The distributions of acid and alkaline phosphatases, 5-nucleotidase, ATPase, non-specific esterase, specific cholinesterase, succinic dehydrogenase and beta-galactosidase are described in the mesencephalic auditory, tegmental and cranial nerve nuclei of the frog (Rana tigrina). The main results of the study are as follows: The laminar, principal, and magnocellular nuclei of the torus semicircularis, which are associated with auditory functions, show intense activity of specific cholinesterase. On the other hand, the commissural and subependymal mid-line nuclei, whose functions are doubtful, show a complete lack of this enzyme. The nucleus isthmi shows intense acid phosphatase, ATPase, non-specific esterase, specific cholinesterase and succinic dehydrogenase activities. Non-specific esterase is virtually absent from all the areas studied except the nucleus isthmi and the 3rd and 4th cranial nerve nuclei. Most of the commissures and fibre tracts show intense activity for beta-galactosidase and 5-nucleotidase. The possible roles of these enzymes in glycolipid and myelin metabolism are discussed.

It was shown that the phenomenon of inactivation of Na, K-ATPase of the non-purified fraction of the rat cortical synaptosomes under electroshock may be related to "modification" of the potassium active center of the enzyme. The anticonvulsant diazepam injected intramuscularly also inhibits Na, K-ATPase of the cerebral membranes. However, in subsequent electrical stimulation of the brain the drug activates Na, K-ATPase as compared to controls. Diazepam also abolishes clonic convulsions induced by electrical stimulation of the brain. At the same time it does not eliminate compensatory shifts in the activity of acetyl-cholinesterase of the rat cerebral and spinal synaptosomes, characteristic of electroshock. The results are discussed from the standpoint that inhibition of the activity of Na, K-ATPase of the nerve endings membranes may underlie the pathogenetic mechanism of the convulsive activity.

Comprehensive investigations were carried out for establishing the biological and nutritional value of low erucic-acid rapeseed oil from a variety of rape called Janpol selected in Poland. The pathophysiological effects of Janpol rapeseed oil were observed after giving it as the only source of fat in the diet or added in different proportions to other edible fats. In all cases the total amount of fat in the diet was 20 p. 100 kcal. The investigations were carried out on 78 young male Wistar rats aged 25 days at the beginning of the experiment. The rats were divided into 7 groups and they were given diets containing: 1) soybean oil; 2) mixed fats; 3) rapeseed oil of high erucic-acid content; 4) mixed fats containing 25 p. 100 of Janpol rapeseed oil; 5) mixed fats with 50 p. 100 of Janpol rapeseed oil; 6) mixed fats with 75 p. 100 of Janpol rapeseed oil; 7) Janpol rapeseed oil only. The experiment lasted 3 months. After its completion the rats were decapitated after 18 hours of starvation. The investigation s included : determination of weight gain, determination of the weight of selected organs (liver-lungs, heart, kidneys, testes, spleen), determination of alkaline phosphatase and pseudocholinesterase activity in the serum, determination of triglycerides and cholesterol in the serum, tests for adrenocortical function, histo-chemical investigations of the liver (alkaline and acid phosphatase, adenosine triphosphatase, fatty infiltration of the liver), macroscopic and microscopic anatomopathological examinations. The authors found the Janpol rapeseed oil caused less pronounced changes in the determined indices of the biological and nutritional evaluation as compared with high-erucic-acid rapeseed oil. Janpol repeseed oil given to experimental animals mixed with other fats in proportions of 25 p. 100 and 50 p. 100 of all fats in the diet, that is 5 p. 100 and 10 p. 100 kcal in the diet derived from Janpol oil gave in most determinations of the investigated parameters results very similar to those observed in animals receiving soybean oil. The results of these investigations show that Janpol rapesed oil can be used for nutrition of man in amounts not exceeding 10 p. 100 of the total caloric content of food.

