The structure of Eubacterium nodatum cell wall peptidoglycan was investigated. The peptide subunit of E. nodatum peptidoglycan has the following structure: L-Ala-D-Glu (Gly)-L-Orn-D-Ala. The carboxyl group of alanine occupying position 4 is attached to the delta-amino group of ornithine of an other subunit by the cross-linking bridge L-Ala-L-Ala-L-Orn. All glycine molecules are connected with the alpha-carboxyl group of glutamic acid with the ratio being 0.5-1. The hydrolysis of E. nodatum peptidoglycan by the S. albus G enzyme proceeds primarily due to the activity of alanyl-alanine endopeptidase, ornithyl-ornithine endopeptidase, ornithyl-alanine endopeptidase, N-acetyl-muramyl-alanine amidase, N-acetylmuramidase and N-acetylglucosaminidase.

Outbreaks in humans, caused by Streptococcus suis serotype 2 (SS2), were reported in 1998 and 2005 in China. However, the mechanism of SS2-associated infection remains unclear. For the first time, a 2-D gel approach combined with MS was used to establish a comprehensive 2-D reference map for aiding our understanding of the pathogenicity of SS2. The identification of 694 out of 834 processed spots revealed 373 proteins. Most of the identified proteins were located in the cytoplasm and were involved in energy metabolism, protein synthesis, and cellular processes. Proteins that were abundant in the 2-DE gels could be linked mainly to housekeeping functions in carbohydrate metabolism, protein quality control and translation. 2-DE of secretory proteins was performed using IPG strips of pH 4-7. Among the 102 protein spots processed, 87 spots representing 77 proteins were successfully identified. Some virulence-associated proteins of SS2 were found, including arginine deiminase, ornithine carbamoyl-transferase, carbamate kinase, muramidase-released protein precursor, extracellular factor, and suilysin. Enolase and endopeptidase have been proposed as putative virulence-associated factors in this study. The 2-D reference map might provide a powerful tool for analyzing the virulence factor and the regulatory network involved in the pathogenicity of this microorganism.

The distinction of hepatocellular carcinoma (HCC) from metastatic tumor in the liver often presents a diagnostic challenge that carries significant impact on prognostication and therapy. The number of diagnostically useful immunohistochemical markers of hepatocytes is limited to hepatocyte paraffin antigen (HepPar-1), polyclonal carcinoembryonic antigen, and CD10, with alpha-fetoprotein and glypican-3 labeling HCCs. Arginase-1 (Arg-1) is a binuclear manganese metalloenzyme that catalyzes the hydrolysis of arginine to ornithine and urea. We used immunohistochemistry to compare the sensitivity of Arg-1 to that of HepPar-1 in 151 HCCs. We found that the overall sensitivities of Arg-1 and HepPar-1 are 96.0% and 84.1%, respectively. The sensitivities of Arg-1 in well, moderately, and poorly differentiated HCCs are 100%, 96.2%, and 85.7%, respectively, whereas, in comparison, HepPar-1 demonstrated sensitivities of 100%, 83.0%, and 46.4% for well, moderately, and poorly differentiated tumors, respectively. There were no HCCs in our study that were reactive for HepPar-1 but nonreactive for Arg-1. We also examined Arg-1 expression in nonhepatocellular tumors, including many that are potential mimics of HCC (renal cell carcinomas, neuroendocrine tumors, melanomas, gastric adenocarcinomas, and adrenocortical carcinomas) and found that only 2 non-HCC tumors were reactive for Arg-1. Arg-1 represents a sensitive and specific marker of benign and malignant hepatocytes that may ultimately prove to be a useful diagnostic tool in routine surgical pathology practice.

The spermatophore of the silkmoth,Bombyx mori, is a reactor with a specific energy-yielding system for sperm maturation, the arginine degradation cascade. On mating, the highly viscous secretions from various glands in the male reproductive tract, which contain many enzymes and their substrates, are transferred to the female bursa (b.) copulatrix to form the spermatophore. In the spermatophore, transferred arginine-rich proteins are digested by initiatorin, an Arg-C endopeptidase of serine-protease type, and a carboxypeptidase. The produced free arginine is then hydrolyzed to urea and ornithine by arginase. Ornithine is metabolized to glutamate, follwed by forming alanine and 2-oxoglutarate. The latter, as a member of TCA-cycle, is a preferred respiratory substrate for spermatozoa and accelerates the post-testicular sperm maturation.In contrast toBombyx mori, Drosophila melanogaster produces only eupyrene spermatozoa and does not form the spermatophore. The sperm of this dipteran insect acquire motility in the v. seminalis of males. As reported forDrosophila, a high glutamate-pyruvate aminotransferase activity was found in the spermatophore as well as the v. seminalis of the silkmoth. The value in the latter organ reaches 58.3% of the whole male reproductive tract that participates in transfer of the seminal fluid.In the male reproductive system ofDrosophila, the concentration of arginine is low, whereas those of urea and ammonia are high. The accessory gland secretion contains much phosphoserine. Theses substances are transferred to female uterus with spermatozoa during mating. Most amino acids increase distinctly at 30 min after the termination of mating (ATM) and then decline, suggesting active degradation of transferred proteins in the uterus. As found inBombyx, urea increases at the post-mating period, while ornithine shows a rather low concentration. Ornithine must be converted to glutamate. In this connection, it is notable that alanine rises markedly at 30 min following mating. As in the silkmoth, the energy metabolism of the fruit fly spermatozoa involves also arginine, ornithine, urea, and proline. These findings suggest that the occurrence of the arginine degradation cascade or related metabolic pathway in this insect.
