1. Acetylcholine (ACh), 7.5 x 10(-5) M, and carbachol, 5 x 10(-6) M (CCh) depressed the frequency of miniature endplate potentials (m.e.p.ps) in the frog (Rana temporaria) sartorius neuromuscular junction with active acetylcholinesterase to about 50-55% of the controls. 2. A similar depression was produced by the nicotinic agonists, nicotine, suberyldicholine and tetramethylammonium. 3. The muscarinic agonists, oxotremorine, methylfurmethide and methacholine were without effect on m.e.p.p. frequency. The muscarinic antagonist, atropine and the nicotinic antagonist, (+)-tubocurarine, had no effect on the depression of m.e.p.p. frequency evoked by CCh. 4. The ganglionic blockers, benzhexonium and IEM-1119, were also without effect on the CCh-evoked depression of m.e.p.p. frequency. 5. Pretreatment of muscles with anticholinesterases did not prevent the CCh-induced drop in m.e.p.p. frequency. 6. The effect of CCh was proportionally the same as in the controls in preparations where the m.e.p.p. frequency was changed by elevation of K+ and in the presence of theophylline, noradrenaline, dibutyryl adenosine 3':5'-cyclic monophosphate (db cyclic AMP) and db cyclic GMP. 7. We have recently shown that the mean muscle chronaxie for nerve pedicle implanted into denervated rabbit strap muscle is comparable to that of normal nerve. This study correlates excitability with histologic characteristics of muscles reinnervated via nerve-muscle pedicles (NMP) and direct nerve implants (DNI). Strength duration curves were measured in 13 rabbits 3.5 to 5 months after reinnervation by NMP (n = 6) and DNI (n = 7). Following this, control (n = 5) and reinnervated straps were harvested immediately before the animals were killed and frozen in liquid nitrogen. The nerve endings of normal hair of the rat's snout, partially digested with trypsin and hydrochloric acid, were studied by scanning electron microscopy. Each lanceolate structure measured ca. 10 microns in length and was arranged around the hair follicle. These palisade-shaped nerve endings were situated almost beneath the sebaceous glands, ran upward, parallel to the axis of the hair follicle, and terminated in pointed shape. 2 kinds of cells, Teloglia cell Type I showing flat profile, and Teloglia cell Type II showing spherical profile and possessing numerous caveolae in its surface were observed at the basal portion of the palisade-shaped endings. The axon was enclosed by Schwann cells in its course to the hair follicle, and was covered with Type I cells at the beginning, and with Type II cells at the end, and constituted the palisade-shaped nerve endings. The palisade structure in silver impregnated tissues observed by backscattered electron microscopy and X-ray analyzer was characterized as comprising neuronal elements. Mothers who smoke cigarettes during pregnancy give birth to babies with lower birth weights than do nonsmoking mothers. One hypothesis to explain this finding is that nicotine depresses the activity of the placental cholinergic system, which has been linked to the placental transport of amino acids and other substances. The levels and activities of several components of the term placental cholinergic system were determined in smokers and nonsmokers to investigate whether this system is involved in the effect of smoking. There were no statistically significant differences in the levels, synthesis or release of acetylcholine in the tissues from smoking and nonsmoking mothers, nor in the activities of the choline uptake system or the enzymes choline acetyltransferase,

. The results do not support the hypothesis that the lower birth weights of babies born to smoking mothers is mediated by an effect of nicotine or other tobacco components on the placental cholinergic system.

Acetylcholinesterase (AchE: EC 3.1.1.7) was identified and purified from the hemolymph of the scorpion Heterometrus bengalensis. The purity of the enzyme was determined by polyacrylamide gel electrophoresis (PAGE). The molecular weight of the enzyme, determined by sodium dodecyl sulfate-PAGE, was 80,000. The purified AchE hydrolysed acetylthiocholine iodide, but it did not react with butyrylthiocholine iodide. BW284C51, a specific inhibitor of AchE, strongly inhibited the enzyme. The known inhibitor (tetramonoisopropylpyrophosphortetramide) of
pseudocholinesterase
did not produce any inhibition of the enzyme activity. The purified AchE of scorpion hemolymph was vulnerable to high substrate concentration. The presence of Cu2+ and Ni2+ reduced the enzyme activity, whereas the metal ion, Sn2+, enhanced AchE activity. Ca2+ produced neither inhibition nor activation. (Na+, K+)-
ATPase
and Mg2+-ATPase activities were greatly enhanced by the purified AchE.

For evaluation of neurotoxicity of 1,3-bis[2-chloroethyl]-1-nitrosourea (BCNU) effects of the compound were examined on histoenzymatic activity of some phosphatases and esterases and on karyometry and DNA cytophotometry in brain neurocytes of the progeny of mothers who had been given the drug during pregnancy. The studies were performed on young, 14 and 28 days old rats of Wistar strain originating from mothers given intraperitoneal BCNU injections on the 12th and 18th day of pregnancy. Material of the studies included neurocytes of frontal cortex, parietal cortex and of trigeminal nerve nucleus in which enzymatic activities of some phosphatases and esterases were estimated and which were subjected to karyometric and DNA cytophotometric measurements, using Morphoquant automated microscopic image analyzer (VEB Carl Zeiss, Jena). The experiments documented decreased nonspecific esterase and

activities and increased activities of acetylcholinesterase,

, alkaline phosphatase and acid phosphatase in neurocytes of experimental animals, as compared to control animals of the corresponding age. The changes were noted both in 14 and 28 days old animals. Changes in thiamine pyrophosphate phosphohydrolase (TTPase) enzymatic activity were of different type in dependence upon age of the animals (increased activity in 28 days old rats, decreased activity in 14 days old rats). In animals subjected to BCNU action in fetal life several karyo- and cytophotometric changes were noted also: circumference and area of cell nuclei cross-sections were decreased, cell nuclei became more round, relative DNA content of cell nuclei increased with parallel increases in compactness and concentration of nuclear chromatin. Karyometric changes showed similar character in both age groups while changes in DNA cytophotometry were more evident in 14 days old rats.

A new antiaggregating chemical, alpha-(p-(fluoren-9-ylidenemethyl)phenyl)-2-piperidineethanol (RMI 10,393), designated FYPE, was found to be an effective inhibitor of platelet aggregation induced by adenosine diphosphate (ADP), thrombin, collagen, or epinephrine. Effects of the antiaggregant on platelets were concentration dependent. Aggregation was prevented by low concentrations of FYPE that produced in the platelet only minor ultrastructural changes consisting of loss of microtubules and of discoid shape. Low levels of FYPE that prevented platelet aggregation had no effect on platelet

activities but did alter clot retraction, the thrombin-induced shift in electrophoretic mobility and platelet

activity. Market decrease in ADP release and increase in adenyl cyclase activity were produced by low levels of FYPE. This study provides a model for evaluation of platelet antiaggregating compounds in vitro.
