Using histochemical technique, the effects of radiofrequency catheter ablation (RFCA) on the activities of LDH, SDH, CCO, and Ca++-ATPase of guinea-pig ventricular myocytes were examined. The histological changes were observed for comparison. Radiofrequency energy (500 kHz) delivered was 20 W x 10 s. The results were as follows: RFCA resulted in significant impairments in all the four kinds of enzymes but without statistical differences in the areas involved in this energy level. No statistically significant difference was found between the ranges of enzymatic damages and areas of pathological lesions. These findings showed a consistency in areas of the histological and histochemical lesions resulted from RFCA.

The experimental rats were randomly divided into three groups: i.e. control group; endotoxic shock group (the model of endotoxic shock was induced by intravenous administration of E. Coli endotoxin, 16 mg/kg); and electroacupuncture (EA) group ("Renzhong" or "Zusanli" acupoint was stimulated for 15 minutes at 1 hr after injection of endotoxin). The experimental rats were decapitated at 75 minutes after injecting endotoxin. Their livers were taken out for cryostat section, and histochemical observation. The results were as follows: 1) The glycogen in the hepatic cells of endotoxic shocked rats were almost completely depleted. The activities of SDH Mg(++)-ATPase and G-6-Pase and 5'-Nase were decreased; especially the activities of 5'-Nase in the biliary canaliculi and sinusoids were apparently reduced. 2) The content of hepatic glycogen in EA group was increased, but some of them was still depleted. The activities of SDH, Mg(++)-ATPase and G-6-Pase were slightly increased as compared with that of the endotoxic shock group. The activity of 5'-Nase was obviously increased after EA. The preliminary results indicated that EA at "Renzhong" or "Zusanli" acupoint of rats with endotoxic shock might play certain role on improving the hepatic metabolism and promoting the membrane transport action.

Thymi of adult chickens (White Leghorn strain), ages of 6, 18, 45, days and 1, 3, 6, months, were studied by histological and histochemical stainings, histoenzymatic reactions (LDH, SDH, alpha-GPDH, NADH, NADPH, Ca++ dependent ATPase, pH 8.5) and by anti-thymostimulin immunoreaction. Positive reactions for thymostimulin-like substance, mucopolysaccharides and enzymatic activities were located in different epithelial compartments of the cortical and medullary zones. In 3 and 6 month-old chickens, there was a progressive reduction of the cortical area and of the number of cortical and medullary epithelial cells, and, in the thymi of the 6 month-old chickens, a partial decline in the immunological reactivity to anti-TS, but the intensity of the histochemical and enzymatic reactivities, in comparison with the thymi of the youngest chickens, was not decreased. This may suggest that the cortical and medullary epithelial cells still present in these thymi are capable of secreting and expressing activities and, thus, may maintain their functions.

Dynamic cardiomyoplasty, a method to support ventricular function by the chronically stimulated latissimus dorsi muscle wrapped around the heart is accompanied by a loss of mass and force of the transplanted muscle. These effects and the fast-to-slow transformation of the muscle could be possibly influenced by the additional administration of anabolic steroids. In this study, the left latissimus dorsi muscles of 12 sheep were electrically conditioned (group A). In 12 other animals (group B), stimulation was combined with the administration of metenolone (100 mg/week). Biopsies were taken from the right and left muscles at the beginning and after 6 and 12 weeks of treatment, frozen and cross-sectioned. The muscle fibre type composition was studied enzymhistochemically (SDH-staining and Myosin-ATPase-reaction) and immunocytochemically (using antibodies against different myosin heavy chains, MHC). Furthermore, the expression of different MHC isoforms was investigated electrophoretically. The untreated latissimus dorsi muscle contains 20% type I fibres expressing slow MHC and 80% type II fibres expressing fast MHC. After 6 weeks, the respective fibre type composition was 42 and 58% (group A) and 80 and 20% (group B). After 12 weeks, the percentage of the type I fibres rose in group A to 59% and in group B to 98%. In accordance with these morphological results, the MHC pattern determined electrophoretically showed a corresponding shift from the fast to the slow isoform. Therefore, the administration of metenolone avoids severe muscle atrophy, and improves and accelerates fast to slow fibre type conversion necessary for successful cardiomyoplasty.

