Discovery of an

by gel chromatography and separation of 3 exopeptidases (a
dipeptidase
, an aminopeptidase and a specific carboxypeptidase) from Lactobacillus casei NCDO 151 by affinity chromatography is described. The 3 exopeptidases were strongly inhibited by the metal chelators EDTA and 1,10-phenanthroline but were reactivated with Co2+ and Mn2+. The pH optima for aminopeptidase,
dipeptidase
and carboxypeptidase activities were 6.5, 7.6 and 7.2, respectively. Maximum activity was obtained at 45 degrees C for the aminopeptidase, at 30 degrees C for the
dipeptidase
and at 40 degrees C for the carboxypeptidase. The substrate specificities of the 3 enzymes were also studied. The properties of these 3 enzymes are compared with those of other bacteria.

Stimulation of [(3)H]thymidine incorporation of thymocytes and splenocytes from guinea pigs by various bacterial cell walls and their peptidoglycans, by enzymatic digests, and by synthetic muramyl dipeptides was studied as an indication of mitogenic activity. Cell wall and peptidoglycan preparations, isolated from 19 strains belonging to 18 different species, definitely increased [(3)H]thymidine incorporation of thymocytes as well as splenocytes, regardless of mycolic acid contents as a non-peptidoglycan component. Both the cell walls from Nocardia corynebacteriodes (containing mycolic acids) and those from Streptomyces gardneri (lacking mycolic acids) showed far stronger mitogenic activities on splenocytes than other cell walls (stimulation index, 25 to 30). Furthermore, water-soluble enzymatic digests, notably the
endopeptidase
digests, which generally were greater in degree of polymerization of peptidoglycan subunits than the glycosidase digests obtained from representative cell walls, were found to have as distinct a stimulating activity on splenocytes as the original cell walls. In contrast, solubilization of the cell walls by enzymes, irrespective of endopeptidases or glycosidases, was accompanied by disappearance of the mitogenic activity on thymocytes. enzyme activity was established to be decreased with 49,2 per cent, glycyl-l-leucine--with 33,2 per cent, glycyl-l-valine with 19,6 per cent, glycyl-l-isoleucine--with 61 per cent, etc. The diminished enzyme activity corresponds, in the majority of the cases, to the severity of the disease and to the degree of the histological changes. It does not reach the decreased degree, found in the patients with coliac sprue. In a series of cases the enzymatic activity does not correspond to the morphological changes: "normal activity" and decreased peptidase activity were found in six cases and in two cases--"partial mucosal atrophy" and normal or elevated peptidase activity. Very likely, the pointed out enzymes, are of a substantial importance for the pathogenetic digestive and resorbtive disturbances in chronic non-specific enteritis, especially protein disturbances.

Both the standard, undifferentiated population and a differentiated population expressed only three peptidases: dipeptidyl peptidase IV, aminopeptidase W and carboxypeptidase M. In the differentiated HT-29 cells the activity of dipeptidyl peptidase IV after 14-21 days was beginning to plateau whereas aminopeptidase W activity was still rising and that of carboxypeptidase M had begun to decline. These differences in activity profiles observed among this group of cell-surface peptidases indicate that these cell lines, especially Caco-2, are useful models to study the regulation of their expression.

The effects of a range of metallopeptidase inhibitors on the activities of the porcine kidney cell surface zinc aminopeptidases, aminopeptidase A (AP-A; EC 3.4.11.2), aminopeptidase N (AP-N; EC 3.4.11.7) and aminopeptidase W (AP-W; EC 3.4.11.16), have been directly compared. Amastatin and probestin were effective against all three aminopeptidases, with the concentration of inhibitor required to cause 50% inhibition (I50) in the low micromolar range (I50 = 1.5-20 microM), except for probestin with AP-N which displayed an I50 of 50 nM. Actinonin failed to inhibit significantly either AP-A or AP-W, and thus can be considered a relatively selective inhibitor (I50 = 2.0 microM) of AP-N. In contrast, bestatin was a relatively poor inhibitor of AP-N (I50 = 89 microM) and failed to inhibit AP-A, but was more potent towards AP-W (I50 = 7.9 microM). Thus, some of the observed chemotherapeutic actions of bestatin may be due to inhibition of cell-surface AP-W. A number of other metallopeptidase inhibitors, including inhibitors of
endopeptidase

