The protein kinase PKR is a major player in the cellular antiviral response, acting mainly by phosphorylation of the alpha-subunit of the eukaryotic translation initiation factor 2 (eIF2-alpha) to block de novo protein synthesis. PKR activation requires binding of double-stranded RNA or PACT/RAX proteins to its regulatory domain. Since several reports have demonstrated that translation is inhibited in apoptosis, we investigated whether PKR and eIF2-alpha phosphorylation contribute to this process. We show that PKR is proteolysed and that eIF2-alpha is phosphorylated at the early stages of apoptosis induced by various stimuli. Both events coincide with the onset of caspase activity and are prevented by caspase inhibitors. Using site-directed mutagenesis we show that PKR is specifically proteolysed at Asp(251) during cellular apoptosis. This site is cleaved in vitro by recombinant caspase-3, caspase-7, and caspase-8 and not by the proinflammatory caspase-1 and caspase-11. The released kinase domain efficiently phosphorylates eIF2-alpha at the cognate Ser(51) residue, and its overexpression in mammalian cells impairs the translation of its own mRNA and of reporter mRNAs. Our results demonstrate a new and caspase-dependent activation mode for PKR, leading to eIF2-alpha phosphorylation and translation inhibition in apoptosis.

In the present study, we have found evidence for ER stress occurring during development of the central nervous system in the mouse. Several ER-resident stress-regulated chaperones, such as calreticulin, glucose regulated protein 78, glucose regulated protein 94, ER protein 57 and protein disulfide isomerase, were expressed at higher levels in embryonic brain and retina, compared with adult tissues. In contrast, calnexin, a chaperone that is not regulated by stress was equally abundant in embryonic and adult tissues. We also detected unfolded protein response during embryonic development. Both eukaryotic translation initiation factor 2 alpha and its phosphorylated form were more abundant in embryonic brain and retina than in adult tissues. Spliced X-box binding protein-1 mRNA was detected in embryonic brain and retina, while it was absent in adult counterparts. Partially glycosylated form of activating transcription factor 6 alpha, another ER stress indicator, was detected predominantly in embryonic brain. Finally, apoptotic pathway components, caspase-7 and -12, were more abundant in embryonic brain than in adult. The pattern of expression of chaperones together with activation of the unfolded protein response factors suggests the presence of ER stress during development of brain and retina. Furthermore, our data suggest that ER stress-like mechanism may induce apoptosis via activation of the caspases during embryonic development of the central nervous system.

Increased levels of misfolded polypeptides in the endoplasmic reticulum (ER) triggers the dissociation of glucose-regulated protein 78 (GRP78) from the three transmembrane ER-stress mediators, i.e., protein kinase RNA-like ER kinase (PERK), activating transcription factor-6 (ATF6), and inositol-requiring enzyme 1alpha, which results in the adaptive unfolded protein response (UPR). In the present studies, we determined that histone deacetylase-6 (HDAC6) binds and deacetylates GRP78. Following treatment with the pan-histone deacetylase inhibitor panobinostat (Novartis Pharmaceuticals), or knockdown of HDAC6 by short hairpin RNA, GRP78 is acetylated in 11 lysine residues, which dissociates GRP78 from PERK. This is associated with the activation of a lethal UPR in human breast cancer cells. Coimmunoprecipitation studies showed that binding of HDAC6 to GRP78 requires the second catalytic and COOH-terminal BUZ domains of HDAC6. Treatment with panobinostat increased the levels of phosphorylated-eukaryotic translation initiation factor (p-eIF2alpha), ATF4, and CAAT/enhancer binding protein homologous protein (CHOP). Panobinostat treatment also increased the proapoptotic BIK, BIM, BAX, and BAK levels, as well as increased the activity of caspase-7. Knockdown of GRP78 sensitized MCF-7 cells to bortezomib and panobinostat-induced UPR and cell death. These findings indicate that enforced acetylation and decreased binding of GRP78 to PERK is mechanistically linked to panobinostat-induced UPR and cell death of breast cancer cells.
