were lower than the control group. Whereas AKP activity of cyst wall in Pra group increased markedly, this is not the case in Meb and Alb groups. Three ATPase activities of cyst wall were inhibited in both Meb and Alb groups, Meb being more potent. No apparent changes in the ATPase activities were seen in Pra group.

The enzymatic examinations were carried out to determine, as early as possible, the functional changes in cells of fetal and maternal part of placenta, as well as pointing out those elements which were the most susceptible to cadmium toxic action. The experiments were carried out on 80 pregnant female rats which were divided into three experimental groups and a control one. The experimental animals were daily administered, intragastrically, aqueous solution of cadmium chloride, between the 7th and 19th day of pregnancy, in doses depending on experimental groups: 2, 10 and 20 mg of CdCl2 per kilogram of body weight, respectively, the animals were killed on the 21st day of pregnancy. Placenta sections were determined for activity of adenosinetriphosphatase stimulated by Mg++ ions (ATP-ase Mg++) E.C.3.6.1.3., and acid phosphatase (AcP) E.C.3.1.3.2. When administered to pregnant females, cadmium chloride was proved to cause a considerable impairment of active transport through biological membranes in placenta, which is indicated by a decrease of reactivity to Mg++ ions stimulated ATP-ase; and an increase in intracellular catabolic processes, which in turn is shown by an increase in reactivity to acid phosphatase. The fetal part of the placenta proves to be more susceptible to the lesion causing activity of cadmium than the maternal part of this organ.
