The characteristics of the glycolytic pathway and the plasma membrane of Lactobacillus and Bifidobacterium were studied. The enzyme system of glycolysis (hexokinase,
glucokinase
and pyruvate kinase) which is the main source of energy in the anaerobic condition was localized in the cell soluble fraction (cytoplasma) of all species examined. Neither electron transfer chain components nor oxidase activities were found in anaerobically cultured Lactobacillus and Bifidobacterium. Adenosine
triphosphatase
(ATPase) activities were mainly localized in the plasma membrane, suggesting that membrane ATPase is playing a key role in membrane transport and ATP synthesis of anaerobic bacilla. SDS-polyacrylamide gel electrophoresis of membranes showed remarkable differences between the polypeptides patterns of B. adolescentis and B. bifidum. Such peculiarities in polypeptide patterns among the same genus may be useful in the identification of species.

Mycelia of a low- and a high-production strain of Streptomyces aureofaciens were converted into protoplasts and divided into five subcellular fractions in order to localize exopolyphosphatases (EC 3.6.1.11),
triphosphatase
(
EC 3.6.1.25
), inorganic diphosphatase (EC 3.6.1.1), apyrase (EC 3.6.1.5) and
glucokinase
(
EC 2.7.1.2
). The highest specific activity of enzymes hydrolyzing polyphosphates was found in cytoplasmic vesicles and membranes. Triphosphatase was detected in the periplasmic fraction. Periplasmic vesicles and cytoplasm exhibited a high activity of diphosphatase. Apyrase was found mainly in the fractions of membranes and cytoplasmic vesicles. Glucokinase was a cytoplasmic enzyme. The enzymes were released from membrane structures into cytoplasm or periplasmic space if benzyl thiocyanate (10 microM) was present in the growth medium.

Preneoplastic liver lesions were produced in female Wistar rats by oral administration of 2-acetylaminofluorene for 165 days succeeded by a carcinogen-free standard diet up to 420 days. During the treatment numerous altered hepatic foci (AHF) and hyperplastic nodules (HN) were detected histochemically by a focal decrease or lack of adenosine-5-

and glucose-6-phosphatase (G-6-Pase) activities. In addition, the immunohistochemically demonstrable amount of L-type pyruvate kinase was clearly reduced. The histochemically demonstrated decrease of G-6-Pase was substantiated by microbiochemical determination of the enzyme activity in microdissected material. Moreover, during the experimental period a continuous decrease in
glucokinase
and an increase in hexokinase was detected microbiochemically within AHF and HN. These alterations indicate a shift in the carbohydrate metabolism from gluconeogenesis to glucose utilization and pentose-phosphate-pathway for biosynthesis of nucleic acids. Beside other oncofetal markers, HK may be used as indicator of the early stages of liver carcinogenesis.

Glycolysis by the supernatant fraction of homogenates of liver from guinea pigs and rats at various stages of development (foetal, newborn and adult) has been examined in a suitably fortified medium by measurement of inorganic phosphate uptake and production of lactate and glycerol 1-phosphate. 2.
