Beta-amyloid (Abeta) peptide is the major component of senile plaques and considered to have a causal role in the development and progression of Alzheimer's disease. There has been compelling evidence that Abeta-induced cytotoxicity is mediated through oxidative and/or nitrosative stress. Compared to N-acetyl-l-cysteine, which mainly scavenges reactive oxygen species, EGT effectively inhibited Abeta-induced cell death by suppressing peroxynitrite formation and subsequent nitration of protein tyrosine residues. The effects of EGT on the cytotoxicity induced by the nitric oxide donor sodium nitroprusside (SNP) and the peroxynitrite-generating 3-morpholinosydnonimine chlorhydrate (SIN-1) were compared. Whereas EGT significantly protected against SIN-1-mediated cell death, it barely affected the cytotoxicity induced by SNP. Thus EGT may attenuate apoptosis caused by Abeta, preferentially by eliminating peroxynitrite derived from the neurotoxic peptide. The importance of diet-derived antioxidants in the management of Alzheimer's disease and other neurodegenerative disorders is discussed.

The role of MCLR induced oxidative stress in the initiation of apoptosis was investigated over a 2-hr period, and it was found that there was an increase in the release of H(2)O(2) in the first 30 min of exposure for both cell lines. Both cell lines exhibited a dose-dependent increase in both micro- and millicalpain after 24 h exposure to MCLR suggesting a role for protease activation in MCLR-induced apoptosis in non-hepatic human derived cell lines.

extract of P. urinaria induced the apoptosis of HL-60 cells as demonstrated by morphological change, DNA fragmentation and increased
caspase-3
activity. However, normal human peripheral mononuclear cells remained viable under the same treatment. The P. urinaria-induced apoptosis of HL-60 cells was associated with the increased Bax gene expression and decreased Bcl-2 gene expression. In addition, the gene expressions of Fas receptor and Fas ligand, but not p53, were also induced in HL-60 cells dose- and time-dependently. The inhibitor of ceramide synthase, fumonisin B1, completely suppressed the apoptosis induced by P. urinaria and this inhibitory effect of fumonisin B1 could be eliminated by the addition of ceramide. It indicated that the activity of ceramide synthase is critical for the P. urinaria-induced apoptosis in HL-60 cells. The P. urinaria-induced apoptosis in HL-60 cells is mediated through a ceramide-related pathway.

1. The clinical use of doxorubicin is limited by the development of severe cardiomyopathies linked, at least in part, to an abnormal increase in the rate of apoptotic cell death. Because cell shrinkage is considered to be a crucial step at the onset of apoptosis, the aim of the present study was to investigate whether a brief hypo-osmotic stress, which leads to an increase in cell volume, could interfere with the induction of apoptosis by doxorubicin in adult cardiomyocytes. 2. Cell volume expansion results in intracellular accumulation of cAMP, so we secondarily tested whether the protective effect of hypo-osmotic stress could be related to the cAMP pathway. Accordingly, apoptosis was induced by doxorubicin (1 micromol/L) in cardiomyocytes freshly isolated from New Zealand adult rabbit hearts. 3. Exposure to doxorubicin in an iso-osmotic medium (290 mOsmol/kg

) induced a rapid decrease in cell volume, as well as increases in annexin V labelling and

activity, two biological markers of apoptosis. These effects of doxorubicin were abolished by 15 min pretreatment with hypo-osmotic stress at 220 mOsmol/kgH2O (HS 220). 4. This cytoprotective effect of HS 220 was still observed when doxorubicin was added to the medium 60 min later, but it was abolished when the pretreatment by HS 220 was associated with the protein kinase A inhibitor KT 5720 (200 nmol/L). 5. Conversely, 15 min pretreatment with either the cAMP analogue 8-bromo-cAMP (0.5 mmol/L) or the adenylate cyclase activator forskolin (10 micromol/L) inhibited apoptosis induced by doxorubicin. 6. In conclusion, these results demonstrate that: (i) apoptosis induced by doxorubicin can be counteracted by a hypo-osmotic stress in adult cardiomyocytes; and (ii) activation of the protein kinase A-dependent pathway plays a major role in the mechanism leading to the cytoprotective effect induced by a hypo-osmotic stress.