Lipid antioxidants, DPPD, DAH, BHT, SQ, retinol and alpha-tocopherol were studied for their effects on normal rat erythrocytes. Retinol, BHT and SQ were found to induce hemolysis while DPPD, DAH and alpha-tocopherol were non-hemolytic. Further more the three antioxidants BHT, SQ and retinol showed a synergistic effect on the lysis. Retinol, BHT or SQ alone, and BHT or SQ along with retinol when administered to rats produced a marked modification of the erythrocyte membrane integrity, simultaneously lowering the levels of membrane bound enzymes--acetyl choline esterase and ATPase. It is concluded that the lipid antioxidants may therefore be classified on the basis of their lytic action in vitro.

Fragmented sarcoplasmic reticulum (FSR) was prepared from the white muscles of the catfish (Amiurus nebulosus). The effect of La3+ on the functional characteristics of FSR was studied. La3+ in a concentration higher than 10(-4) M was found to decrease or arrest Ca2+ accumulation, cholinesterase activity and the activation of ATPase by Ca2+. La3+ added after the elimination of membrane-bound Ca2+ of FSR (1 mM EGTA, pH 7.1) does not substitute Ca2+ in its functions. The cholinesterase activity of FSR solubilized by deoxycholate and purified by gelfiltration is inhibited by La3+ present in a concentration higher than 10(-4) M and simultaneously with inhibition, the absorbance at 280 nm is increased. Ca2+ gives rise to similar changes only in concentrations higher than 10(-2) M.

The structure and histochemistry of the palmar and plantar skin were studied in four adult male marmosets (two Callithrix jacchus and two Callithrix penicillata). In this skin there exist well-developed epidermal ridges, to which are attached one or two ducts of sweat glands. A thick stratum corneum can be seen in the epidermis, while a distinct stratum lucidum cannot be isolated from the other layers. The stratum granulosum is constituted by one or three layers of cells containing keratohyalin granules. Melanin granulations are mainly concentrated in the basal cells of the epidermal ridges. Dendritic melanocytes and amelanotic melanocytes containing alkaline phosphatase are found among the epidermal cells. Glycogen, UDPG-GT and phosphorylases are mainly present in the middle and lower Malpighian cells of the epidermal ridges. Alkaline phosphatase, ATPase, alanyl amino-peptidase and leucine aminopeptidase were absent in the epidermal cells. SDH, cytochrome oxidase, MAO and a certain number of NAD-dependent dehydrogenases (LDH, ADH, MDH, alpha-GPDH, beta-OHBDH and GDH) showed a stronger reactivity in the basal cells and Malpighian layer. The NADP-dependent enzymes (G-6-PDH, 6-PGDH, cis-aconistase and ICDH) were more reactive in the upper Malpighian layer and stratum granulosum. The stratum corneum showed some acid phosphatase and nonspecific esterase reactivity. The collagenous fibers intertwined with a small number of very thin elastic ones and a larger amount of reticular fibers run almost parallel to the epidermal ridges in the papillary body. In the reticular dermis some fibers are disposed transversely to the epidermal ridges. Meissner corpuscles reactive to butyrylcholinesterase, acetylcholinesterase, nonspecific esterase and G-6-PA are disposed at regular intervals and frequently at each side of the epidermal ridges. Pacinian corpuscles were found only in the hypodermis. The eccrine sweat glands contain glycogen, UDPG-GT and phosphorylase in their secretory, ductal and myoepithelial cells. The secretory part shows a uniform reactivity for every dehydrogenase because it contains only one type of cells (clear cells). The intraepidermal segment of the ducts shows a stronger reactivity to nonspecific esterase and NADP-dependent dehydrogenases than the epithelial cells around it.

The dermal cells in grey, xanthic, and white goldfish integuments were cytochemically characterized for the following enzymatic activities: tyrosinase, DOPA-oxidase, cytochrome oxidase, monoamine oxidase, peroxidase, non-specific esterase, cholinesterase, NAD-diaphorase, NADP-diaphorase, aryl sulfatase, nucleotide phosphodiesterase, beta-glucuronidase, acid phosphatase, alkaline phosphatase, adenosine triphosphatase, thiamine pyrophosphatase, glucose-6-phosphatase, aldolase, as well as succinate, malate, isocitrate, glutamate, glucose-6-phosphate, 6-phosphogluconate, alpha-glycerophosphate, alcohol, lactate, and beta-hydroxybutyrate dehydrogenases. It was found that the epidermis was a significant barrier to the access of cytochemical reaction substrates. Removal of the epidermal barrier provided dermal cell localizations of enzymatic activities which were reproducible. Further, alterations in reaction times and temperatures from the mammalian methodology provided conditions fe various integumental cells were compared for possible interrelationships. The basic foundations for future work with the dermis of poikilothermic vertebrates on an experimental basis were established. In addition, a previously undescribed non-pigmented dermal cell, the "x"-cell, was found to have enzymatic characteristics similar to both melanophores and lipophores. The "x"-cell may be the common precursor of both types of pigment cells.

