Immunohistochemical studies were carried out on the new type of cerebral cortical astrocytic inclusions recently discovered in a 20-year-old patient with maldeveloped brain and micropolygyria. The inclusions appeared as eosinophilic structures (hematoxylin and eosin stain) and did not exhibit argyrophilia (modified Bielschowsky method). The inclusions were strongly stained by the antibody against S-100 protein (S 100) and to a lesser extent by the antibody to microtubule-associated protein 1B (MAP 1B). In contrast to Rosenthal fibers, the astrocytic inclusions did not react with antibodies to alpha B-crystallin, glial fibrillary acidic protein and ubiquitin. No positive reactions were obtained with antibodies against heat-shock protein 27 (HSP 27), HSP 72, actin, vimentin, desmin, cytokeratin, myelin basic protein, beta-tubulin, MAP 2, tau protein, paired helical filament, phosphorylated neurofilament protein (NFP), nonphosphorylated NFP, synaptophysin,

By immunoelectron microscopy, the products of the reaction with the anti-S 100 antibody appeared as heterogeneous granular deposits and with the antibody to MAP 1B they were randomly scattered throughout the astrocytic inclusions. Our results demonstrate that the immunohistochemical profile of the recently described inclusions differs from that of Rosenthal fibers. Whether the novel inclusions are involved in congenital astrocyte dysfunction and cerebral malformation remains to be established.

, alpha 1-AT, and alpha 1-AChy was identical in ectopic and intrauterine gestation: there was labeling of villous syncytiotrophoblast and a proportion of Hofbauer cells but no labeling of villous cytotrophoblast. The majority of interstitial extravillous trophoblast yielded negative results, but alpha 1-AT and alpha 1-AChy were consistently demonstrated in endovascular trophoblast. FXIII was not found in any trophoblast population but was demonstrated in Hofbauer cells, stromal fibroblasts, and interstitial dendritic cells. Granular, extracellular FXIII reactivity was present among sheets of infiltrating extravillous trophoblast in ectopic pregnancy but only occasionally in early intrauterine pregnancy. The results document further the heterogeneity of trophoblast, with the endovascular trophoblast forming an immunophenotypically distinct population. Furthermore, the pattern of extravillous trophoblastic invasion of maternal tissues in ectopic pregnancy appears to differ from intrauterine pregnancy; the poor decidualization of tubal mucosa in ectopic pregnancy may play a role in this variation.

A stable, tamoxifen-resistant subline, MCF-7/TAMR-1, of the human breast cancer cell line MCF-7 has been established in tissue culture after long-term treatment with 10(-6) M tamoxifen. The MCF-7/TAMR-1 cell line grows equally well in the presence and absence of tamoxifen, whereas the steroidal antiestrogens ICI 164,384 and ICI 182,780 exert profound inhibitory activity on cell proliferation, although higher concentrations are required to inhibit these cells compared to the parent cells. The MCF-7/TAMR-1 cells grown in tissue culture deviate from parent characteristics by the complete lack of expression of progesterone receptors even when grown with estradiol, by an altered tamoxifen regulation of M(r) 52,000

The sensitivity to the growth inhibitory activity of ICI 164,384 and ICI 182,780 may be ascribed to the maintenance of the pure antagonistic effect of these steroidal antiestrogens on MCF-7/TAMR-1 cells. Our results indicate that treatment with pure antiestrogens may be effective when patients become refractory to tamoxifen therapy.

However, it has not been considered that the uPA system is interacting with several other protease/inhibitor systems, neither has a comparable prognostic role of these factors been investigated. We reported earlier the potential of tomato
cathepsin D
inhibitor (SlCDI) as an in-built stabilizing agent for the protection of recombinant proteins in transgenic plant leaf crude extracts (Plant Biotechnol J.4, 359-368). Here we document the potential of SlCDI for the in situ protection of proteins in potato leaves. Total protein assays with control and SlCDI-expressing potato lines indicated a positive impact of slcdi transgene expression on leaf protein content, with a mean relative increase of 35%-40% depending on the light regime. Out of approximately 700 proteins detected on 2-D gels, only 20 exhibited a significantly altered level on a protein-specific basis, whereas most proteins were up-regulated on a leaf fresh weight basis, albeit at variable rates. Quantitative reverse trancriptase-PCR assays for rubisco activase showed similar transcript levels in leaves of test and control lines despite protein levels increased by two- to threefold in SlCDI-expressing lines. These observations, along with the unrelated biological functions assigned to MS-identified proteins up-regulated in leaves and protease assays showing slightly increased proteasome activity in protein extracts of SlCDI-expressing lines, suggest a general, proteasome-independent protein stabilizing effect of SlCDI in planta. Transient expression assays with human alpha(1)-
antichymotrypsin
also showed a stabilizing effect for SlCDI on heterologous proteins, leading to net levels of the human protein increased by approximately 2.5-fold in SlCDI-expressing plants. These data illustrate, overall, the potential of SlCDI as an in vivo protein-stabilizing agent in transgenic plant systems, useful to improve protein levels and recombinant protein accumulation.