water
chlorination. MX, MCA, and MCF also promote foci formation in the two-stage cell transformation assay. The cellular mechanisms underlying this apparent promotional effect are not known. In the present study, the effects of MX, MCA, CMCF, and MCF on gap junctional intercellular communication (GJIC) were measured in BALB/c 3T3 cells using the scrape loading dye technique. The effect of MX on apoptosis in the same cell line was explored by assaying
caspase-3
-like protease activity. All the four CHFs inhibited GJIC after 30 min exposure in a dose-dependent fashion but there was a marked difference in the ranges of their active concentrations. MX was almost as potent an inhibitor of GJIC (inhibition at nanomolar concentrations) as 12-O-tetradecanoylphorbol-13-acetate (TPA) (positive control), while MCA was 10 times weaker, CMCF 10,000 times weaker, and MCF 20,000 times weaker than MX. After prolonged exposure periods (up to 6 h), GJIC recovered somewhat upon MX and MCA exposures, the inhibition of GJIC by MCF remained constant but CMCF showed an irreversible increasing inhibitory effect. MX caused apoptosis as a "window" effect at concentrations 2000-4000-fold higher than those needed to inhibit GJIC. The results indicate that MX is a potent inhibitor of GJIC in BALB/c 3T3 cells and this inhibition might be one mechanism by which MX can promote malignant foci formation. MCA also has a specific potential to inhibit GJIC whereas MCF and CMCF affected GJIC at concentrations, similar to those evoking genotoxicity in vitro.

Efficacy of a safe and clinically utilized polyethylene glycol formulation (PEG-3350) to suppress intestinal tumors was investigated in the Apc(min) mouse-model of experimental carcinogenesis. Furthermore, based on our previous finding on the induction of apoptosis in HT-29 cells by PEG, we evaluated its ability to stimulate epithelial cell apoptosis in both Apc(min) mouse as well as AOM-treated rat as a potential molecular mechanism of chemoprevention. Twenty-two Apc(min) mice were randomized equally to PEG or vehicle (control) supplementation. . Supplementation of Apc(min) mice with 10% PEG 3350 (in drinking

) resulted in a 48% (P<0.05) reduction in intestinal tumor burden and induced 2-3 fold increase in mucosal apoptosis. Dietary supplementation of polyethylene glycol (5%) also stimulated colonic mucosal apoptosis 4-5 fold in AOM-treated rats, the regimen that we previously reported to reduce tumor burden by 76% (P<0.05). In summary, we demonstrate, for the first time, that PEG does protect against Apc(min) mouse tumorigenesis. The correlation between pro-apoptotic actions and chemopreventive efficacy of PEG in these models strongly implicates induction of apoptosis as one of the impending mechanisms of chemoprevention.

The cells of the kidney medulla are exposed routinely to high extracellular concentrations of various solutes including NaCl, urea and ammonium (NH4+). Although it is well established that the expression of a variety of osmosensitive genes and proteins, which confer cytoprotection on renal medullary cells, is induced by high NaCl concentrations, the role of NH4+ in these cellular responses is unclear. This study thus addressed the effect of NH4+ on the expression of the betaine/GABA transporter (BGT-1), the sodium/myo-inositol cotransporter (SMIT), aldose reductase (AR), and heat shock protein 70 (HSP70) in Madin-Darby canine kidney (MDCK) cells, using Northern and Western blot analyses and enzyme-linked immunosorbent assay (ELISA). Qualitatively similar results were obtained for SMIT and AR mRNAs. Exposure to both isotonic and hypertonic, NH4Cl-containing medium enhanced apoptosis compared with equiosmolar, NaCl-containing media. These results suggest that, in addition to NaCl, NH4Cl may play a role in regulating the intracellular accumulation of organic osmolytes, the abundance of HSP70 and cell turnover in the renal medulla in vivo.

Gagam-whanglyun-haedoktang (GWH) is a newly designed herbal drug formula based on the traditional oriental pharmacological knowledge for the purpose of treating tumorous diseases. Apoptosis is an evolutionarily conserved suicide program residing in cells. In the present study, apoptosis inducing activities of the decocted
water
extract of GWH were studied. Results of the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay showed that GWH had a strong cytotoxic effect on HL-60 cells. The number of live cells was less than 20% after exposure to 1 mg/ml GWH for 48 h. GWH increased cytotoxicity of HL-60 cells in a dose- and time-dependent manner. The percentage of apoptotic cells by flow cytometric analysis of the DNA-stained cells increased to 28%, 31%, and 37% at 24 h and to 37%, 44%, and 81% at 48 h after treatment with 0.01, 0.1, and 1 mg/ml GWH, respectively. DNA fragmentation also occurred in apoptosis and was characterized by a ladder pattern on agarose gel. In addition, GWH increased the secretion of tumor necrosis factor-alpha. GWH-induced apoptosis was accompanied by activation of
caspase-3
. These results suggest that GWH induces activation of
