The immobilization of
alpha-chymotrypsin
, trypsin and invertase on hydrated oxides of tin,
titanium
and aluminium was investigated. The degree at which the enzymes were bound upon immobilization was 83.2-2.6%. The amount of bound proteins was 64.2 mg/g carrier. The specific activity of enzymes reached the highest level in the case of hydrated tin oxide and amounted to 76.8%, 49.9% and 99.6%, of activity of native
alpha-chymotrypsin
, trypsin and invertase, respectively. The thermal stability of immobilized proteases was considerably higher and that of immobilized invertase was significantly lower than that of native enzymes. The pH optimum of immobilized enzymes shifted by 0.6-2.6 units towards the alkaline region.

Biological and biochemical characteristics of the leukotoxin of Fusobacterium necrophorum were determined. Culture supernatant of F necrophorum was toxic to polymorphonuclear neutrophilic leukocytes from cattle and sheep, but not to those from pigs and rabbits. Culture supernatant and sonicated bacterial cell fractions had low hemolytic activity and did not cause dermonecrosis in a guinea pig. Supernatant-derived leukotoxin was inactivated at 56 C for 5 minutes and became unstable at pH > 7.8 or < 6.6. Chemical treatment with 0.1% sodium dodecyl sulfate, 0.25% sodium deoxycholate, 5.2% sodium sulfide, or 0.25 mM

(III) citrate markedly decreased leukotoxicity. Enzymatic treatment with protease, trypsin, and
chymotrypsin
inactivated the toxin completely, whereas amylase had no effect. Use of protease inhibitors failed to prevent loss of leukotoxin activity. Using membrane partition chromatography and gel filtration, the estimated molecular weight of the toxin was > 300,000. On reduction and denaturation, the toxin dissociated into several components by use of sodium dodecyl sulfate-polyacrylamide gel electrophoresis.

We prepared a new composite gel fiber by the gel formation of cellulose acetate and
titanium
iso-propoxide. The fiber is harder than alginate gel; it is also stable in common solvents, phosphate solution, and electrolyte solutions over a wide range of pH from 3 to 10. The fiber shows amphoretic adsorption properties depending on pH, namely, it acts anionic with decreasing pH and cationic with increasing pH. However, the fiber had no adsorption property for a pyrogen endotoxin. The beta-galactosidase and
alpha-chymotrypsin

The use of enzymes in conjunction with inorganic photocatalysts requires stability against photooxidation. In this paper, we describe enhanced stabilization of a model enzyme,
chymotrypsin
, to photooxidation driven by
titanium
dioxide exposed to ultraviolet light (TiO(2)-UV). Stabilization is achieved conjugating the enzyme with an oligomeric adduct of UV-absorbing (2-[3-(2H-benzotriazol-2-yl)-4-hydroxyphenyl]ethyl methacrylate) (HBMA) and free radical-absorbing 2-methacryloyloxyethyl-6-hydroxy-2,5,7,8-tetramethylchroman-2-carboxylate (Trolox-HEMA). Juxtaposition of the antioxidant Trolox with the UV absorber HBMA within a single chain reduced the rate of deactivation of the former by TiO(2)-UV. This enables modified enzyme, which is adsorbed on TiO(2), to absorb both UV-light and free radicals and locally reduce the rate of photooxidation. Interestingly, Trolox was more readily deactivated by TiO(2)-UV when it was conjugated separately to
chymotrypsin
that had been pre-modified with HBMA moieties.

This preliminary study aims to investigate the effects of a maximal implant retention procedure. The authors retrospectively reviewed the use of negative pressure wound therapy with
chymotrypsin
irrigation treating implant infection/exposure in
titanium
mesh cranioplasty by comparing patients with
titanium
mesh totally retained, partially removed, or totally removed according to the evaluation during the surgery. Negative pressure wound therapy with
chymotrypsin
irrigation was applied 5 days after the surgery. The negative pressure was set at -125 to -150 mmHg. A total of 21 patients were included, 4 patients treated with
titanium
mesh totally removed; 3 patients treated with
titanium
mesh partially removed; and 14 patients treated with U-shape debridement with
titanium
mesh preserved completely. However, 1 patient in the U-shape group required a second debridement to remove all implant. Negative pressure wound therapy with
chymotrypsin
irrigation is a novel procedure and could be used to treat implant-related infection without the exchange of implant.
