expression is transcriptionally regulated, we isolated the

gene promoter by genome walking and investigated the mechanisms regulating
caspase-11
gene expression in macrophages that are treated with LPS and interferon-gamma. Transient transfections with
caspase-11
promoter-luciferase reporter constructs and deletion/mutation analysis revealed an essential role for NF-kappaB binding in the up-regulation of
caspase-11
in response to LPS. In the case of interferon-gamma stimulation, signal transducer and activator of transcription 1 binding to the
caspase-11
promoter could be shown to be required for
caspase-11
expression.

-deficient mice are resistant to LPS-induced shock. Here we report that LPS-induced

regulates lymphocyte apoptosis by activating both caspase-3 and caspase-7. The activation of

preceded that of

and caspases-3/-7, and in the absence of

, the activation of caspases-3/-7 was significantly reduced. The early activation of caspases-3/-7 by

was not affected by blocking of

Myocardial ischemia is the leading cause of death in both men and women; however, very little information exists regarding the effect of testosterone on the response of myocardium to acute ischemic injury. We hypothesized that testosterone may exert deleterious effects on myocardial inflammatory cytokine production, p38 MAPK activation, apoptotic signaling, and myocardial functional recovery after acute ischemia-reperfusion (I/R). To study this, isolated, perfused rat hearts (Langendorff) from adult males, castrated males, and males treated with a testosterone receptor blocker (flutamide) were subjected to 25 min of ischemia followed by 40 min of reperfusion. Myocardial contractile function (left ventricular developed pressure, left ventricular end-diastolic pressure, positive and negative first derivative of pressure) was continuously recorded. After reperfusion, hearts were analyzed for expression of tissue TNF-alpha, IL-1beta, and IL-6 (ELISA) and activation of p38 MAPK,

-like protease with pro-apoptotic and pro-inflammatory activities. Here we have identified a novel exon at the 5'-end of the human caspase-5 gene. This novel exon was present in six alternatively spliced caspase-5 mRNA variants expressed in human peripheral blood mononuclear cells (PBMC) and encoded the previously unknown amino-terminus of caspase-5. The genomic region upstream of this exon contained sequence elements homologous to those of the
caspase-11
promoter in the mouse, and transcription of caspase-5 was upregulated by lipopolysaccharide (LPS) in a
caspase-11
-like manner in human PBMC in vivo. Taken together, our findings call for a revision of the structure of caspase-5 at the genomic level as well as at the mRNA and protein levels.

C/EBP homologous protein (CHOP) is one of the main mediating factors in the ER stress pathway. To elucidate the role of the ER stress-CHOP pathway in experimental pancreatitis, wild-type (Chop(+/+)) and Chop deficient (Chop(-/-)) mice were administered cerulein, a cholecystokinin analogue, or both cerulein and lipopolysaccharide (LPS). In cerulein-induced acute pancreatitis, ER stress, serum amylase elevation and histological interstitial edema were induced. However, there was no remarkable activation downstream of the CHOP pathway regardless of the presence or absence of CHOP. Whereas, in the cerulein and LPS model, inflammation-associated caspases (

caspase-1
) and IL-1beta, but not apoptosis-associated caspases, were activated. In Chop(-/-) mice, the expression levels of these mediators returned to basal levels resulting in a milder pancreatitis and decreased serum amylase level.
