A cell fraction enriched in biliary epithelial cells (BEC) has been isolated from the liver of normal rats. The procedure involved proteolytic digestion by trypsin and mild mechanical disruption of biliary ductular and connective tissue that remained undigested after collagenase-
hyaluronidase
perfusion. An adherence procedure removed the large majority of contaminating Kupffer cells. The majority (87.4 +/- 3.5%) of the cells were positive to an indirect immunofluorescence staining that used an antiserum against bovine hoof prekeratin that specifically recognizes intermediate filaments of biliary epithelium. Similar results were obtained by histochemical staining for
gamma-glutamyltranspeptidase
activity. The contamination of the BEC fraction with Kupffer cells and hepatocytes was approximately 7% and 2%, respectively. The viability of the BEC population was always more than 90%. The BEC and hepatocytes were analysed for their lipid composition; the BEC were found to have a cholesterol content approximately 6-times higher than hepatic parenchymal cells, with a cholesterol/phospholipid molar ratio of 0.53 in comparison to a value of 0.11 for hepatocytes. No detectable evidence of cytochrome P-450 or cytochrome P-450-related enzymatic activities was found in the BEC.

The effect of exploitation and adrenergic system drugs on sexual reflexes and properties of the semen in somatically mature Polish Large White boars has been presented. At the first stage 8 males were exploited twice a week. Then, previous to semen collection, the animals were given beta-adrenolytic (Propranolol--Polfa), after 30 minutes followed by the preparation Levonor--Polfa (alpha-adrenomimetic). 15 minutes after last injection the semen was collected, this procedure having been repeated twice a week. At the second stage of the experiment the same males were exploited once a week. Like in the first stage, the animals were given the same doses of the drugs in the same sequence. 15 minutes after last injection the semen was collected once a week. At the third stage of the experiment the boars were exploited every day. Again, like in the previous stages, the animals were given the above mentioned beta-adrenolytic and alpha-adrenomimetic to collect the semen 15 minutes after last injection. In all the stages of the experiment 6-8 ejaculates were collected from each boar. Apart from preliminary assessment of concentration and morphology of spermatozoons there was determined the level of protein and activity of selected enzymes in the semen. The sexual drive as well as the time of searching reflex and ejaculation were observed. The drugs applied have been proved to be able to prolong the time of ejaculation reflex. Simultaneous application of beta-adrenolytic and alpha-adrenomimetic to animals exploited twice a week appeared to increase the number of spermatozoons in the whole ejaculate, the percentage of spermatozoons with proper movement and the activity of
GGT
and fucosidase. Intensive exploitation of a boar with parallel application of the preparations described bring about a gradual decrease in spermatozoons and protein in the ejaculate as well as in the activity of
hyaluronidase
in its plasma and in conversion to mg protein of semen plasma.

Daily administration of 2g/kg/day di(2-ethylhexyl)phthalate (DEHP) to immature rats was found to cause testicular atrophy and reduce zinc concentration. Specific activities of testicular enzymes associated with postmeiotic spermatogenic cells, such as lactate dehydrogenase isozyme-X,
hyaluronidase
and sorbitol dehydrogenase, were lower than those of control by day 10, coincident with degeneration of spermatogenic cells. The specific activities of enzymes associated with premeiotic spermatogenic cells, Sertoli cells or interstitial cells (beta-glucuronidase,
gamma-glutamyl transpeptidase
and malate dehydrogenase) were higher than those of control by day 10. The specific activities of alcohol dehydrogenase and aldolase, zinc containing enzymes, increased after DEHP treatment in spite of the decrease in zinc concentration in the testis. In conclusion, changes in several testicular cell-specific enzymes appear to be useful biochemical markers of testicular injury induced by testicular toxicants such as DEHP. However, these changes occurred after or simultaneous with massive histological or morphological changes rather than prior to such changes.

levels, biochemical markers for testicular function, were evaluated in male rats. Treatment of 21-day-old rats with DL-204 for 7 or 15 days produced cryptorchid condition. Testicular
hyaluronidase
and
gamma-glutamyl transpeptidase
levels reveal that DL-204 acts on the testes, possibly in two ways; one, by reducing the gonadotropin levels thereby reducing the levels of androgens as reflected by reduced accessory reproductive organ weights and, secondly, by a direct action on the testes. Thus, we conclude that DL-204 is acting as an antispermatogenic agent, possibly acting in more than one way on the testes.
