Effects of
bromelain
(BR) on rabbit sputum consistency were investigated in vitro and in vivo. On the sputum showing relatively low viscosity, BR and other enzymes such as serratiopeptidase (SP), the mixed preparation of pronase and pancreatin, and
lysozyme
exerted lowering effects; and the effect of BR was the most potent. However, bromhexine had virtually no effect. On the sputum showing relatively high viscosity, BR exerted more potent lowering effects on the viscosity and yield value of sputum than those of SP. Furthermore, 320,000 U/head BR and 120,000 U/head SP lowered the viscosity significantly and yield value of sputum in rabbits with oral administration for 3 days. The lowering effect on the yield value of BR was more potent than that of SP. BR also increased the sputum volume in rabbits. BR and SP showed tendencies to decrease the contents of acid glycoprotein and sialic acid in sputum. It can be considered that these results support the effectiveness of BR as an expectorant in clinical use.

Protease supplementation has been shown to attenuate soft tissue injury resulting from intense exercise. The aim of this study was to evaluate the effects of protease supplementation on muscle soreness and contractile performance after downhill running. Ten matched pairs of male participants ran at a -10% grade for 30 min at 80% of their predicted maximal heart rate. The participants consumed two protease tablets (325 mg pancreatic enzymes, 75 mg trypsin, 50 mg papain, 50 mg
bromelain
, 10 mg amylase, 10 mg lipase, 10 mg
lysozyme
, 2 mg chymotrypisn) or a placebo four times a day beginning 1 day before exercise and lasting a total of 4 days. The participants were evaluated for perceived muscle soreness of the front and back of the dominant leg, pressure pain threshold by dolorimetry of the anterior medial, anterior lateral, posterior medial and posterior lateral quadrants of the thigh, and knee extension/flexion torque and power. The experimental group demonstrated superior recovery of contractile function and diminished effects of delayed-onset muscle soreness after downhill running when compared with the placebo group. Our results indicate that protease supplementation may attenuate muscle soreness after downhill running. Protease supplementation may also facilitate muscle healing and allow for faster restoration of contractile function after intense exercise.

Polyhydroxybutyrate (PHB) is the most studied among a wide variety of polyhydroxyalkanoates, bacterial biodegradable polymers known as potential substitutes for conventional plastics. This work aimed at evaluating the use of enzymes to recover and purify the PHB produced by Ralstonia eutropha DSM545. Screening experiments allowed the selection of trypsin,
bromelain
and
lysozyme
among six enzymes, based on their efficiency in lysing cells of a non-PHB producing R. eutropha strain. Then, process conditions for high efficiency in PHB purification from the DSM545 cells were searched for the enzymes previously selected. The best result was achieved with 2.0% of
bromelain
(enzyme mass per biomass), equivalent to 14.1 U ml(-1), at 50 degrees C and pH 9.0, resulting in 88.8% PHB purity. Aiming at improving the process efficiency and reducing the enzyme cost, experiments were carried out with pancreatin, leading to 90.0% polymer purity and an enzyme cost three times lower than the one obtained with
bromelain
. The molecular mass analysis of PHB showed no polymer degradation. Therefore, this work demonstrates the potential of using enzymes in order to recover and purify PHB and bacterial biopolymers in general.

Bromelain is a natural mixture of proteolytic enzymes derived from pineapple stem that has been shown to have anti-inflammatory activity when administered orally. Although most proteins given orally without adjuvant (e.g., food) result in tolerance, we previously reported that long-term oral exposure to
bromelain
stimulated the development of high serum anti-
bromelain
antibody titers. The purpose of these studies was to further investigate the mechanisms responsible for the immunogenicity of oral
bromelain
. Results showed that repeated exposure was required for development of anti-
bromelain
antibodies, with strong antibody responses in all mice that received at least 12 doses of
bromelain
either orally or intragastrically over 3-6 weeks. Proteolytic activity was required for strong oral immunogenicity in the absence of conventional adjuvant, with strong serum antibody responses generated against proteolytically active
bromelain
and trypsin, but not against ovalbumin,
lysozyme
, or inactivated
bromelain
. Significantly higher anti-
bromelain
antibody titers were seen in IL-10-deficient versus wild-type mice, suggesting that simultaneous treatments that decrease IL-10 activity may further enhance systemic antibody responses following oral exposure. The antibodies generated did not affect the proteolytic activity of
bromelain
. The data demonstrate that proteolytically active antigens such as
bromelain
can stimulate both systemic and mucosal immune responses following repeated oral exposure. Further studies of the mechanisms involved in generation of immune responses following oral exposure to proteolytically active antigens can lead to a better understanding of mechanisms of oral tolerance and to the development of novel adjuvants for oral vaccines.

