The effects of the recombinant human cytokines interleukin 2 (IL-2) and IL-7 on the proliferation of T-acute lymphoblastic leukaemia (T-ALL) cells were tested in a clonogenic assay. Highly purified leukaemic cells were obtained by immunomagnetic depletion of mature T cells and fluorescence-activated cell sorting for immature leucocyte markers (CD1, CD10, CD34). Of 9 cases tested, only 3 showed evidence of stimulation by cytokines. One was stimulated by both IL-2 and IL-7, one by IL-2 only, and the third by IL-7 alone. A further case showed proliferation without addition of cytokines. The remaining 5 cases were completely unresponsive. While both IL-2 and IL-7 are capable of stimulating leukaemic cells from some cases of T-ALL, the molecules regulating the proliferation of T-ALL cells in vitro remain to be more fully elucidated.

Phorbol ester (TPA)-induced down-regulation of the common ALL (CALLA) antigen was studied by continuous flow immunocytometry with the aid of several CD10 monoclonal antibodies, including a new CD10 monoclonal antibody (DGH-10-1-A9), shown to be of IgG1 isotype, recognizing a 100 kDa cell surface protein and effectively inhibited by a series of reference CD10 monoclonal antibodies. The TPA-induced down-regulation of CALLA on REH cells was demonstrated with the aid of the following CD10 monoclonal antibodies: J-5, VIL-A1 and DGH-10-1-A9. No major modulations in cell surface expression of CALLA on REH cells were observed after induction with 1,25-(OH)2 vitamin D3, retinoic acid, recombinant interferon (IFN) alpha 2a and recombinant interleukin 2.

The reactivity spectrum of an anti-CALLA/CD10 monoclonal antibody for cutaneous melanoma was analysed by immunohistochemistry in a series of lesions of different Breslow thickness. Similar proportions of small primary tumours, advanced primary tumours and metastatic lesions were found to express CALLA/CD10 (31-47%). However the proportion of stained cells within a given lesion increased with tumour progression. Up to 23% of the advanced primary lesions (> 3.0 mm) showed 26-50% cells stained with the anti-CALLA/CD10 antibody and up to 14% of the metastatic lesions showed 76-100% stained cells. The expression of CALLA/CD10 was further analysed in 15 ocular melanoma lesions of different histiotype. All five spindle type lesions, three of six epitheloid and two of five mixed type lesions stained positively with the anti-CALLA/CD10 antibody. The percentage of stained cells within a given lesion varied from 30% to 100%. A total of 63% of the ocular melanomas and 38% of the cutaneous melanomas tested expressed CALLA/CD10. Experiments with cultured melanoma cell lines showed that the surface expression of CALLA/CD10 can be modulated in vitro by treatment with interleukin 2 (IL-2) and an adenosine 3',5'-cyclic monophosphate (analogue).

This study addressed several questions concerning age-related changes in human B lymphopoiesis. The relative abundance of pro-B, pre-B, immature, naive, and mature B cells among the CD19(+) lymphocyte fraction of human bone marrow was found not to change appreciably over the interval between 24 and 88 years of age. Moreover, proliferation of pro-B and large pre-B cells in adult marrow equaled that observed with fetal marrow specimens. Exceptionally low numbers of lymphocyte precursors were found in some marrow samples, and the values obtained were used to determine parameters that best reflect B lymphopoiesis. Cord blood always contained higher incidences of functional precursors than adult cells. However, sorted CD34(+) Lin(-) CD10(+) progenitors from cord blood and adult marrow had equivalent potential for differentiation in culture, and notable age-related changes were found in more primitive subsets. A recently described subset of CD34(+)CD38(-)CD7(+) cord blood cells had no exact counterpart in adult marrow. That is, all adult CD34(+)Lin(-)CD7(+)CD10(-) cells expressed CD38, displayed less CD45RA, and had little B-lineage differentiation potential. The CD7(+) fractions in either site contained progenitors for erythroid and natural killer (NK) lineages, and ones sorted from marrow expressed high levels of transcripts for the CD122 interleukin 2 (IL-2)/IL-15 receptor required by NK-lineage precursors. Dramatic changes in human B lymphopoiesis occur early in life, and more information is required to construct a probable sequence of differentiation events prior to the acquisition of CD10.

As shown by R5 antibody-based sandwich and competitive enzyme-linked immunosorbent assay (ELISA), selected sourdough lactobacilli, in combination with fungal proteases, hydrolyzed gluten (72 h at 37 degrees C) of various cultivars of Triticum turgidum L. var. durum to less than 20 ppm. Complementary electrophoretic, chromatography, and mass spectrometry techniques were used to characterize the gluten and epitope hydrolysis. Nine peptidases were partially purified from the pooled cytoplasmic extract of the sourdough lactobacilli and used to hydrolyze the 33-mer epitope, the most immunogenic peptide generated during digestion of Triticum species. At least three peptidases (general aminopeptidase type N [PepN], X-prolyl dipeptidyl aminopeptidase [PepX], and endopeptidase PepO) were necessary to detoxify the 33-mer without generation of related immunogenic epitopes. After 14 h of incubation, the combination of all or at least six different peptidases totally hydrolyzed the 33-mer (200 mM) into free amino acids. The same results were found for other immunogenic epitopes, such as fragments 57-68 of alpha 9-gliadin, 62-75 of A-gliadin, and 134-153 of gamma-gliadin. When peptidases were used for fermentation of durum wheat semolina, they caused the hydrolysis of gluten to ca. 2 ppm. The in vivo digestion was simulated, and proteins/peptides extracted from pepsin-trypsin (PT) digestion of durum wheat semolina fermented with selected sourdough lactobacilli induced the expression of gamma interferon and interleukin 2 at levels comparable to those of the negative control. Durum wheat semolina fermented with sourdough lactobacilli was freeze-dried and used for making Italian-type pasta. The scores for cooking and sensory properties for this pasta were higher that those of conventional gluten-free pasta.

This study compared the gene expression profiles of primary leukaemic cells from infants versus children with acute lymphoblastic leukaemia (ALL). Our analyses provided unprecedented evidence that remarkably different pathognomonic transcriptomes dominate the biology of infant versus paediatric high risk ALL. The genetic signature of infant ALL is characterized by concomitant overexpression of mitogenic and anti-apoptotic genes, some of which have been associated with early relapse in ALL. Our study demonstrated that primary leukaemia cells from infant ALL patients expressed significantly higher levels of genes for cytokines that mediate their biological effects through stimulation of the JAK-STAT signal transduction pathway including interleukin 1a, interleukin 1b, interleukin 2, and interleukin 7. We further showed that the JAK/STAT signalling pathway is constitutively active in CD10(-) infant ALL cells and treatment with a JAK3 inhibitor or a pan-JAK kinase inhibitor effectively triggered their apoptosis. These findings identified JAK3 as an attractive molecular target for disrupting the constitutively deregulated anti-apoptotic STAT3 and STAT5 signalling pathways in infant ALL cells.
