alcalase was used to catalyze the synthesis of the target dipeptide in

-organic cosolvent systems. The conditions of the synthesis reaction were optimized by examining the effects of several factors, including organic solvents,
water
content, temperature, pH, and reaction time on the yield of Z-Asp-Val-NH(2). The optimum conditions using alcalase as the catalyst are pH 10.0, 35 degrees C, in acetonitrile/Na(2)CO(3)-NaHCO(3) buffer system (9:1, V/V), reaction time 5 h, with a yield of 63%. The dipeptide product was confirmed by LC- MS.

The degree of copper-induced oxidative stress (OS), established by measuring lipid peroxidation, protein oxidation, and cellular glutathione level, as well as activities of antioxidant enzymes--catalase, superoxide dismutase, and gultathionine peroxidase, depended on the mode of copper administration. Chronic copper administration (mild oxidative stress) did not affect

activities, whereas acute copper treatment (severe oxidative stress) caused a decline in chymotryptic- and tryptic-like activities. Hypothalamic norepinephrine (NE) release regulates arterial pressure by altering sympathetic nervous system activity. Because angiotensin (Ang) (1-7) decreases hypothalamic NE release and this effect may be correlated with a diminished NE synthesis, we hypothesize that Ang-(1-7) down-regulates tyrosine hydroxylase (TH), the rate-limiting enzyme in catecholamines biosynthesis. We investigated the effect of Ang-(1-7) on centrally TH activity and expression. TH activity was evaluated by the release of tritiated
water
from (3)H-l-tyrosine. TH expression and phosphorylation were determined by western blot. Hypothalami from normotensive or spontaneously hypertensive rats pre-incubated with Ang-(1-7) showed a significant decrease in TH specific activity. Ang-(1-7) caused a decrease in TH phosphorylation at Ser19 and Ser40 residues. The heptapeptide induced a decrease in TH expression that was blocked by an AT(2) receptor antagonist and not by an AT(1) or Mas receptor antagonist, suggesting the involvement of AT(2) receptors. The proteasome inhibitor MG132 blocked the Ang-(1-7)-mediated TH reduction. In addition, Ang-(1-7) increased the amount of TH-ubiquitin complexes, indicating that the Ang-(1-7)-mediated TH degradation involves ubiquitin conjugation prior to
proteasome
degradation. We conclude that Ang-(1-7) down-regulates TH activity and expression centrally leading to a decrease in the central NE system activity.

The objective of this study was to further investigate the effects of music on fish physiology, bearing in mind available information regarding the involvement of endogenous and exogenous factors in fish farming. Therefore, Cyprinus carpio (50.5 +/- 0.36 g) were reared in a recirculating

system under 80 and 200 lux and subjected to no music at all (control, ambient noise only), 4 h of Mozart's "Eine Kleine Nachtmusik", or 4 h of anonymous "Romanza-Jeux Interdits" for 106 days. Both music treatments resulted in increased growth performance at both light intensities, with Romanza treatment at 200 lux resulting in better growth performance than Mozart treatment. plays a critical role in the regulation of several important cellular processes and has drawn extensive interest in the field of anti-tumor research. Peptide aldehydes can inhibit the 20S
proteasome
activity by covalently binding to the active site of the beta subunits. In this work, covalent docking in conjunction with molecular dynamics (MD) simulation was used to explore the binding mode of MG132. Two conformations with the lowest docking energy were selected as the representative binding modes. One of the conformations was confirmed as a more reasonable binding mode by molecular dynamics simulations. The binding mode analysis revealed that a space demanding aromatic group with a short linker at the P4 site of the peptide aldehyde inhibitor would form favorable hydrophobic contacts with the neighboring subunit. A bulky substituent at the P2 position would also increase the binding stability by reducing

accessibility of the covalent bond. This study contributed to our understanding of the mechanism and structure-activity relationship of the peptide aldehyde inhibitors and may provide useful information for rational drug design.

The abilities to accumulate cadmium (Cd) are different among cultivars (cv.) in many species. The characteristic of Cd concentration among cultivars is heritable and is probably controlled by genes, but rather limited information about the relevant genes in vegetable crops has been published. In the present study, a suppression subtractive hybridization (SSH) approach was used to identify genes induced by Cd in two
water
spinach (an important vegetable in southern China) cultivars that differ in Cd accumulation in their edible parts. The two cultivars were cv. Qiangkunqinggu (QK), a low Cd accumulative cultivar and cv. Taiwan 308 (TW), a high Cd accumulative cultivar. In the construction of QK and TW libraries, the plants without Cd treatment were taken as drivers and the plants exposed to 6 mg L(-1) Cd for 24 h as testers. Four hundred clones were sequenced, and 164 nonrepeated sequences (112 from the QK library and 52 from the TW library) were assigned to being functional genes or proteins. A tremendous difference in Cd-induced gene expressions between the two libraries was observed. In the QK library, genes implicated in disease/defense comprised one of the largest sets (20.6%), whereas the proportion was only 8.8% in the TW library. QK and cv. TW using the RT-PCR method, and obvious differences were detected. The relationships between the identified differences in the expressions of the genes and the Cd accumulation of the two cultivars were discussed, and it was concluded that the SSH approach is useful for finding the difference in expression of Cd-induced gene even at the cultivar level and is applicable in the investigation of the mechanisms of low Cd accumulation.
