Buffalo sperm acrosome resembles its counterpart in other species, being rich in hydrolytic enzymes. Of the enzyme activities estimated, acid phosphatase, beta-N-acetylglucosaminidase and hyaluronidase were low compared to those of ram semen. However, the aryl sulphatase activity was high. GOT activity estimated in sperm preparation may not be of acrosomal origin.

The changes in the weight, histology and biochemical composition of the epididymis (caput, corpus and cauda segments) in prepuberal rabbit and rhesus monkey in response to testosterone treatment were investigated. The increase in the weight of the organ was accompanied by increased levels of RNA and DNA. Androgen therapy caused an increase in the concentration of sialic acid, phospholipids and glycerylphosphorylcholine and activities of alkaline phosphatase, acid phosphatase and hyaluronidase. The cauda region of the epididymis exhibited relatively higher levels of sialic acid and glycerylphosphorylcholine. These findings are discussed in relation to the functional maturation of the organ and the role of androgen in this process.

Chromatography of honeybee venom on Sephadex G-150 super fine revealed a high molecular weight (HMW) fraction that elutes prior to hyaluronidase (HYAL) and comprises 2% to 4% of the venom weight. HMW appears to exist in polymeric form, and the polymer which is present in greatest concentration has an estimated molecular weight of 105,000 D. The 12% nitrogen content of HMW suggests it may not be all protein. HMW is antigenically and enzymatically distinct from HYAL and phospholipase A2 (PHOS A). The acid phosphatase activity known to be present in honeybee venom was found in the HMW fraction. Since it reacts by RAST with the sera of most individuals known to be sensitive to honeybee venom, and releases histamine from the peripheral leukocytes of such individuals, its role as an allergen is confirmed. Since individuals react to different degrees to HMW, HYAL, and PHOS A, there does not appear to be a single principal allergen in honeybee venom.

The effects of epinephrine, indomethacin, acetylsalicylic acid, dexamethasone, and cyclic AMP on lysosomal hyaluronidase activity in in the rabbit iris were studied in vitro. Indomethacin and acetylsalicylic acid inhibited the lysosomal hyaluronidase activity. Dexamethasone and cyclic AMP (adenosine-3',5'-cyclic monophosphate) had little effect on the enzyme activity. Epinephrine activated the enzyme activity. These drugs had no effect on the activities of either bovine testicular hyaluronidase or rabbit iridial acid phosphatase. The possible role of lysosomal hyaluronidase in the rabbit eye and the drug effects were considered.

In order to elucidate the cytochemical properties of the membranous structure between enamel and ameloblasts of the rat incisor at the maturation stage, chromic phosphotungstic acid (Cr-PTA) and periodic acid-silver methenamine (PA-silver) techniques for electron microscopy were employed in combination with a digestion test with hyaluronidase, neuraminidase, collagenase or trypsin. Also, acid phosphatase activity of ameloblasts at the maturation stage was examined with a modified GOMORI's metal salt method. An intensely Cr-PTA reactive band approximately 0.1 micron thick appeared along the surface layer of enamel at the transitional stage, and at the very beginning of the maturation stage another intensely Cr-PTA reactive band which was seen by uran-lead stain to be a delicate electron-dense membranous structure appeared as well between enamel and ameloblasts. A lot of cytoplasmic small vesicles or tubular structures, both intensely reactive to Cr-PTA, were observed near the apical membranes of the overlying ameloblasts indicating that those organelles must have been responsible for the secretion of the latter band. Acid phosphatase activity was clearly demonstrated at Cr-PTA reactive large vesicles in the cytoplasm of those cells. The PA-silver staining technique manifested a band heavily deposited with silver grains along the surface layer of enamel, i.e., where the former band existed, but showed no particular reaction at the latter, the band-like layer between enamel and ameloblasts. Hyaluronidase or neuraminidase treatment remarkably decreased the Cr-PTA reaction of the latter band. Trypsin or collagenase treatment, on the other hand, not only eliminated the Cr-PTA reaction but digested the band itself. These results suggest that the membranous structure between enamel and ameloblasts of a rat incisor is not so-called enamel cuticle but a basal lamina produced by overlying ameloblasts and that the basal lamina contains collagenous components even though it lies on enamel.

The effects of castration and testosterone replacement therapy on the histology and biochemical composition (RNA, DNA, total protein, alkaline phosphatase, acid phosphatase, hyaluronidase, sialic acid, glycogen, phospholipids, and glycerylphosphorylcholine [GPC]) of the epididymis of the rabbit and rhesus monkey were investigated. Castration produced marked ponderal, histologic, and biochemical changes in the epididymis. In the androgen-deficient state the tubular diameter and epithelial cell height were reduced and there was an increase in interbular stroma. The levels of RNA, DNA, phospholipids, and GPC were also reduced in castrated animals. Testosterone treatment restored the histologic features and the levels of various biochemical constituents to a great extent but not to the intact control level. The importance of endocrine and exocrine factors of the testis in relation to epididymal function is discussed.

Yellow jacket venom (YJV) contains acid phosphatase, hyaluronidase and phospholipase but neither allergen C nor melittin as bee venom (BV). YJV enzymes did not cross react with antisera to BV enzymes. YJV was separated into seven fractions, all of which had some activity in RAST. Sera were found specific for the fractions containing phospholipase and acid phosphatase. The other three protein fractions also exhibited substantial RAST activity, suggesting there are at least five allergens in YJV.

The mammalian toxicity of the potently algogenic venom of the ant Pogonomyrmex badius is greater than that reported for any other insect venom. This enzyme-rich venom contains high concentrations of phospholipase A2 and B, hyaluronidase, acid phosphatase, lipase, and esterases. This hemolytic secretion from the poison gland products unusual symptoms in mammals and appears to have been evolved as a deterrent for vertebrate predators.

Distribution of acid phosphatase, beta-glucuronidase, and lysosomal hyaluronidase in the anterior segment of the rabbit eye was studied biochemically. Acid phosphatase activity was higher in the anterior uvea and cornea but lower in the sclera. Beta-Glucuronidase activity was higher in the anterior uvea but lower in the corneoscleral tissues. Lysosomal hyaluronidase activity was higher in the anterior uvea. The inner layer of the corneoscleral junction showed the highest specific activity of beta-glucuronidase and lysosomal hyaluronidase among the corneoscleral tissues. Lysosomal hyaluronidase activity was detected in all corneoscleral tissues.

Tissues of rats with adjuvant arthritis manifested differences in activity and distribution between free, latent and membrane-bound forms of acid catepsins, alpha-D- and beta-D-galactosidases, alpha-D- and beta-D-glucosidases, beta-D-glucorunidase, hyaluronidase, acid phosphatase, arylsulphatases (A+B). Activation of certain hydrolytic enzymes is observed in tissues of the liver, kidneys, heart and spleen: a rise in total activity (of arylsulphatase in the liver and acid catepsins in the spleen; hyaluronidase in the kidneys, beta-D-glucuronidase in the heart) and a change in the ratio of different forms with a simultaneous increase in the activity of free form (of hyaluronidase in the spleen, acid phosphatase in the heart and liver). Inhibition of alpha-D-glucosidase in the liver and beta-D-glucosidase in the spleen is also detected. A decrease in the activity of beta-D-glucuronidase in the spleen is pronounced in a significant decrease in the activity of each enzyme free form with no changes in the total activity.