EC 3.4.24.11
) and
membrane dipeptidase
(EC 3.4.13.11), and the carboxylalkyl and phosphoryl inhibitors of angiotensin converting enzyme (EC 3.4.15.1) failed to inhibit significantly AP-A, AP-N or AP-W. However, AP-W was inhibited with I50 values in the micromolar range by the sulphydryl converting enzyme inhibitors rentiapril (I50 = 1.6 microM), zofenoprilat (I50 = 7.0 microM) and YS 980 (I50 = 17.7 microM). Neither AP-A nor AP-N were affected by these sulphydryl compounds. Inhibition of AP-W may account for some of the side effects noted with the clinical use of the sulphydryl converting enzyme inhibitors. The availability of compounds which are totally selective for AP-W over any of the other mammalian cell surface zinc aminopeptidases may aid in identifying endogenous substrates, and thus physiological or pathophysiological role(s) of AP-W.

A (angiotensin converting enzyme), aminopeptidase-N and dipeptidyl peptidase IV are present in the microvessels, and aminopeptidase-N is also seen in the perineurial connective tissue. Of this group of peptidases, only aminopeptidase-W is a neuronal marker in normal nerve. Macrophages were identified by two antibodies, 74-22-15 and 40D (which recognises Ia). Short-term cultures of macrophages obtained by alveolar lavage were positively stained by both antibodies and about half of the cells also expressed aminopeptidase-N and dipeptidyl peptidase IV. Staining by 40D and 74-22-15 revealed the presence of significant numbers of macrophages in normal nerve, but none of the membrane peptidases colocalized with these cells. Seven days after section of the nerve, the distal segment showed morphological changes typical of Wallerian degeneration. Endopeptidase-24.11 was no longer visible in myelin sheaths, but remained a marker for the surface of Schwann cells (defined also by staining for glial fibrillary acidic protein). The macrophage markers revealed marked changes in the morphology of these cells, often consistent with their phagocytic activity. Two peptidases, aminopeptidase-N and aminopeptidase-W, also appeared at this time to be associated with cells exhibiting the morphology of activated macrophages. This association could be confirmed in many instances by double staining with 74-22-15 and antibodies to the peptidases. Angiotensin converting enzyme retained its single location in microvessels at 7 days after section, but at 14 days a new pattern emerged as it, too, was expressed by macrophages. Dipeptidyl peptidase IV was not shown to be a macrophage marker in the degenerating nerve. Thus Wallerian degeneration leads to remarkable changes in the cellular expression of membrane peptidase;

-24.11 associated with a battery of exopeptidases. The manner by which some neuropeptides are degraded by the combined attack of the peptidases of this membrane has been investigated. The contribution of individual peptidases was assessed by including inhibitors (phosphoramidon, captopril, amastatin and di-isopropyl fluorophosphate) with the membrane fraction when incubated with the peptides. Substance P, bradykinin and angiotensins I, II and III and insulin B-chain were rapidly hydrolysed by kidney microvilli. Oxytocin was hydrolysed much more slowly, but no products were detected from [Arg8]vasopressin or insulin under the conditions used for other peptides. The peptide bonds hydrolysed were identified and the contributions of the different peptidases were quantified. For each of the susceptible peptides, the main contribution came from

activity. Its pI is 4.10, its molecular weight is 21000 Da and its A1%(1 cm) at 280 nm is 9.75. It rapidly hydrolyzes casein and hemoglobin.