Three triblock copolymers based on the poly(lactide) or poly(lactide-co-glycolide) and poly(ethylene glycol) or poly(ethylene oxide) blocks were synthesized and characterized. The weight average molecular weight and number average molecular weight were determined by gel permeation chromatography and proton nuclear magnetic resonance spectroscopy, respectively. Fourier transform infrared spectroscopy was used to determine the completion of synthesis of polymers. Thermoreversible sol-gel transition temperature and concentration were determined by an inverted tube method. Two formulations each of three synthesized polymers containing 5% (w/v) of
lysozyme
or
bromelain
but differing in polymer concentrations (20-30%, w/v) were prepared and studied for in vitro release of the incorporated protein. In vitro biocompatibility of the delivery systems was studied by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) cell viability assay. Biological activities of
lysozyme
and
bromelain
were determined by enzyme activity assays. Critical gelling concentrations were found in the range of 20-30% (w/v). In vitro biocompatibility study showed that all the formulations were biocompatible. Increasing the polymer concentration led to a decrease in burst release and extended the in vitro release of proteins. Furthermore, biological activities of
lysozyme
and
bromelain
in released samples were found to be significantly (p<0.05) greater in comparison to the control. Thus, the above thermosensitive polymers were able to deliver proteins in biologically active forms at a controlled rate for 2-8 weeks.

Mimivirus was investigated by atomic force microscopy in its native state following serial degradation by
lysozyme
and
bromelain
. The 750-nm diameter virus is coated with a forest of glycosylated protein fibers of lengths about 140 nm with diameters 1.4 nm. Fibers are capped with distinctive ellipsoidal protein heads of estimated Mr=25 kDa. The surface fibers are attached to the particle through a layer of protein covering the capsid, which is in turn composed of the major capsid protein (MCP). The latter is organized as an open network of hexagonal rings with central depressions separated by 14 nm. The virion exhibits an elaborate apparatus at a unique vertex, visible as a star shaped depression on native particles, but on defibered virions as five arms of 50 nm width and 250 nm length rising above the capsid by 20 nm. The apparatus is integrated into the capsid and not applied atop the icosahedral lattice. Prior to DNA release, the arms of the star disengage from the virion and it opens by folding back five adjacent triangular faces. A membrane sac containing the DNA emerges from the capsid in preparation for fusion with a membrane of the host cell. Also observed from disrupted virions were masses of distinctive fibers of diameter about 1 nm, and having a 7-nm periodicity. These are probably contained within the capsid along with the DNA bearing sac. The fibers were occasionally observed associated with toroidal protein clusters interpreted as processive enzymes modifying the fibers.

Aqueous Two-Phase Extraction is known to be a gentle separation technique for biochemical molecules where product partitioning is fast. However, the reason for the high mass transfer rates has not been investigated, yet. Many researchers claim that the low interfacial tension facilitates the formation of very small droplets and with it a large interfacial area causing a fast partitioning. However, an experimental evidence for this hypothesis has not been published yet. In this study, the mass transfer coefficients of two proteins, namely
lysozyme
and
bromelain
, were determined by providing a defined interfacial area for partitioning. Compared to low molecular weight solutes the mass transfer coefficient for the proteins investigated was small proving for the first time that the large interfacial area and not fast diffusion seems to be the reason for fast protein partitioning.