We investigated the effects of eserine and ouabain on the permeability of the blood-brain barrier (B.B.B.) as related to the febrile response induced with LPS in rabbit. Results are as follows; The febrile response induced by LPS (0.02 and 1.0 mug/kg) i.v. was suppressed by administration of ouabain (0.06 mg/kg, i.v.). Contrary to the febrile response of LPS given i.c. (10(-4) and 10(-3) mug/kg), the febrile response was not suppressed with the same dose of ouabain. The pyrogenicity of cerebrospinal fluid (CSF) withdrawn at two hours after rabbits had been injected with LPS (25 mug/kg) was suppressed by ouabain (0.06 mg/kg, i.v.). Pyrogenic response was enhanced by pretreatment with eserine (0.5 mg/kg, s.c.) given one hr before LPS (1 mug/kg, i.v.). The pyrogenicity of CSF was also potentiated to a greater extent by pretreatment of eserine than with LPS alone. In the eserinized rabbit (0.5 mg/kg, s.c.), the pyrogenicity of CSF was potentiated to a greater extent by ACh (10 mug/kg, i.v.) than by LPS (1 mug/kg, i.v.) alone. From these data, it is concluded that the inhibition of Na, K-ATPase by ouabain decreases the pyrogenicity of LPS, while the inhibition of cholinesterase by eserine enhances the pyrogenicity.

Previous studies have shown that iodoarachidonates (IAs) prevent goiter production in rats. In the present studies we show that both IL-d and IL-w (IAs bearing the iodine atom at the positions 6 and 14, respectively), cause a significant involution of preformed goiter. This effect was evident when IAs were administered either orally or via i.p., although the first one required larger doses to obtain the same degree of inhibition. No changes were observed in serum protein, urea, cholesterol, cholinesterase, T3 or T4. In vitro studies with FRTL-5 cells showed that both IAs inhibit iodide and alpha-AIB uptake, as well as ATPase activity.

Alterations in erythrocyte plasma membrane properties (enzymatic activities and membrane fluidity) have been observed in patients affected by insulin-dependent diabetes mellitus (IDDM) and non-insulin-dependent diabetes mellitus (NIDDM). In order to verify whether these alterations are present also in gestational diabetes mellitus (GDM) we studied the plasma membranes obtained from two different cellular types (erythrocyte from both mother and cord blood and placenta syncytiothrophoblast cell) of 16 healthy pregnant women and 15 women affected by GDM. The following determinations were performed on the membrane preparations: Na+/K(+)-ATPase activity, acetyl-cholinesterase (AchE) activity, membrane fluidity and cholesterol:phospholipid ratio. We observed a reduction of both enzymatic activities and a decrease of membrane fluidity in maternal and cord blood erythrocytes and in syncytiotrophoblast plasma membranes in GDM pregnant women in comparison with controls. The cholesterol to phospholipid ratio was significantly lower in the erythrocyte membranes of women affected by GDM than in normal pregnant women, while it was increased in the cord blood erythrocyte membranes and in placental membranes in GDM in comparison with controls. The present study found, in GDM patients, a membrane alteration similar to the abnormality reported in IDDM and NIDDM (i.e. decreased Na+/K(+)-ATPase activity), while opposite modifications were observed with regard to other membrane activities and properties. The different membrane alterations observed in GDM with respect to IDDM and NIDDM might be linked to the different degree of metabolic control, on the contrary the reduced Na+/K(+)-ATPase activity might be a primary event in the pathogenesis of diabetes mellitus per se and might constitute a signal of high risk of developing the disease later in the women affected by GDM during pregnancy.