The study was performed on rats divided into 9 groups. Groups 1-3 served as controls. In groups 4 and 5 rat livers were subjected to 90-min ischemia followed by 12- or 24-hour reperfusion. In groups 6 and 7 rats were injected with intraperitoneal chlorfenvinphos (2 mg/kg b.w.) and sacrificed after 12 or 24 hours. In groups 8 and 9 rat livers were subjected to 90-min ischemia, 12- or 24-hour reperfusion and then rats were injected with chlorfenvinphos (2 mg/kg b.w.). Liver sections were evaluated morphologically, histochemically (SDH, LDH, G6Pase, glycogen, Mg2+ ATPase and AcP). The microsomal fraction of the liver was evaluated for cytochrome P450 content and NADPH-cytochrome P-450 reductase activity. It has been found that liver ischemia and reperfusion result in extensive necrosis, enzymatic disturbances, particularly in acinar zone 3. Ischemia as well as reperfusion decrease the cytochrome P450 content of hepatocytic microsomes and the activity of NADPH-cytochrome P-450 reductase. Intraperitoneal injection of chlorfenvinphos during ischemia and reperfusion dramatically intensifies damage to the liver, although chlorfenvinphos alone produces only mild nonspecific effects on the morphological and enzymatic structure of the liver.

Thiram was administered to male rats through gavage at doses 5, 10 and 25 mg/kg/day for 180 and 360 days. Thiram has caused marginal increase in the relative weight of testes and epididymis and decrease in the weight of seminal vesicle and prostate. Marked degenerative changes were observed in seminiferous tubules together with alterations in testicular enzyme profile. The activity of testicular enzymes such as ACP, SDH and ATPase (Na+ + K+ dependent) was decreased whereas activity of LDH, G-6-PDH and ALP increased. The levels of serum cholesterol and testicular free sialic acid were enhanced, while the level of testicular protein was lowered. It is evident from the present study that long term treatment of thiram at tested dose levels has resulted in dose and time dependent morphological and biochemical changes in testes of rat.

Effects of medroxy progesterone acetate (MPA; 5 mg/kg) and MPA + testosterone enanthate (TE) (3 mg + 2 mg/kg) were investigated in vas deferens and on fertility (along with reversibility study) for 60 days through histopathology, morphometric and certain biochemical parameters such as total proteins, sialic acid, ATPase, SDH and testosterone. The study revealed for altered histopathology and contratile pattern of vas deferens resulting in reduced fertility. The study also indicated androgen antagonistic effect. These effects were found to be reversible 4 and 3 months after withdrawal of MPA and MPA + TE injections respectively. Thus, both types generated functional sterility in the rat, but MPA affected histophysiology of vasal tissue.

Histochemical and ultrastructural studies were performed on lateral musculature from individual female weakfish (Cynoscion striatus Cuvier). Based upon SDH, PhR, Sudan and myosin-ATPase determinations, different kinds of red, pink and white fibres were discerned at the anterior, medium and posterior regions. Levels of glycolytic activity increased from red to white fibres, being intermediate in the pink ones. Contrarily, oxidative activity decreased from red to pink and white fibres. Histochemical stain showed a complex distribution of fibre types in each layer. A considerable variation in size and distribution was also found among fibres.

Our study investigated the effects of Corylus avellana extract on acetaminophen and carbon tetrachloride intoxicated liver of young rats. Hepatocytolysis was determined by measuring the level of serum transaminases (GPT and GOT), steatosis by Sudan black staining, histological structures by hamatoxylineosin staining and the activity of enzymes such as SDH, GtDH, G-6-Pase and ATPase. Comparatively, the most serious lesions appeared in CCl4 intoxication. Corylus avellana extract had some beneficial effects in CCl4 toxicosis: it reduced hepatocytolysis as well as histological lesions and returned the activity of some enzymes to normal values.

Malignant Hyperthermia (MH) represents a functional myopathy triggered by volatile anesthetics and depolarizing muscle relaxants, and leading to metabolic disturbances of intracellular Calcium homeostasis. Central-Core-like-structures (CCLS) were recently described as central defects in enzyme-histochemical stains and well correlated to the autosomal-dominant MH-predisposition. We studied the correlation of a MH-predisposition with specific myopathological signs. Skeletal muscles of suspected MH-individuals were histochemically stained by SDH-, NADH-, COX-, Gomori-Trichrome-, ATPase-, Acid Phosphatase-, Oil-red O- und PAS-stain und evaluated without knowing MH-diagnosis by the in-vitro-contracture test. Out of 118 patients (30% MHS ["susceptible"], 63% MHN [normal], 7% MHE ["equivocal"]) 19% revealed pathological findings corresponding to CCLS. 45% of these findings were associated with MHS/MHE. With HE-staining internal nuclei were not specific, but increased with the probability of MHS/MHE from 24% to 80%. Central Cores were correlated in 100% with MHS/MHE (4 out of 118 patients). CCLS were found with about similar frequency in skeletal muscle of MHS/MHE and MHN individuals. Internal nuclei were, however, not specifically, associated with MHS. In contrast, Central Cores correlated significantly with MHS/MHE diagnosis. In conclusion, histopathological findings in skeletal muscle seem to be a reliable marker for MH-predisposition only with Central Cores.
