The gastrointestinal invasive stages of two parasitic nematodes, Ancylostoma caninum and Anisakis simplex, were each found to release in vitro a hydrolytic enzyme that degrades the glycosaminoglycan hyaluronic acid. The parasite hyaluronidases were partially purified by ion-exchange chromatography and biochemically characterized. The
hyaluronidase
from A. caninum adult worms had a molecular weight of 65,000 and a pH optimum of 6 with activity at neutral pH, while the
hyaluronidase
from A. simplex larvae had a molecular weight of 40,000 and a pH optimum of 4 with no activity at neutral pH. Both parasite hyaluronidases also degraded the glycosaminoglycan chondroitin sulfate A. Cupric sulfate and high concentrations of sodium chloride were inhibitory. , attributed to the NagH protein, contributes to the pathogenicity of this organism. The family 84 catalytic module of one of the three homologues of NagH found in C. perfringens (ATCC 13124) has been cloned. The 69 kDa catalytic module of NagJ, here called GH84C, was overproduced in Escherichia coli and purified by immobilized metal-affinity chromatography (IMAC). Crystals belonging to space group I222 or I2(1)2(1)2(1) with unit-cell parameters a = 130.39, b = 150.05, c = 155.43 A were obtained that diffracted to 2.1 A. Selenomethionyl crystals have also been produced, leading to the possibility of solving the phase problem by MAD using synchrotron radiation.

Until now, Enterococcus cecorum (EC) has been known as a pathogen for broilers, broiler breeders, and Pekin ducks. In the present report, we describe a fatal systemic EC infection in a young racing pigeon (Columba livia forma domestica). EC was isolated from the heart, liver, spleen, and intestine of the bird in pure culture. In the pathologic examination, the pigeon showed
enteritis
and an ulcerative gastritis, which may have been predisposing factors for the development of the generalized EC infection. An accumulation of gram-positive cocci in spleen tissue was found in the histopathologic examination and confirms the presence of a systemic EC infection in the pigeon. Additionally, EC was isolated from cloacal swabs of other pigeons in the same loft, but no additional pigeons were submitted for necropsy. All EC isolates tested were negative by PCR for the enterococcal virulence factors cytolysin, enterococcal surface protein, aggregation substance,
hyaluronidase
, and gelatinase. Therefore, the reason for the enhanced virulence of the EC isolate remains unknown. Our report confirms EC as a disease-causing agent in pigeons and presents the first data concerning the analysis of EC for virulence factors.
